The Effects Of Social And Nonsocial Contextual Stimuli On The Renewal Of Cocaine Seeking, Bree Humburg

Those with substance use disorders can undergo craving and relapse when re-exposed to a drug-associated context. This study determined if renewal of cocaine seeking is differentially controlled by contexts consisting of social and/or nonsocial stimuli. Experiment 1, rats self-administered cocaine in Context A which included a social peer and house light illumination. Following self-administration, rats were randomly assigned to an AAA or ABA group for extinction and renewal. For the AAA rats, context was similar to self-administration; for ABA rats, the drug-associated stimuli (peer and house light) were removed (Context B). Cocaine Use Disorder (CUD) is characterized partly by the use of cocaine at the expense of other alternatives, in other words, it is a decision-making pathology (Kalivas & Volkow, 2005). Concurrent choice tasks assess decision-making in a dynamic scenario that more closely resembles real life. Value-based decision-making is an important facet of understanding the addictive properties of drugs of abuse. In order to compare two value-based theories of addiction (habit theory and relative value theory), a concurrent choice task was run in tandem with an escalation procedure. Drug-specific reward and associated effects on neural signaling are often studied between subjects, where one group self-administers drug and a separate group self-administers a natural reinforcer. However, exposure to drugs of abuse can cause long-term neural adaptations that can affect how an organism responds to drug reward, natural reward, and their reward-associated stimuli. Thus, to isolate drug-specific effects it is important to use models that expose the same organism to all of the aforementioned. Until recently, there were few rodent models available to study the interaction of post-traumatic stress disorder (PTSD) and drug taking. Like PTSD, single prolonged stress (SPS) produces hypothalamic-pituitary-adrenal (HPA) axis dysfunction and alters psychostimulant self-administration. Other stressors, such as isolation stress, also alter psychostimulant self-administration. However, it is currently unknown if isolation housing combined with SPS can alter the acquisition or maintenance of cocaine self-administration. While preclinical self-administration research has provided invaluable insight into the neurobehavioral mechanisms that underlie cocaine abuse, cocaine use outside of the laboratory occurs within an environment where other goods are also available ubiquitously. Estrogens seem to play a role in the locomotor activating effects of cocaine. Japanese quail provide a good model for hormonal manipulation as alterations of their photoperiod controls hormone levels. The current study aims to examine the role of early life photoperiod manipulation in cocaine-induced behavioral sensitization in quail. It was expected that if quail were raised on a short photoperiod, they would have a reduction in gonadal hormones and this reduction in hormones would affect the acquisition of cocaine-induced behavioral sensitization. Background—Numerous studies in behavioral economics have demonstrated that individuals are more sensitive to the prospect of a loss than a gain (i.e., loss aversion). Although loss aversion has been well described in “healthy” populations, little research exists in individuals with substance use disorders. This gap is notable considering the prominent role that choice and decision-making play in drug use. The purpose of this pilot study was to evaluate loss aversion in active cocaine users.

Background: Early exposure to enriched environments has been shown to decrease the locomotor effects induced by repeated injections of cocaine and modify basal and cocaine-induced total protein levels of the transcription factor ΔFosB in the whole striatum of mice. In this study, we aimed at characterizing whether the profile of ΔFosB accumulation induced by enriched environments and cocaine would be similar or different in terms of brain areas and cell type.

Background: Cocaine is the number one abused psychostimulant drug that reaches addiction criterion in the US. In animals, repeated administration of cocaine results in behavioral sensitization which is thought to represent adaptations in the mesolimbic dopamine neural circuitry, the reward pathway. Cocaine-induced behavioral sensitization is evident in rodents and quail when cocaine is administered intraperitoneally (IP).

Cocaine users display impaired inhibitory control. The influence of cocaine-related stimuli on inhibitory control has not been assessed. The Attentional Bias-Behavioral Activation (ABBA) task uses cocaine and neutral images as cues to determine if drug-related images impair inhibitory control in cocaine users. This dissertation was designed to assess the influence of cocaine images on inhibitory control in cocaine users through the conduct of studies designed to address four aims. Research has indicated that gonadal hormones may mediate behavioral and biological responses to cocaine. Estrogen, in particular, has been shown to increase behavioral responding to cocaine in female rats relative to male rats. The use of Japanese quail may add to our knowledge of sex differences in drug abuse because of their advanced visual system and the ability to control their gonadal hormones via alterations in photoperiod. In three experiments, cocaine-induced behaviors were examined using this avian model.

Rodents with the propensity to sign track are thought to be vulnerable to drug abuse. Heightened attentional bias towards cocaine-related stimuli relative to neutral stimuli is a characteristic observed in cocaine-use disorders. Response time is an indirect measure of attention and research has failed to consistently demonstrate evidence of clinical relevance. Eye tracking presents a novel tool for directly measuring attentional allocation. The aim of this dissertation was to assess the sensitivity, reliability, and specificity of attentional bias through fixation and response time during the visual probe task.

In the visual probe task, substance-related and matched neutral images were presented side-by-side on a computer screen. Eye-tracking technology measured time spent fixating on each image. A …
