Protein A And Multimodal Anion-Exchange Membrane Adsorbers For Downstream Purification Of Therapeutic Biomolecules, Joshua Osuofa

This dissertation explores development and characterization of membrane chromatography tools for downstream purification of therapeutic biomolecules. Convective technologies, particularly membrane chromatography, are emerging in the field of bioseparations as an alternative to resin chromatography due to their improved throughput capability. On the forefront of emerging membrane technologies are multimodal and Protein A membrane adsorbers. The overarching theme of this dissertation research was to investigate advances in these membrane chromatography tools. The primary objective was to develop and characterize novel Protein A membrane adsorbers. A secondary objective was to characterize newly commercialized Protein A and multimodal anion exchange membrane adsorbers.

Thermoplastic polymers have been widely used in the industry due to their high toughness and impact resistance along with their remolding capabilities as compared to thermoset polymers, which also makes them an attractive choice for polymer composites. Many desirable features of thermoplastic polymers like ease of processing, low weight and cost, and corrosion resistance make thermoplastics a viable option for applications in the automotive, aerospace, sporting goods and many other industries. Analytical chemistry uses high performance liquid chromatography (HPLC) to separate desired macromolecules from a fluid. This thesis is concerned with monolithic column environments used in HPLC separations. The monolithic column environment under consideration consists of many long polymer fibers suspended in a tube. Between the fibers are voids (interstices) which regulate the separation process. The goal of this thesis is both to identify interstices of a size such that the interstice contributes to the separation process, and to identify the boundaries of the fibers along such interstices. Disclosed are open-framework solids that possess superior ion-transport properties pertinent to the electrochemical performance of next-generation electrode materials for battery devices. Disclosed compounds including compositions and architectures relevant to electrical energy storage device applications have been developed through integrated solid-state and soft (solution) chemistry studies. The solids can adopt a general formula of A.sub.xM.sub.y(XO.sub.4).sub.z, where A=mono- or divalent electropositive cations (e.g., Li.sup.+), M--trivalent transition metal cations (e.g., Fe.sup.3+, Mn.sup.3+), and X.dbd.Si, P, As, or V. Rational design of nanoparticle surface chemistry offers the ability to control nanoparticle characteristics such as size, polydispersity, shape, dispersibility in various solvents, functionality and end fate. Ligand exchange has proved to be is a versatile method for modifying the surface of plasmonic nanoparticles. Ligand exchange has provided a “green” alternative to traditional biphasic syntheses that require large amounts of phase transfer catalysts. Ligand exchange can also be used to reduce the amount of post synthesis processing and waste when it is conducted on nanoparticles that have been synthesized with a method that affords control over nanoparticle size and polydispersity. Accretion is a fundamental process which establishes the dynamics of the protoplanetary disk and the final properties of the forming star. In solar-type stars, the star–disk coupling is determined by the magnetic field structure, which is responsible for funneling material from the disk midplane to higher latitudes on the star. Here, we use pan-chromatic data for the Herbig Ae star MWC 480 to address whether similar processes occur in intermediatemass stars. MWC 480 has X-ray emission typical of actively accreting Herbig Ae stars, but with ∼10× more photoelectric absorption than expected from optical and FUV data. We consider three sources …
