Interactive Effects Of Co2, Temperature, And Nitrate Limitation On The Growth And Physiology Of Marine Cyanobacterium Synechococcus Sp. Ccmp 1334, Alyssa K. Sharbaugh

The marine cyanobacterium Synechococcus sp. CCMP 1334 was grown in a continuous culture system on a 12:12 h light:dark cycle at all combinations of low and high pCO2 (400 and 1000 ppmv, respectively), nitrate availability (nitrate-limited and nutrient-replete conditions), and temperatures of 21°C, 24°C, 28°C, 32°C, and 35°C. The maximum median nutrient-replete growth rate was ~1.15 d−1 at 32 –35°C. Median growth rates at 1000 ppmv pCO2 were higher than those at 400 ppmv at all temperatures, but most of the differences were statistically insignificant. The refractive index of phytoplankton has never been directly measured before despite its importance towards studying the optical characteristics of marine particles. Previous attempts to measure it have been done through indirect methods. While these methods have proven useful, they contain assumptions about the particles morphology and composition that cause for uncertainty with the measurements. Through the use of the 3D Cell Explorer, a high precision holotomographic microscope, the RI of phytoplankton can be directly measured for the first time. Upward advection or mixing of iron-rich deep waters due to circulation changes driven by the rate of basal ice shelf melt was shown to be a primary control on chlorophyll a production in coastal polynyas over the Antarctic continental shelf. Here, the effects of atmospheric changes projected in 2100 on this relationship were examined with a 5-km resolution ocean/sea ice/ice shelf model of the Southern Ocean with different simulated dissolved iron sources and idealized biological uptake. Tidal Mixing Fronts (TMFs) are prominent hydrographic features of tidally energetic shallow shelf seas, representing the transition from mixed to stratified waters. These frontal boundaries often host enhanced phytoplankton primary productivity, as complete vertical mixing exhumes nutrients from depth to the light-lit surface. Existing observational programs for locating TMFs include infra-red satellite imagery of sea surface temperature (SST) and vertical profiling of temperature and density. However, challenges in observationally distinguishing mixed from mixing using only conservatively mixed hydrographic properties persist. The Southern Ocean, which flows around the Antarctic continent, is home to vast numbers of unique and remarkable animals, including penguins, albatrosses, petrels, seals, and whales. Constraining the role of dust deposition in regulating the concentration of the essential micronutrient iron in surface ocean waters requires knowledge of the flux of seawater-soluble iron in aerosols and the replacement time of dissolved iron (DFe) in the euphotic zone. Here we estimate these quantities using seasonally resolved DFe data from the Bermuda Atlantic Time-series Study region and weekly-scale measurements of iron in aerosols and rain from Bermuda during 2019. The primary nitrite maximum (PNM) is a ubiquitous feature of the upper ocean, where nitrite accumulates in a sharp peak at the base of the euphotic zone. This feature is situated where many chemical and hydrographic properties have strong gradients and the activities of several microbial processes overlap. Near the PNM, four major microbial processes are active in nitrite cycling: ammonia oxidation, nitrite oxidation, nitrate reduction and nitrite uptake. The first two processes are mediated by the nitrifying archaeal/bacterial community, while the second two processes are primarily conducted by phytoplankton. Particulate phases transport trace metals (TM) and thereby exert a major control on TM distribution in the ocean. Particulate TMs can be classified by their origin as lithogenic (crustal material), biogenic (cellular), or authigenic (formed in situ), but distinguishing these fractions analytically in field samples is a challenge often addressed using operational definitions and assumptions. These different phases require accurate characterization because they have distinct roles in the biogeochemical iron cycle. The growth of diatoms in the Southern Ocean, especially the region surrounding the West Antarctic Peninsula, is frequently constrained by low dissolved iron and other trace metal concentrations. This challenge may be overcome by mutualisms between diatoms and co-occurring associated bacteria, in which diatoms produce organic carbon as a substrate for bacterial growth, and bacteria produce siderophores, metal-binding ligands that can supply diatoms