Compact 1 Ghz Electron Paramagnetic Resonance Spectrometers For Preclinical Imaging Studies, Tanden A. Hovey
Electronic Theses and Dissertations
Magnetic resonance is one of the most generally useful scientific techniques. New horizons of understanding are revealed as technological advances make possible explorations of nuclear spins and electron spins at higher and lower magnetic fields and electromagnetic frequencies. While the greatest current effort is on ever higher magnetic fields and microwave frequencies, both in nuclear magnetic resonance (NMR) and electron paramagnetic resonance (EPR), the Eaton lab focusses on developing high signal-to-noise performance EPR at low microwave frequencies. EPR provides information about O2, pH, viscosity, glutathione concentrations, and reactive oxygen species in living organisms that MRI cannot. To yield clinically or …

A Computational Micropatterning Approach To Study Caveolae And Protein Localization In Vascular Biology, Andrew B. Grespin
A Computational Micropatterning Approach To Study Caveolae And Protein Localization In Vascular Biology, Andrew B. Grespin
Electronic Theses and Dissertations
Caveolae are specialized, flask-shaped membrane invaginations highly expressed in endothelium and dysregulated in atherosclerosis. Caveolae play a central role in buffering membrane tension, yet the principles governing their spatial organization remain elusive. Thus, we sought to generate the most comprehensive and systematic analysis of blood vessel caveolar spatial organization. However, cell culturing, the backbone of human-focused biological research, does not standardly do well to model physiologically relevant cell behaviors such as migration or polarity-based tissue formation, particularly for punctate proteins and structures like caveolae. Micropatterns are cell-adhesive shapes that biophysically confine cell(s) to a user defined geometry which stereotype organelle …
An Integrated Sensor-Fusion Framework For Performance-Based Compensation In Construction, Aly Ayman El-Ashry
An Integrated Sensor-Fusion Framework For Performance-Based Compensation In Construction, Aly Ayman El-Ashry
Theses and Dissertations
Construction productivity has historically stagnated due to a reliance on subjective labor monitoring and “lagging” performance indicators. This research proposes a novel compensation model with the use of Total Energy Management System (TEMS) to translate labor effort and efficiency into payment incentives with the integration of Internet of Things (IoT) wearables and Statistical Process Control (SPC) charts.
This research addresses the systemic problem of declining construction productivity caused by industry’s inability to objectively quantify human effort. While traditional models rely on post-facto production tallies, a significant research gap exists in real-time physiological labor monitoring. To bridge this, the proposed TEMS …

Tissue engineering is widely used in research for investigating cellular proliferation, behavior, and responses to various stimuli. However, the predictive value of preclinical studies using cell culture plates is limited by the inability to recapitulate the complexity of the physiological microenvironment. Synthetic three-dimensional (3D) scaffolds can be engineered to mimic the complex morphology of the extracellular matrix of native tissues and can serve as physiologically relevant platforms for preclinical studies. In this study, 3D electrospun scaffolds were characterized to aid in breast cancer research. Unlike previous studies that focused primarily on scaffold fabrication or cell viability, this work systematically evaluates …

Design And Development Of A Perfusable Hydrogel Platform With Bioprinted Human-Endothelialized Microvascular Channels, Calvin A. Chernyatinskiy
All Theses
We developed an integrated bioprinting and perfusion platform to generate vascularized hydrogel constructs that support perfusable microchannel networks. Multiple hydrogel formulations, sacrificial ink systems, device geometries, and perfusion conditions were systematically evaluated to optimize print fidelity, mechanical stability, and cellular compatibility. Internally gelling alginate–calcium carbonate hydrogels were identified as a robust matrix, providing tunable gelation kinetics and structural stability during embedded printing. In parallel, Pluronic F-127 fugitive ink enabled reliable fabrication of continuous hollow channels due to its shear-thinning behavior and efficient post-print removal.
The finalized platform integrates stereolithographically fabricated perfusion chambers with extrusion-based bioprinting to produce three-dimensional, perfusable vascular …

