Bacterial Chondronecrosis With Osteomyelitis In Broilers: Genomics, Phylogenomics, And Methods To Detect Specific Pathogens During Outbreaks., Abdulkarim Abdulaziz A. Shwani

Graduate Theses and Dissertations

Lameness is a major issue in animal welfare and the broiler industry. Bacterial chondronecrosis with osteomyelitis (BCO) is one of the main causes of lameness. Many staphylococcal species, including Staphylococcus agnetis isolate 908, have been isolated from the bones and blood of lame broilers at the University of Arkansas. Staphylococcus agnetis is a coagulase-variable, Gram-positive bacterial species that has been previously associated with subclinical or mild clinical cases of mastitis in dairy cattle. The annotated complete genome of hypervirulent strain 908 was published at NCBI. In this study, it has been compared to nine genomes we assembled for hypervirulent isolates …

Preparation And Evaluation Of Single-Stranded Dna Aptamer-Based Immunological Adjuvant In Broiler Chickens, Adil Sabr Al-Ogaili

Graduate Theses and Dissertations

Mineral oils and metal salts are commonly used as adjuvants to enhance acquired immunity. Recently, monoclonal antibodies (MAbs) and recombinant peptides agonist CD40 receptor have shown remarkable promise for induction of rapid and robust immune responses. Limitations of this approach MAb production costs and multiple administrations due to anti-MAb immune responses. Here we demonstrate the development of a unique and sophisticated DNA aptamer-based alternative for CD40-directed delivery of universal antigens as an alternative in chickens, and potentially other vertebrate species. This receptor, expressed by antigen-presenting cells, acts as a costimulatory molecule for activated T helper lymphocytes. After initially selecting for …

Quantitative Microbial Risk Assessment For Parts, Ground, And Msc Poultry Product Including Intervention Analysis And Exploration Of Enterobacteriaceae As An Indicator Organism In Poultry Processing, Leigh Ann Parette

Graduate Theses and Dissertations

Samples collected at five different large bird poultry processing facilities over a period of 7 months from prescald to post debone locations were enumerated for Enterobacteriaceae, Salmonella spp., and Campylobacter spp. and the results were used to create Quantitative Microbial Risk Analyses (QMRA) models for parts, ground, and mechanically separated chicken (MSC) products. Sensitivity analyses indicated the points in the process at which reductions would be most advantageous to the endpoint and simulation models were run to test reductions required to meet the current USDA performance standards.

These data were analyzed to determine the reductions from one node (location) to …

Effect Of Endophyte-Infected Tall Fescue Toxins On Growth Performance And The Microbial Community In The Rumen And Feces In Pregnant Ewes, Saleh Alrashedi Dec 2017

Effect Of Endophyte-Infected Tall Fescue Toxins On Growth Performance And The Microbial Community In The Rumen And Feces In Pregnant Ewes, Saleh Alrashedi

Graduate Theses and Dissertations

Fifty pregnant Katahdin ewes grazed pastures with high (HE) and moderate (MI) levels of Neotyphodium coenophialum-infected tall fescue between February 1 and May 24 to determine the impacts of tall fescue toxins on body weight (BW), body condition scores, FAMACHA scores, fecal egg counts (FEC), and serum Cu, Zn, prolactin, and non-esterified fatty acids (NEFA). Within those 50 ewes, 15 were selected randomly for fecal and rumen sampling to assess the impacts of HE and MI on the microbiome community. Total bacterial DNA was extracted from rumen and fecal samples, and alpha and beta diversities were used to relate microbiome …

Evaluation And Selection Of A Bacillus Based Direct-Fed Microbial Candidate For In Situ Enzyme Production To Improve Gut Health Integrity, Bone Quality And Growth Performance In Poultry, Juan David Latorre Cardenas

Graduate Theses and Dissertations

During the last decade, the increasing interest in renewable energy sources has been changing the distribution of corn utilization from human and animal consumption to biofuel production, leading to a continuous rise in feed costs of livestock diets. Therefore, alternative feed ingredients such as distillers dried grains with solubles (DDGS), as well as cereals like wheat, barley, and sorghum have become part of the feed matrix to maintain or reduce production costs. However, these raw materials often contain a higher concentration of antinutritional factors in comparison to corn, including non-starch polysaccharides (NSP) which increase digesta viscosity and reduce nutrient absorption …

Evaluation Of Porcine Ipec-J2 Cell Line Growth Rate And Immune Response To E. Coli (0111:B4) Lps, Xiaofan Wang

Graduate Theses and Dissertations

The objective of this study was to evaluate the intestinal porcine epithelial cell-jejunum 2 (IPEC-J2) cell line as a model to study the innate immune function of live pigs. Growth rates of IPEC-J2 cells in T-75 flasks and 96-well plates were evaluated using a hemocytometer and spectrophotometer for cell quantity measurements to determine growth rate and doubling time. Growth rates of IPEC-J2 cells in T-75 flasks and 96-well plates were 0.4016 and 0.2851 times of doubling/day respectively with a doubling time of 1.73 d and 2.43 d, respectively. Confluent IPEC-J2 monolayers were tested at five time intervals (0, 1, 2, …

Linoleic acid isomerase (LAI) is the enzyme that converts linoleic acid to conjugated linoleic acids (CLAs) which appear to be capable of lowering cancer risk and enhancing immunity. There is an interest in developing commercial processes for the production of single isomers of CLA by biotransformation of LA using microbial cells and enzymes. However, the evaluation of these strains suggest that growth and LA isomerase production levels by these anaerobes are insufficient to support economic commercial production of single CLA isomers. A better alternative is to clone the isomerase gene and to generate new production strains using recombinant technology. Bifidobacterium …