with metals upon uptake as well as other useful secondary compounds for diatom growth like vitamins. It is widely assumed that phytoplankton abundance and productivity decline during temperate winters because of low irradiance and temperatures. However, winter phytoplankton blooms commonly occur in temperate estuaries, but they are often undocumented because of reduced water quality monitoring in winter. The small body of in situ work that has been done on winter blooms suggests they can be of enormous consequence to ecosystems. The California Current System is a diatom‐dominated region characterized by seasonal coastal upwelling and additional elevated mesoscale activity. Cyclonic mesoscale eddies in the region trap productive coastal waters with their planktonic communities and transport them offshore with limited interaction with surrounding waters, effectively acting as natural mesocosms, where phytoplankton populations undergo ecological succession as eddies age. Phytoplankton exhibit diverse physiological responses to temperature which influence their fitness in the environment and consequently alter their community structure. Here, we explored the sensitivity of phytoplankton community structure to thermal response parameterization in a modelled marine phytoplankton community. Using published empirical data, we evaluated the maximum thermal growth rates (μmax) and temperature coefficients (Q10; the rate at which growth scales with temperature) of six key Phytoplankton Functional Types (PFTs): coccolithophores, cyanobacteria, diatoms, diazotrophs, dinoflagellates, and green algae. Coastal environments such as seagrass meadows span a wide range of spatial scales and can experience disturbances that cause rapid shifts to ecosystem dynamics. Increased pressure to the coastal zone has highlighted the need for constant, real-time monitoring to monitor current ecosystem status. A custom, low cost, high resolution, in situ sensor network was designed and tested in the Lower Laguna Madre, Texas to characterize primary productivity in seagrass meadows through gradient patterns of dissolved oxygen concentration, with the purpose of determining whether seagrass or phytoplankton were the more dominant primary producer. Fully and accurately reconstructing changes in oceanic productivity and carbon export and their controls is critical to determining the efficiency of the biological pump and its role in the global carbon cycle through time, particularly in modern CO2 source regions like the eastern equatorial Pacific (EEP). Here we present new high-resolution records of sedimentary 230Th-normalized opal and nannofossil carbonate fluxes and [231Pa/230Th]xs ratios from site MV1014-02-17JC in the Panama Basin. As primary producers, phytoplankton play an integral role in global biogeochemical cycles through their production of oxygen and fixation of carbon. They also provide significant ecosystem services, by supporting secondary production and fisheries. Phytoplankton biomass and diversity have been identified by the Global Ocean Observing System (GOOS) as Essential Ocean Variables (EOVs), properties that need to be monitored to better understand and predict the ocean system. Phytoplankton identification and enumeration relies on the skills and expertise of highly trained taxonomic analysts. The Mississippi Bight (MSB) is a river-dominated continental margin influenced by multiple large river systems, including the Mississippi River, Alabama and Tombigbee rivers via Mobile Bay, and numerous smaller rivers, creeks, and bayous. This is part of a biologically-rich ecosystem that supports the second largest fishery industry by volume in the United States. Alexandrium tamerense is a well-studied dinoflagellate known for its ability to produce the neurotoxin that causes paralytic shellfish poisoning. Until 1970 Alexandrium tamerense was only found in Europe, North America, and Japan but has been increasingly found all over the globe. Alexandrium is characteristically found in temperate and subtropical regions and as the Arctic warms, there is considerable concern that it may be expanding into the Arctic. The aggregation of individual phytoplankton into marine snow allows particles to sink more quickly, thus resulting in the transport of particulate organic carbon from surface waters to the deep ocean. Aggregate formation has previously been measured in experiments indirectly by quantifying how particle size or particle concentration changes over time. Here, I present my thesis in which I applied high-resolution imaging to quantify aggregate formation using two different methodologies.

