Dissolved organic matter (DOM) is an extremely complex mixture of organic molecules ubiquitous in aquatic systems and a critical component of the global carbon cycle. Little is known about DOM structural composition at the molecular level. The work presented in this dissertation summarizes the development of a novel analytical toolbox based on trapped ion mobility spectrometry and Fourier transform ion cyclotron resonance mass spectrometry (TIMS-FT-ICR MS) that has significantly contributed to expand our knowledge of DOM molecular complexity and diversity. DREAM belongs to the NCS family, and it is involved in several physiological processes mediated by Ca2+ association, which produces changes in its oligomerization state. The mechanism of how Ca2+ binds to DREAM remains unknown. A kinetic study of the binding of Ca2+ to DREAM suggested that DREAM dimerization is a two-step process, the first step( τ1= 8±0.1ms) is associated with the binding of Ca2+ to the monomer, while the second step (τ2 = 3.6 ±0.4 s ) corresponds to a conformation relaxation that leads to the dimerization. The lack of solubility of the chitosan in organic solvents hinders its potential for chemical modifications. For this reason, it was hypothesized that the insertion of the phenyl carbamoylated guanidine moiety could lead to increased solubility in DMSO and new potential modifications and uses.

This research involved the synthesis of S-methyl phenyl carbamoylated guanidine (SMPCG) starting from the commercially available S-methyl isothiourea. This was followed by the reaction of the SMPCG with chitosan to yield phenyl carbamoylated guanidine functionalized chitosan (PCGCs) with a degree of substitution of 14%.

Protein-based therapeutics have gained significant attention for treating or curing various diseases, especially those untreatable by small molecule-drugs. Despite their widespread success and adoption, the cellular membrane remains a major obstacle for proteins to reach the inner compartments of the cell and have had limited applicability spanning only extracellular targets. Consequently, the efficient intracellular delivery of proteins offers tremendous potential to reach new, unexplored, and otherwise “undruggable” targets. The characterization of crude oils at the compositional level remains challenging due to their high chemical complexity and isomeric diversity. The present work expands the analytical separation space of chemical components in crude oils by coupling gas chromatography (GC), trapped ion mobility spectrometry (TIMS), and mass spectrometry (GC-TIMS-MS and TIMS-FT-ICR MS) for the effective separation and identification of isomeric compounds. The coupling of GC and TIMS-MS platform enhanced the peak capacity and showed complementary isomeric separation. The goal of this project was to develop a fast, microwave-based extraction technique that could be employed for direct and rapid DNA analysis. The hypothesis was that the use of a microwave could increase the yield of DNA by opening the cell membrane, rendering the DNA available without the use of any other chemical treatment, and improving results from very low quantity samples.

The development of an aptamer-based voltammetric sensor and biosensor is described in this dissertation. The mammalian high-mobility-group protein AT-hook 2 (HMGA2) is a small DNA-binding protein and consists of three positively charged “AT-hooks” and a negatively charged C-terminal motif. It is a multifunctional nuclear protein linked to obesity, human height, stem cell youth, human intelligence, and tumorigenesis. Previous results showed that HMGA2 is a potential therapeutic target of anticancer and anti‐obesity drugs through inhibiting its DNA‐binding activities. Here a miniaturized, automated AlphaScreen ultra‐high‐throughput screening assay is developed to identify inhibitors targeting HMGA2‐DNA interactions. After screening the LOPAC1280 library, several compounds are identified that strongly inhibit HMGA2‐DNA interactions including suramin, a negatively charged antiparasitic drug. …

The dinoflagellate Karenia brevis, blooms annually in the Gulf of Mexico, producing a suite of neurotoxins known as the brevetoxins. The cellular toxin content of K. brevis, however, is highly variable between or even within strains. I investigated biochemical differences between high (KbHT) and low (KbLT) toxin producing cultures both derived from the Wilson strain, related to energy-dependent quenching (qE) by photosystem II, and the content of reduced thiols of the proteome. Modified nucleosides gained great attention as potential anticancer and antiviral therapeutics. In this dissertation, synthesis and reactivity of (β-iodovinyl)sulfone and β-ketosulfone groups incorporated into purine nucleosides at C8 or C2 positions and DNA incorporation of their 5' triphosphates have been developed. Moreover, synthesis of novel antibiotic arsinothricin (AST) as well as polycyclic aromatic hydrocarbons (PAHs) have been discussed. The 8-(1-iodo-2-tosylvinyl)-2'-deoxyadenosine and 8-(1-Iodo-2-tosylvinyl)adenosine were synthesized employing iodovinylsulfonation of 8-ethynyl precursors with TsNa/I2/NaOAc. The rhodizonate dianion is of importance for forensic testing, specifically with regard to the sodium rhodizonate test for lead in gunpowder residues. In an effort to understand this test from a chemical perspective, the coordination of rhodizonate and its decomposition products, croconate and oxalate, with several metals has been investigated with a variety of structural and spectroscopic methods.

Improving the accuracy and reliability of odor detection dogs is of utmost importance particularly for legal reasons. Field testing in conjunction with headspace analysis of volatile organic compounds (VOCs) has in recent times allowed for these improvements, by providing scientifically based recommendations for optimum training protocols. A field study was conducted to observe the natural development of cannabinoids in three daylength sensitive industrial hemp varieties Bubba Kush (BK), Emerald Flower (EF), and Golden Sunset (GS). Plants were configured in a randomized block design with 3 replications. Once 50% of the plants within a variety reached reproductive growth, plants were sampled weekly until senescence and analyzed through a HPLC-UV/DAD. The results from the study indicate that all three varieties of industrial hemp tested in field study reached reproductive growth within the first week of transplanting. Most production of Technetium-99 (99Tc) occurred as a byproduct of development of legacy nuclear weapons research. Singlet oxygen, 1O2, can be easily generated by a sensitizer, solar radiation, and molecular oxygen. 1O2 is very important in a range of biological and environmental systems. The products and mechanisms of the 1O2 reactions are well studied in organic solvents, and despite well-established solvent effects, studies on the reactions of the singlet oxygen in aqueous media are limited. Herein, 1O2 reactions with biologically and environmentally relevant conjugated π systems in aqueous solutions are presented. A great challenge facing humanity in the 21st century is finding inexhaustible and inexpensive energy sources to power the planet. Renewable energies are the best solutions because of their abundance, diversity, and pollution-free emission. Solar energy is the cleanest and most abundant renewable energy source available. In the continuing quest for efficient and low-cost solar cells, perovskite solar cells (PSCs) have emerged as a potential replacement for silicon solar cells. Surface Enhanced Raman Spectroscopy (SERS) is an analytical technique in which nanostructured substrates amplify the inherently weak Raman signal of an adsorbed species by several orders of magnitude, enabling the detection of trace compounds, up to the single molecule level. While this may be an exceptional tool for any analytical scientist, SERS is at present relegated to the role of academic sensation, and is underutilized in everyday analytical practice. The use and abuse of synthetic cannabinoids has increased significantly in recent years due to their easy access and growing popularity. Despite having more known drugs become illegal, new synthetic versions of these drugs are being made that have not yet been recognized or classified as illegal substances. Therefore, standard methods may not be able to detect these drugs.

Spent nuclear fuel (SNF) – or used nuclear fuel (UNF) – contains long-lived minor actinides such as 241Am, 245Cm, and 237Np, together with fission products that include lanthanides. Minor actinides are responsible for much of the radiotoxicity and heat generation that limits the capacity of geological repositories. Thus, removing minor actinides from UNF can reduce storage time required for decay to natural levels of activity by several orders of magnitude.
