Grus antigone sharpii จัดเป็นนกกระเรียนที่เคยสูญพันธุ์ไปจากธรรมชาติของประเทศไทย การเพาะเลี้ยงและขยายพันธุ์นกกระเรียนในกรงเลี้ยงเป็นขั้นตอนสำคัญที่สามารถเพิ่มจำนวนประชากรเพื่อนำไปปล่อยคืนสู่ธรรมชาติในอนาคตได้ โดยสิ่งสำคัญที่ต้องคำนึงถึงคือความหลากหลายทางพันธุกรรม โดยเฉพาะอย่างยิ่งในกลุ่มของพ่อแม่พันธุ์ตั้งต้น ในการศึกษาครั้งนี้มีวัตถุประสงค์เพื่อคัดกรองเครื่องหมายไมโครแซทเทลไลท์จำนวน 28 ตำแหน่ง ซึ่งเป็นไพรเมอร์ที่เคยใช้ศึกษาในนกกระเรียน G. americana และนกกระเรียน Anthropoides paradisea เพื่อใช้วิเคราะห์ความหลากหลายทางพันธุกรรมของนกกระเรียนพันธุ์ไทยจากสวนสัตว์เปิดเขาเขียว (KKOZ, n = 11) และสถานีวิจัยเพาะเลี้ยงนกน้ำบางพระ (BB, n = 17) จังหวัดชลบุรี ผลการศึกษาพบว่าไพรเมอร์ทั้ง 28 คู่สามารถเพิ่มปริมาณไมโครแซทเทลไลท์ดีเอ็นเอของนกกระเรียนพันธุ์ไทยได้ แต่มีเพียง 22 คู่ที่แสดงความเป็น polymorphism และมีตำแหน่งไมโครแซทเทลไลท์ที่ปรากฏค่า linkage disequilibrium 8 ตำแหน่ง การเบี่ยงเบนออกจากสมดุล Hardy-Weinberg พบที่โลคัส Gram8 และ Gpa38 ซึ่งเป็นผลมาจาก null allele และการเพิ่มขึ้นของอัลลีลแบบ heterozygous ตามลำดับ จากการวิเคราะห์ค่าความหลากหลายทางพันธุกรรมพบว่า ค่าเฉลี่ยของ expected และ observed heterozygosity มีค่าเท่ากับ 0.63 และ 0.69 ค่า inbreeding coefficient มีค่าเท่ากับ -0.100 ซึ่งแสดงว่านกกระเรียนมีความหลากหลายทางพันธุกรรมค่อนข้างสูง และเกิด inbreeding น้อย นอกจากนี้ยังพบว่านกกระเรียนจาก KKOZ และ BB มีความแตกต่างทางพันธุกรรมต่ำ (FST = 0.035) และไม่มีการแยกออกเป็นกลุ่มประชากรย่อย (K = 1) จากผลการศึกษาครั้งนี้บ่งบอกได้ว่านกกระเรียนพันธุ์ไทยจาก 2 พื้นที่การศึกษาน่าจะเหมาะสมที่จะใช้เป็นพ่อแม่พันธุ์เพื่อเพิ่มจำนวนประชากรในกรงเลี้ยงให้มากขึ้น และสามารถนำไปปล่อยคืนสู่พื้นที่ชุ่มน้ำตามธรรมชาติในอนาคตได้

This study has evaluated biological activities of isolated compounds from Tinospora baenzigeri stems collecting from two different areas, and from mangrove-derived endophytic fungi isolated from trees inhabiting in Songkhla province. Four new rearranged clerodane-type diterpenes (66-69), one new glycoside (70), and six known compounds (61, 71-75), were extracted from plant obtained from local market in Bangkok. The alternansucrase (ALT, EC 2.4.1.140) catalyses transferring of glucose from sucrose to produce a polymer with α-1,6 and α- 1,3 linkages, so-called "alternan". The ALT-encoding gene from Leuconostoc citreum ABK-1 (WTalt) was successfully cloned and expressed in Escherichia coli BL21 (DE3). The two versions of truncated SH3-like domains, ∆3SHALT and ∆7SHALT, were successfully constructed. The WTALT, ∆3SHALT and ∆7SHALT possessed optimum temperature of 40 °C, while optimum pH of WTALT was at 5.0 but ∆3SHALT's and ∆7SHALT's were shifted to pH 4.0. Alpha-glucosidase [EC 3.2.1.20; α-D-glucoside glucohydrolase] is an exohydrolase which catalyzes non-reducing end of substrates to release D-glucose. In addition, alpha-glucosidase also displays a transferase activity, which bring about the formation of α-glucosylated compounds such as soluble starch and glycogen. This research aims to express and characterize α-glucosidase from Weissella confusa BBK-1 (WcAG). WcAG gene was expressed in modified pET28b vector. Bacterial isolate HM7 was isolated from Dynastes hercules larvae excrement. It was identified as Bacillus sp. by 16S rRNA gene analysis. When the bacteria were cultured for β-mannanase production in medium containing konjac flour it had the highest specific activity of 138 U/mg at 36 h, and when cultured in medium containing coconut meal it had the highest specific of 114 U/mg at 48 h. The 1089 base pairs long of a β-mannanase encoding gene (Man26HM7) was successfully cloned and expressed in Escherichia coli BL21 (DE3), using pET21-b expression system. Oxidized carbon nanospheres (OCSs) and graphene oxide sheets (GOShs) were prepared by oxidation of graphite. Oxidized carbon nanotubes (OCTs) was synthesized from single-walled carbon nanotubes. All three shapes of oxidized carbon nanoparticles (OCNs) were used for investigation of their intracellular trafficking in human embryonic kidney cells (HEK293T), mouse macrophage cells (RAW264.7) and human liver cancer