The main concern of this study is looking into the neural mechanisms of associative learning with a nicotine stimulus. The goal of this study was to find out if inactivating the corticostriatal pathway through DREADDs (Designer Receptors Exclusively Activated by Designer Drugs) inhibited goal-tracking behavior in rats. In this experiment, rats underwent two surgeries. One was to inject DREADDs into the infralimbic cortex and the other in the dorsomedial caudate-putamen (dmCPu). Rats were initially trained on levers through water deprivation. Once they reached the criterion, they moved on to the self-administration phase. Risky choice can be operationally defined as the choice for a larger, uncertain reinforcer over a smaller, certain reinforcer. Research suggests smokers engage in more risky or maladaptive decisions when compared to nonsmokers. The relation between nicotine and risky choice could benefit from further investigation, since nicotine is the active substance of tobacco products that maintains tobacco addiction. The objective of the present study was to determine the effectiveness of the opioid antagonist naltrexone at reducing the consumption of EtOH and nicotine in female alcohol-preferring (P) rats. P rats have been selectively bred to have a genetic predisposition for alcohol abuse, which allows them to be used as an animal model of alcoholism. P rats readily self-administer i.v nicotine (Le et al., 2016), have EtOH consumption during adolescence that is similar to that seen in adulthood, and operantly respond for EtOH until they are impaired/intoxicated (Bell et al., 2006). Co-users of alcohol and nicotine are the largest group of polysubstance users worldwide. Although pharmacotherapies are available for alcohol (EtOH) or tobacco use disorders individually, it may be possible to develop a single pharmacotherapy to treat heavy drinking tobacco smokers through capitalizing on the commonalities in their mechanisms of action. Towards this goal, several models of concurrent access to EtOH and nicotine were explored as potential preclinical models of co-use using female alcohol-preferring (P) rats. Additionally, potential pharmacotherapeutics for the treatment of EtOH and nicotine co-use disorder were tested using different variations of our model. Tobacco smoking in the United States is used by approximately 25% of adults. Many studies using animal models have suggested that nicotine has rewarding properties. Contrastingly, several studies have also found it to be a weakly reinforcing substance at low and high dose levels. Due to this, other tobacco constituents, such as the monoamine oxidase inhibitor norharmane which is found in tobacco leaf and smoke, may be responsible for tobacco addiction by potentiating the rewarding properties of nicotine. Little is known about how electronic cigarette (EC) users manipulate device parameters, what factors drive their use, and how non-nicotine ingredients influence the stimulus effects of EC aerosols. The ingredients propylene glycol (PG) or vegetable glycerin (VG) serve as the base for virtually all electronic cigarette liquids, and information on how they affect the using experience would provide important groundwork for the study of other ingredients. Neonatal quinpirole (NQ) treatment to rats increases dopamine D2 receptor sensitivity persistent throughout the animal’s lifetime. In Experiment 1, we analyzed the role of α7 and α4β2 nicotinic receptors (nAChRs) in nicotine behavioral sensitization and on the brain-derived neurotrophic factor (BDNF) response to nicotine in NQ- and neonatally saline (NS)-treated rats. In Experiment 2, we analyzed changes in α7 and α4β2 nAChR density in the nucleus accumbens (NAcc) and dorsal striatum in NQ and NS animals sensitized to nicotine. Introduction: An active area of tobacco regulatory science research focuses on examining the effects of varying the nicotine content of cigarettes as part of a potential national policy to lower their nicotine content levels to reduce addiction potential. The present study examines differences in the behavioral effects of reduced nicotine content cigarettes related to their menthol status. Menthol is the only cigarette flavoring that is still legally permissible according to Food and Drug administration regulations.

Electronic cigarettes (ECIGs) aerosolize an often nicotine-containing solution for user inhalation. ECIG nicotine delivery may depend on liquid nicotine concentration and user puffing behavior (topography). This study examined the relationship among liquid nicotine concentration, puff topography, and plasma nicotine concentration. Thirty-three ECIG-experienced and 31 ECIG-naïve individuals completed four laboratory sessions that differed by ECIG liquid nicotine concentration (0, 8, 18, or 36 mg/ml). Little is currently known about the ability of electronic cigarettes to manage tobacco withdrawal symptoms and their abuse liability. In the current study eight conventional cigarette smokers completed nine within-subject study sessions. In the first session participants practiced using an electronic cigarette containing 16 mg/ml of nicotine over six 10-puff bouts. Remaining study sessions were comprised of four two-day blocks (one for each condition), which assessed measures of tobacco withdrawal symptoms and abuse liability following unrestricted cigarette smoking and 24-hour tobacco deprivation. Smoking and tobacco dependence are serious health concerns in the United States and globally. Reward via the pharmacological effects of nicotine are believed to be the principal motivating factor that drive tobacco dependence. Research reveals differences in sensitivity between males and females to the motivational effects of nicotine in tobacco use. Enhancement of reinforcement value of non-nicotine rewards contributes to overall nicotine reward. Similar value-enhancing effects have been observed by the two most commonly prescribed smoking cessation aids, bupropion and varenicline. Nicotine has both unconditioned and conditioned stimulus properties. Conditioned stimulus properties of nicotine may contribute to the tenacity of nicotine addiction. The purpose of this experiment was to use neurohistochemical analysis of rapidly developing c-Fos protein to elucidate neurobiological loci involved in the processing of nicotine as an interoceptive conditioned stimulus (CS). The Effects Of Nicotine On Gambling Behavior Of Smoking And Nonsmoking Undergraduate Students, Ellen Meier, Jeffrey N. Weatherly

Analysis of Gambling Behavior

Gambling and smoking have been linked in the literature. The present study recruited smokers and nonsmokers to gamble on a slot machine after they chewed nicotine or non-nicotine gum. Re-sults showed that gambling behavior, both in terms of persistence and risk taking, did not differ as a function of either smoking status or type of gum the participants chewed. Although the present study has a number of limitations, the results highlight that factors correlated with gambling do not necessarily lead to differences in gambling behavior when people actually gamble.
