Porcine Parainfluenza-1 (PPIV-1) is a Paramyxovirus inthe genus Respirovirus. To evaluatethe frequency of detection of PPIV-1 on farms in the United States, theIowa State University Veterinary Diagnostic Laboratory (ISU VDL) conducted an analysis of clinical samples submitted from Spring toFall 2016.Thirty-four percent of diagnostic samples tested both prospectively (70/204) and retrospectively (472/1385) were positive by polymerase chain reaction (PCR), which suggests PPIV-1 is widespread.

The liver plays a variety of roles in energy metabolism, digestion, and immune response. During the challenge by anenvironmental stressor, such as heat, the liver is one of the key organs that determines the chicken’s ability to cope with the hostile environment. Sequencing RNA extracted from liver samples from a heat-susceptible broiler line and a heat-resistant Fayoumi line, we have identifieda set of genes that were differentially expressed due to an acuteheat stress challenge. Initial antibody titers are maternally-derived from colostrum, then decay with age. Change in antibody titer levels were compared between four viruses contributing to the Bovine Respiratory Disease Complex (BRDC), and evaluation of response to vaccination indicated that antibody production will not occur when high levels of maternal antibodies are present. The maternal antibodies were found to decay with calf age for each of the four viruses, which allowed for the estimation of a maximum circulating titer level under which a positive antibody response to vaccination could occur. Hyperphosphatemia in chronic kidney disease (CKD) patients is a risk factor for cardiovascular events, progressive kidney failure, and mortality. Improved therapeutic interventions to control hyperphosphatemia depend greatly on robust animal models that recapitulate the CKD disease process. Murine-based models of CKD as compared to rat models present significant advantages due to available genetic knockout lines that permit mechanistic dissection of CKD etiologies. A Severe Combined Immunodeficient (SCID) Yorkshire pig line has been identified at Iowa State University. Since SCID animals lack an adaptive immune system they are instrumental models for biomedical and immunological research. An important goal for this thesis and for development of the ISU SCID pig is to characterize its natural immune parameters, and use the SCID pig as a biomedical model for exploring research questions.

Equine protozoal myeloencephalitis (EPM), caused by the protozoan parasite Sarcocystis neurona, is one of the most important neurological diseases of horses in the Americas. While seroprevalence of S. neurona in horses is high, clinical manifestation of EPM occurs in less than 1% of infected horses. Porcine epidemic diarrhea virus (PEDv) causes disease and mortality to piglets worldwide. Most vaccines used to combat the disease have been ineffective live attenuated virus vaccines. The goal of this project was to produce a plant-made subunit vaccine based off the membrane protein of the virus. This is the first time this protein has been produced in plants. An elastin-like polypeptide fusion membrane protein accumulated up to 0.8 mg/g of fresh leaf weight when transiently expressed in Nicotiana benthamiana. Aflatoxin contamination in feed and feed ingredients is of high worldwide prevalence, posing serious risks to the livestock industry. A series of studies were conducted to better understand the comprehensive impact of aflatoxicosis and its interrelationships with dietary modifications in broiler chicks and ducks. Results of these studies emphasize the many factors that the severity of aflatoxicosis depends on, including animal species, age, presence of mycotoxin adsorbent, and concentration of major dietary nutrients. Bovine herpes virus 1 (BHV-1) and Herpes simplex virus 1 (HSV-1) are crucial etiological viral agent of clinical diseases. HSV-1 and BHV-1 establish latent infection in sensory neurons. Periodically, reactivation from latency occurs resulting in virus excretion and transmission. Stress increases corticosteroid levels and the incidence of HSV-1 and BHV-1 reactivation from latency. The synthetic corticosteroid, dexamethasone (DEX) mimics stress and induces BHV-1 and HSV-1 reactivation. Hemorrhagic colitis occasionally progresses to hemolytic uremic syndrome (HUS), an important cause of acute renal failure in children and morbidity and mortality in adults. Transmissible spongiform encephalopathies (TSEs) are neurodegenerative diseases caused by prions. Although these infections usually remain asymptomatic for years, the disease is always progressive and fatal once the clinical signs develop.