cells (HepG2). The results revealed all three shapes of OCNs were internalized into through HEK293T and RAW264.7 cells without endosomal trapping, including localization of all the three OCNs in HEK293T's lysosome. In this study, isolation, screening, fermentation optimization, and determination of the expression of key enzymes involving in D-lactic acid production were conducted for selection of the novel D-lactic acid isolate with the high yield, productivity, and optical purity. From the screening experiment, the novel isolate, NK26-11T, was obtained. The isolate NK26-11T was a Gram-stain-positive, catalase-positive, facultatively anaerobic, spore-forming, rod-shaped bacterium isolated from soil sample in Thailand. This isolate homofermentatively fermented glucose for D-lactic acid and grew at the wide range of temperature between 20 and 45 °C and the pH of 5-8.5. In this dissertation, immunoassays on paper-based microfluidic devices have been developed using different types of label for various applications in agriculture safety and medical diagnosis, which can be divided into three parts. In the first part, the conjugation method of ractopamine (RAC) and protein carrier is developed for fabricating paper-based lateral flow strip test of RAC using gold nanoparticles (AuNPs) as a label. The developed conjugation method provided a simple and one-step method with the use of less chemicals. Bioactive Peptide Derived From Trypsin Hydrolysis Of Unicellular Cyanobacterium Synechococcus Sp., Rutairat Suttisuwan

A free radicals can promote denaturing reactions in many cellular components, which create oxidative stress and it results to damage in body tissue. The free radicals can causing many diseases including heart diseases, Alzheimer, inflammation and cancer. This research aimed to investigate the effect of bioactive peptides derived from Synechococcus sp. cultured for 21 days on antioxidant, anti-inflammation and anticancer activities. The Synechococcus sp. protein hydrolysate was prepared by trypsin digest and purified by ultrafiltration with molecular weight cut off membranes of 10, 5 and 3 kDa. The MW <3 kDa fraction showed the highest NO, ABTS and DPPH radical scavenging activities with IC50 values of 34.51±9.8 µg protein/mL, IC50 11.54±0.3 µg protein/mL and IC50 13.63±0.15 µg protein/mL, respectively. The F2 fraction from gel filtration chromatography showed the strongest NO and ABTS radical scavenging activities with IC50 values of 7.68±0.64 µg protein/mL and 9.74±0.2 µg protein/mL, respectively. The F2 was purified by RP-HPLC to yield four fractions. The 30 – 40 min sub-fraction (F2-4) was selected for further analysis by mass spectroscopy. Five isolated peptides with amino acid sequences of AILQSYSAGKTK; 1,265.69 Da, ALNKTHLIQTK; 1,265.74 Da, LLVHAPVK; 875.55 Da, IPDAHPVK; 875.48 Da and VVVLRDGAVQQLGTPR; 1,706.97 Da were identified. The F2-4 had higher DPPH and NO radical scavenging activity compared to the synthetic peptide. Moreover, AILQSYSAGKTK had the highest ABTS radical scavenging activity. Furthermore, the F2 fraction protected oxidation-induced DNA damage in pBR322, pKS and pUC19 cells. F2 fraction was selected to study anti-inflammatory and anticancer properties. Anti-inflammatory effect, F2 fraction showed no cytotoxicity toward RAW264.7 macrophage cells. RT-PCR and qRT-PCR results showed that F2 reduced gene expression of pro-inflammatory cytokines iNOS, TNF-α, COX-1, COX-2 and IL-6. For anticancer activity, F2 fraction showed high anticancer activities in the human colon cancer cells (SW620) according to cytotoxic activity (MTT assay), with IC50 values of 106.58±21.46 µg protein/mL. The F2 fraction activated the apoptotic pathway in SW620 cells after treatment for 24, 48 and 72 hours. The highest activities of caspases 3, 8 and 9 were observed after treatment for 72 hours. These findings suggested that unicellular cyanobacterium Synechococcus sp. may be used to develop for natural anti-inflammation and natural anticancer drugs.