A Computational Approach To Graduated Centrifuge Profiles For Partial Gravity Planetary Environments, Cabell W. Jones
Mechanical & Aerospace Engineering Faculty Publications
Missions such as NASA’s Artemis and SpaceX’s Starship aspire towards creating permanent settlements beyond Earth. In order to achieve this goal, it must be considered that the gravity present on planetary surfaces such as the Moon (0.165 g) and Mars (0.378 g) may be insufficient to support human physiology for long term habitation. On near-Earth missions, issues such as muscular atrophy, neuro-ocular changes, cardiopulmonary variations, and bone mineral density loss have been observed proportional to the duration spent in microgravity. Further studies have suggested that the severity of these conditions scales linearly with decreasing gravity, meaning that astronaut health can …

The Effect Of Fascicular Elastin On The Mechanical And Functional Properties Of Healthy, Damaged, And Healing Tendon, Shawn Pavey
The Effect Of Fascicular Elastin On The Mechanical And Functional Properties Of Healthy, Damaged, And Healing Tendon, Shawn Pavey
McKelvey School of Engineering Graduate Student Theses & Dissertations
Mechanical properties of tendon are highly influenced by structural protein composition and microscopic sub-structures. Within the largest subunit of tendon, the fascicle, the role of the elastin protein remains understudied despite impressive extensibility and fatigue resistance of its resulting elastic fibers. While previous work catalogued contributions of fascicular elastin across tendon type and species, the anticipated effects of elastin in fatigue and healing have not yet been explored. While previous knockout mouse models showed that disruption of elastic fibers led to altered mechanical properties (e.g., increased linear modulus), these models depended on heterozygous elastin deficiency or indirect knockout of proteins …

Heavy metals are ubiquitous environmental pollutants, contaminating air, soil, and water via the erosion of natural deposits, as well as originating from anthropogenic sources, such as agriculture, industries, transportation, and landfills. The increasing utilization of heavy metals over the years, combined with the persistent nature of metals in the environment poses a direct threat to human and environment health. Although regulatory limits have been established for toxic metals, assessing the associated health risks using real-life exposure scenarios remains challenging. In this review, we summarize the development and use of in vitro models based two- and three-dimensional cell culture systems, focusing …

Triboelectric nanogenerators (TENGs) have garnered significant research interest due to their ability to harvest mechanical energy efficiently. In this study, we report a TENG composed of polydimethylsiloxane (PDMS) and polyvinyl alcohol (PVA) as triboelectric layers. To enhance charge generation in the PDMS composite polymer, we incorporated lithium niobate (LiNbO3) nanoparticles, leveraging their piezoelectric and ferroelectric properties. The LiNbO3 nanoparticles were synthesized using a solid-state reaction method, resulting in two distinct phases: triclinic LiNbO3 and monoclinic LiNb3O8. Various weight percentages of LiNbO3 and LiNb3O8 nanoparticles were added to the PDMS matrix to optimize power generation. The maximum open-circuit voltage (VOC) and …

Dissertations
As the global population ages, the demand for wearable assistive technologies continues to rise, driven by their potential to enhance mobility and independence in older adults. Effectively designed controllers for lower-limb exoskeletons to assist sit-to-stand (STS) and walking are crucial for delivering efficient, safe, and comfortable assistance during daily activities. Traditionally, controller optimization involves biomechanical modeling and user-specific customization. Musculoskeletal simulations play a central role in this process by providing insights into human-exoskeleton interaction dynamics, thereby informing and refining control strategies.
This work presents a simulation-driven approach for developing exoskeleton controllers for walking and STS using two distinct methods: optimal …

Polyphenols For The Mitigation Of Vascular Calcification; In Vitro And In Vivo Studies, Abdullah Altuhami
All Theses
Globally, cardiovascular diseases, in most cases associated with calcification, are the main cause of death. However, currently, the only effective cure is to undergo surgery to augment or receive a healthy donor heart, which could be very difficult due to the long waiting list and the associated risks. The possibility of body rejection of the foreign organs is attributed to immunoreaction or the risk of infection, since the donor must be on immunosuppressant drugs that lower the body's immunity and facilitate the body's acceptance of the new organ as one of its own. Taking that into consideration, using tissue-engineered vascular …

