Analysis of the pathogenicity of 800 progeny from a HI-path (Blue) x LO-path (Cream) cross showed that the quantitative genetic control of pathogenicity in Nectria haematococca MPI on hypocotyls of Cucurbita maxima was determined by 6-12 ‘effective factor’ or quantitative trait loci (QTL). In addition there was evidence for a virulence/colony colour gene(s) with an effect that was superimposed on the pathogenicity phenotype.

We have constructed 2 tn5-containing plasmids, pLH1 and pLH3, specialized towards mutagenesis of genes from fungi which are auxotrophic for arginine or sensitive to hygromycin (such as the filamentous fungi Aspergillus nidulans and Magnaporthe grisea.) These plasmids are also a useful means of integrating additional marker genes in the plasmid backbone.

Vigorous growth occurred when conidia of two heterokaryon-incompatible auxotrophic strains of N. crassa were mixed on minimal medium. The syntrophic growth could often, but not always, be propagated through several subcultures using minute conidial inocula. We demonstrate that growth of auxotrophic strains occurred without hyphal fusion by cross-feeding.

A method is described for the reliable preparation of DNA from fungal spores and mycelia and from plant tissues. A number of fungal and plant species were used in the study to indicate the generality of the method. The DNA prepared by this protocol was digested by restriction endonucleases and served as template using standard polymerase chain reaction conditions.

We developed a simple ascospore plating assay for aneuploidy, based on identifying disomic progeny that inherit two independently selectable mtr alleles. We validated the assay using a known meiotic mutant, mei-2. We used this assay to demonstrate that elevated frequencies of aneuploidy previously reported to be associated with reduced DNA methylation were not, in fact, due to the methylation deficiencies. A new allele of the mei-1 gene was responsible for some of the high aneuploidy.

Generally niaD mutants of fungi are selected by spontaneous mutations on appropriate minimal medium supplemented with various concentrations of KClO3 and a nitrogen source (Daboussi et al. 1989 Curr. Genet. 15:453-456; Johnstone et al. 1990 Gene 90:181-192; Malardier et al.1989 Gene 78:147-156; Unkles et al.1989 Gene 78:157-166). But in case of entomopathogenic fungi it has been observed that niaDmutants isolated simply by spontaneous mutation on chlorate were not stable, (Table-1). Therefore a method has been developed to isolate stable niaD mutants of these fungi by treating protoplasts with ethane methane sulfonate (EMS).

Street Noises: Parisian Pleasure, 1900-1940 by Adrian Rifkin and Shanghai on the Métro: Spies, Espionage, and the French Between the Wars by Michael B. Miller are test cases for issues of historiography related to period and duration in the study of France between 1919 and 1940. Of added relevance to these issues is the fact that Rifkin and Miller both question distinctions between elite literary cultures linked to book publishing and new forms of mass reproduction enhanced by technologies of sound reproduction and illustration. The strategies of placemaking developed during the late seventies and implemented in the early eighties have proven insufficient to grapple with many of the irregularities of the contemporary metropolis. So while contextualism, collage, and other formalist design tools have been viable means for architects to reintegrate the fabric of pedestrian cities, such a fabric simply never existed in many American cities, particularly at their periphery. The essay examines the preoccupation with information technologies as a dominant theme of German poetry in the 1980s and 1990s. Outlining the historical shift from a critique of mass media to the ironic affirmation of hi-tech media, the investigation seeks an understanding of its driving forces. The analysis of Enzensberger's polemical essays on the culture industry shows the parallels and the difference in attitude between him and younger poets of the 1980s. A 35-d experiment was conducted to compare the effects of increasing spray-dried animal plasma and a high protein whey concentrate (73% CP) on starter pig performance. Spray-dried animal plasma and whey protein concentrate replaced dried skin1 milk on an equal lysine basis. Pigs fed increasing spray-dried animal plasma protein had increased ADG and ADFI from d 0 to 7 after weaning, but not for any other period in the study. Increasing whey protein concentrate had no effect on growth perforn1ance in relation to the pigs fed dried skim milk.; Swine Day, Manhattan, KS, November 20, 1997

The objective of this experiment was to study the influence of tetracycline on the transfer of antibiotic resistance in an in vivo swine model experimentally infected with antibiotic-resistant and antibiotic susceptible Salmonella spp. Tetracycline reduced the amount and duration of shedding of tetracycline- susceptible Salmonella. However, tetracycline had no effect on shedding of resistant Salmonella. We also have evidence that resistance was transferred from the resistant to the susceptible strain of Salmonella.; Swine Day, Manhattan, KS, November 20, 1997