UV-A and -B blocking agents are essential components for the formulation of topical cream for psoriasis phototherapy. In this research, many natural compounds were discovered as potent UV-A and UV-B blocking agents, but small number could absorb narrowband UV-B. Quercetin hydrate, oxyresveratrol and curcumin were good UV-A blocking agents, while catechin, hesperetin, lutein and naringenin were proper for UV-B blocking agents. Berberine chloride hydrate, demethoxycurcumin and mansonone G were UV-A and -B blocking agents. โรคกระดูกพรุนเป็นปัญหาสุขภาพที่สำคัญอย่างหนึ่งของสังคมผู้สูงอายุ สาเหตุหลักของภาวะดังกล่าวเกิดจากความไม่สมดุลของเซลล์ที่ทำหน้าทีสร้างกระดูก (osteoblast) และเซลล์ที่ทำหน้าที่สลายกระดูก (osteoclast) การลดลงของมวลกระดูกในผู้สูงอายุนำมาซึ่งความเสี่ยงในการแตกหักของกระดูก จุดประสงค์ของการศึกษาครั้งนี้ คือ การคัดกรองสารออกฤทธิ์ทางชีวภาพที่มีฤทธิ์ในการต้านการเปลี่ยนสภาพของเซลล์สลายกระดูกและกลไกการออกฤทธิ์ของสารนั้นๆ ในกลุ่มสารทดสอบอนุพันธ์สาร (3S)-1-(3,4-dihydroxyphenyl)-3-hydroxy-7-phenyl-(6E)-6-heptene (DHPH) และ กรดไซเปอเรโนอิก (Cyperenoic acid) ซึ่งสกัดได้จากส่วนเหง้า Curcuma comosa Rox. และราก Croton crassifolius Geiseler. ตามลำดับ มีฤทธิ์แรงในการต้านการเกิดเซลล์สลายกระดูก ในระดับ in vitro ใช้เซลล์จากไขกระดูกที่ถูกกระตุ้นด้วย receptor activator of nuclear factor-κB ligand (RANKL) DHPH มีฤทธิ์ในการยับยั้งการเปลี่ยนสภาพไปเป็นเซลล์สลายกระดูก โดยยับยั้งการเกิด TRAP-positive multinucleated cells (TRAP+ MNCs) ที่ระดับความเข้มข้นยับยั้งกึ่งหนึ่ง (IC50) 325±1.37 นาโนโมลาร์ การยับยั้งดังกล่าวสอดคล้องกับการลดต่ำลงของสัญญาณภายใต้การกระตุ้นของ RANKL โดยรวมถึงวิถี Mitogen-activated protein kinases (MAPKs) เช่น ERK (p44/42) และ p38 แต่ไม่มีผลต่อวิถี NF-κB โดยผลของสารดังกล่าวยังส่งผลกระทบต่อระดับของ transcription factor เช่น nuclear factor of activation T cells (NFATc1) และ c-Fos ซึ่งมีความสำคัญมากในการเปลี่ยนสภาพเป็นเซลล์สลายกระดูก DHPH ลดการกัดกร่อนกระดูก (bone resorption) บนแผ่นกระดูก (bone slices) นอกจากนั้นสาร DHPH ยังสามารถเพิ่มกิจกรรมของเอนไซม์ alkaline phosphatase (ALP) ที่เกี่ยวข้องกับการเปลี่ยนสภาพของเซลล์สร้างกระดูกในเซลล์ชนิด MC3T3-E1 ในกรณีของกรดไซเปอเรโนอิกนั้น พบว่าสามารถลดการเกิด TRAP+ MNCs ที่ระดับความเข้มข้น IC50 ที่ 36.69±1.02 …