Effects Of Environmental Stressors On Human Tissue-On-A-Chip Platforms, Andie Padilla
Open Access Theses & Dissertations
As space exploration begins to extend beyond low earth orbit, it has become increasingly critical to understand the interaction of the extreme environment of space flight with human systems. While it is known that space-travel induces a vast array of complications to cardiac, neural, musculoskeletal, and immune systems, the mechanisms by which these complications occur are poorly understood. Current research to study the effects of microgravity and radiation are limited to ground simulations, which rarely account for the multifactorial stressors experienced during spaceflight, or long duration studies aboard the International Space Station. Similarly, traditional two-dimensional (2D) models lack the ability …

Epithelial cells are among the most abundant cell types in the human body, making up sheets of epithelial tissues that cover the linings of organs and body cavities. Cellular adhesion and force transmission across epithelial tissues are crucial in maintaining tissue integrity and ensuring proper tissue regeneration. Defects in cellular force transmission have been found in various diseases, including cancer. Force transmission at cell-cell and cell-ECM junctions drive cell survival and development. It has been previously shown that changes in cell-ECM traction directly modulates forces at cell-cell contacts. In this study, we aim to determine the effects of perturbations in …

Long-duration space missions pose significant challenges to astronaut health, primarily due to the detrimental effects of microgravity on musculoskeletal and cardiovascular systems. This research proposal aims to address the critical gap between pre- and post-spaceflight physiological conditions, focusing on the marked decline in muscle mass, bone density, and cardiovascular function. Existing countermeasures, such as resistive exercise, are insufficient to prevent muscle atrophy and bone demineralization, highlighting the need for novel solutions. Neuromuscular Electrical Stimulation (NMES) emerges as a promising intervention, capable of eliciting muscle contractions to maintain function and mitigate atrophy. By integrating NMES into existing exercise regimens, we hypothesize …

Cancer is a complicated disease and one in every three diagnoses are women with breast cancer. Recent research shows that the progression of cancer depends strongly on interactions with other non-cancerous cells and non-cellular components. Metastasis, the spread of cancer to other parts of the body, is a key step in cancer progression and makes treatment harder, leading to poor response to treatments culminating in death in ~90% of patients. This dissertation focused on creating a series of microdevices as novel pre-clinical models of breast cancer. Two different devices were fabricated to (1) study how exposing cancer cells to the …

In an attempt to reduce reliance on petroleum-based polymers, lignin – an abundant biopolymer – has gained popularity as a sustainable alternative. However, research on lignin-containing soft composites – i.e., hydrogel (bio)composites – has largely been heuristic, providing limited insight into the fundamental structure-processing-property relationships that govern performance improvements in this emerging class of ‘green’ soft composites. As such, this dissertation presents a systematic investigation of how lignin molecular weight (MW), composite composition, and the concentrations of crosslinker and accelerator impact the mechanical and transport characteristics of soft composites composed of lignin, poly(N-isopropylacrylamide) (PNIPAm), and poly(vinyl alcohol) (PVA).

Background
Head and neck squamous cell carcinoma (HNSCC) presents significant treatment challenges, particularly in cases unrelated to human papillomavirus (HPV). The chemokine receptor CXCR4, interacting with its ligand CXCL12, plays a crucial role in tumor proliferation, metastasis, and treatment resistance. This study explores the therapeutic potential of engineered monomeric and dimerized CXCL12 variants (CXCL121 and CXCL122, respectively) in HNSCC and evaluates potential additive effects when combined with radiation therapy.
Methods
Clinical HNSCC biopsies were evaluated for CXCR4 expression in both previously untreated and radiorecurrent disease. HNSCC cell lines were then treated with combinations of CXCL12 variants and radiotherapy and interrogated …
