A method for enhancing resistance to necrotrophic and/or hemibiotrophic pathogens by overexpressing glycerol-3-phosphate dehydrogenase using an expression vector in a plant species. For example, the present method can be used to enhance resistance to C. higginsianum by overexpressing glycerol-3-phosphate dehydrogenase in a plant such as Arabidopsis plant, using an expression vector in a plant.

The invention provides Bean pod mottle virus (BPMV) vectors useful for expression of heterologous proteins in plants such as soybean. The BPMV vectors are also useful for virus-induced gene silencing. The invention also provides methods for expressing a heterologous polypeptide in a plant such as soybean. The invention additionally provides methods for virus-induced gene silencing, particularly in a soybean plant, which can be used to determine the function of a gene of interest.

Loline alkaloids (LA), which are 1-aminopyrrolizidines with an oxygen bridge, are produced by Epichloë (anamorph=Neotyphodium) species, endophytes of grasses. LA are insecticidal, thus helping protect host plants from insect herbivory. Suppression subtractive hybridization PCR was used to isolate transcripts up-regulated during loline alkaloid production in cultures of Neotyphodium uncinatum. Subtracted cDNAs were cloned, and a λ-phage cDNA library from an LA-expressing N. uncinatum culture was screened with subtracted cDNA. In BLAST searches, several cDNAs identified had sequence similarities to aspartate kinases, and another with O-acetylhomoserine-(thiol)lyase. A low-profile cutter assembly for use on a microscope and related cutting method are provided. The cutter assembly includes a support subassembly having a mounting ring for receiving and engaging the objective of a microscope. A cutter subassembly carried by the support subassembly is displaceable between a home and a forward position in response to actuation. Upon reaching the forward position, additional actuation causes the front end of the cutter subassembly to pivot toward the stage, thereby placing a cutter adjacent to or in contact with the specimen. The present invention provides, inter alia, dmaW nucleic acid sequences and the proteins for which they encode. Also provided are methods for the utilization of knockout mutants of the sequences which are useful for engineering ergot alkaloid-deficient fungal symbionts (endophytes) of plants. Other methods and materials related to these sequences are also provided.