Phytoplankton productivity and export sequester climatically significant quantities of atmospheric carbon dioxide as particulate organic carbon through a suite of processes termed the biological pump. Constraining how the biological pump operated in the past is important for understanding past atmospheric carbon dioxide concentrations and Earth's climate history. However, reconstructing the history of the biological pump requires proxies. The expected reduction of ice algae with declining sea ice may prove to be detrimental to the Pacific Arctic ecosystem. Benthic organisms that rely on sea ice organic carbon (iPOC) sustain benthic predators such as the Pacific walrus (Odobenus rosmarus divergens). Phytoplankton iron contents (i.e., quotas) directly link biogeochemical cycles of iron and carbon and drive patterns of nutrient limitation, recycling, and export. Ocean biogeochemical models typically assume that iron quotas are either static or controlled by dissolved iron availability. We measured iron quotas in phytoplankton communities across nutrient gradients in the Pacific Ocean and found that quotas diverged significantly in taxon‐specific ways from laboratory‐derived predictions. Iron quotas varied 40‐fold across nutrient gradients, and nitrogen‐limitation allowed diatoms to accumulate fivefold more iron than co‐occurring flagellates even under low iron availability. The Kuroshio current separates from the Japanese coast to become the eastward flowing Kuroshio Extension (KE) characterized by a strong latitudinal density front, high levels of mesoscale (eddy) energy, and high chlorophyll a (Chl). While satellite measurements of Chl show evidence of the impact of mesoscale eddies on the standing stock of phytoplankton, there have been very limited synoptic, spatially resolved in situ estimates of productivity in this region. Aquatic stressors are known to cause biological impairment that can result in biodiversity loss in several Marine Protected Areas (MPA), including in Menjangan Island, West Bali National Park. Electromagnetic theory predicts spectral dependencies in extinction efficiency near a narrow absorption band for a particle with an index of refraction close to that of the medium in which it is immersed. These absorption band effects are anticipated in oceanographic beam-attenuation (beam-c) spectra, primarily due to the narrow red peak in absorption produced by the phytoplankton photopigment, chlorophyll a (Chl a). Here we present a method to obtain Chl a absorption and size information by analyzing an eigendecomposition of hyperspectral beam-c residuals measured in marine surface waters by an automatic underway system. In this study, phytoplankton community and carbon dynamics were examined in the optically complex estuarine-coastal regions of the northern Gulf of Mexico (nGOM) from field and satellite ocean color observations. As part of this study, bio-optical ocean color algorithms for i) dissolved organic carbon (DOC), ii) phytoplankton pigment composition, iii) adaptive estimation of Chl a and iv) phytoplankton size fractions were developed to facilitate the study of biogeochemical cycling in the nGOM.

Harmful algal blooms (HABs) are increasing globally in frequency, persistence, and geographic extent. HABs pose a threat to economic stability, and ecosystem and human health. To date no incidences of marine toxins produced by phytoplankton have been recorded in Mozambique, which may be due to the absence of a monitoring program and general awareness of the potential threat. Sulfur (S) is a major heteroatom in organic matter. This project evaluated spatial variability in the concentration and molecular-level composition of organic sulfur along gradients of depth and latitude. We measured the concentration of total organic sulfur (TOS) directly from whole seawater. Our data reveal high variability in organic sulfur, relative to established variability in total organic carbon or nitrogen. The deep ocean contained significant amounts of organic sulfur, and the concentration of TOS in North Atlantic Deep Water (NADW) decreased with increasing age while total organic carbon remained stable. Phytoplankton accessory pigments are commonly used to estimate phytoplankton size classes, particularly during development and validation of biogeochemical models and satellite ocean color-based algorithms. The diagnostic pigment analysis (DPA) is based on bulk measurements of pigment concentrations and relies on assumptions regarding the presence of specific pigments in different phytoplankton taxonomic groups. Of all the Antarctic coastal polynyas, the Amundsen Sea Polynya is the most productive per unit area. Observations from the 2010-2011 Amundsen Sea Polynya International Research Expedition (ASPIRE) revealed that both light and iron can limit the growth of phytoplankton (Phaeocystis antarctica), but how these controls manifest over the bloom season is poorly understood, especially with respect to their climate sensitivity. Using a 1-D biogeochemical model, we examine the influence of light and iron limitation on the phytoplankton bloom and vertical carbon flux at 12 stations representing different bloom stages within the polynya. Previous studies showed that satellite‐derived estimates of chlorophyll a in coastal polynyas over the Antarctic continental shelf are correlated with the basal melt rate of adjacent ice shelves. A 5‐km resolution ocean/sea ice/ice shelf model of the Southern Ocean is used to examine mechanisms that supply the limiting micronutrient iron to Antarctic continental shelf surface waters. Four sources of dissolved iron are simulated with independent tracers, assumptions about the source iron concentration for each tracer, and an idealized summer biological uptake. Iron from ice shelf melt provides about 6% of the total dissolved iron in surface waters. The contribution from …
