Nicotine dependence is causally linked to increased risk of mood/anxiety disorders in later life. Females are reported to experience a higher prevalence of anxiety/depressive disorders and challenges in smoking cessation therapies, suggesting a potential sex-specific response to nicotine exposure and mood/anxiety disorder risk. However, pre-clinical evidence of sex-specific responses to adolescent nicotine exposure is unclear. Thus, to determine any sex differences in anxiety/depressive-related outcomes, adolescent male and female Sprague Dawley rats received nicotine (0.4 mg/kg; 3x daily) or saline injections for 10 consecutive days, followed by behavioural testing, in-vivo electrophysiology and Western Blot analyses. A single bout of exercise improves executive function (EF); however, the physiological mechanism(s) contributing to this benefit are unclear. One candidate mechanism for the benefit is an exercised-mediated increase in cerebral blood flow (CBF) that improves neural efficiency. In my thesis, I conducted two experiments to assess the relationship between an increase in CBF and EF. In Experiment 1, I examined passive exercise, and in Experiment 2, I examined a 2.5% hypercapnic environment given that both interventions increase CBF independent of the metabolic demands of volitional muscle activity. Individuals with mild traumatic brain injury (mTBI) can experience high levels of emotional distress, leading to decreased quality of life and increased health care costs. Bowen's feasibility framework was used to examine the feasibility, acceptability, and limited efficacy of an innovative clinician-guided internet-delivered cognitive behavioural therapy program (ICBT) specialized for people with mTBI to improve their wellbeing. The data illustrated strong adherence and program satisfaction from participants. Likewise, statistically and clinically significant changes in symptoms of unhappiness and anxiety were found, along with improved self-efficacy and quality of life. Clinical and preclinical studies indicate prenatal cannabis exposure (PCE) pathologically affects fetal brain development and may increase vulnerability to neuropsychiatric disorders, including schizophrenia and mood/anxiety disorders. In review research from our lab suggests that fetal exposure to Δ9-THC sex-selectively impairs mesocorticolimbic (MCL) circuit function. However, there is a distinct lack of focus on PCE models on the BLA. The BLA plays a central role within the MCL where it directly interacts with the VTA, PFC and HIPP. The enteric bacterial metabolite, propionic acid (PPA), elicits physiological and behavioural changes in rodents reminiscent of autism spectrum disorder (ASD). This includes abnormal sensory processing and social behaviour. ASD may contribute to social deficits through impaired habituation; therefore, the present study examined the effects of intraperitoneal PPA on the habituation to social and non-social odours. Adult male rats were injected daily with PPA or the vehicle control, and for 3 days, habituated to a conspecific odour or vanilla extract for 10 minutes. Anticipatory Nausea (AN) is a form of classical conditioning in which the effects of a nausea-inducing toxin become associated with a specific environmental context. AN is often experienced by individuals receiving chemotherapy treatment, whereby the emetic effects of the chemotherapy agents become associated with the treatment context (i.e. hospital, clinic), such that exposure to the context alone can cause an individual to experience nausea and potentially withdraw from treatment. In ‘targeted memory reactivation’ (TMR) paradigms, information learned during wakefulness is paired with a cue, and reactivated during sleep by presenting that same cue. TMR improves memory. In a prior study (Antony et al., 2012), participants learned two melodies. One melody was cued during a nap, and performance was better than for the uncued melody. The current study reanalyzed these data to characterize sleep spindle density during TMR cue-periods relative to non-cued periods, and whether spindle density correlated with performance. Eating disorders (EDs) are serious mental illnesses often with poor prognosis. Personalizing evidence-based treatments based on an individual’s reasons for engaging in ED behaviours – or the functions of EDs – may improve treatment outcomes; however, no validated measures assessing these functions exist. Age-related cognitive decline greatly impacts quality of life for older adults. Previous research has indicated that meditation may act as a neuroprotective factor to prevent age-related cognitive decline. This thesis sought to replicate previous findings and investigate if a four-week meditation intervention would improve sustained attention. Participants 60 years and older (n=27, 17 female) were recruited and assigned to a focused-attention (FA) meditation or relaxation group which met for four weeks, three times a week. Attention is the cognitive processing that facilitates the ability to target and attend to relevant environmental stimuli, while filtering out irrelevant or distracting stimuli. Control over selective attention is theorized to be dependent on organized neural communication that stems from the medial prefrontal cortex (mPFC). To evaluate selective and sustained attention, mice were trained on the novel touchscreen rodent continuous performance task (rCPT), a task designed to emulate the human CPT. Background: The mesolimbic dopamine system consists of dopamine neuron projections from the ventral tegmental area (VTA) to the nucleus accumbens (NAc). The NAc regulates VTA dopamine release through inhibitory GABA projections to the VTA. Hyperactive mesolimbic dopamine signaling is implicated in anxiety. Cannabidiol, a compound found in cannabis, demonstrates promising therapeutic potential for anxiety through the regulation of the mesolimbic dopamine system. Previous studies have revealed that cannabidiol infusions into the NAc decreases mesolimbic dopamine activity - potentially through the inhibitory GABA signaling to the VTA. White matter integrity is crucial to healthy executive function, the cognitive domain that enables functional independence. However, in the ageing brain, white matter is highly vulnerable. White matter inflammation increases with age and Alzheimer disease (AD), which disrupts the normal function of white matter. One way that the body actively responds to an impending stressor is by increasing systemic glucocorticoids through the activation of the hypothalamic-pituitary-adrenal (HPA) axis. While it is essential for short-term adaptation to stress, the sustained activation of the HPA axis during chronic stress can be detrimental and is linked to stress-related psychiatric conditions such as anxiety and depression. Patients with Parkinson disease are prescribed dopamine agonists such as pramipexole to improve motor symptoms. Several studies have found that patients taking dopaminergic medication develop impulse control disorders. In contrast, other studies suggest that some behaviors become less impulsive with pramipexole. We evaluated the performance of 20 young, healthy participants who received pramipexole (0.5 mg) and 20 participants who received placebo, on the Go/No-Go, the Stop Signal Task, and the Balloon Analogue Risk Task. We found that the pramipexole group had more timed out Go trials on the Go/No-Go task than the placebo group, suggesting reduced motor impulsivity. There were …

Maternal alcohol consumption during pregnancy results in a spectrum of behavioural and cognitive deficits collectively known as Fetal Alcohol Spectrum Disorders (FASD). Currently, little is know about if and how the external environment may modulate these deficits. I have used C57BL/6 mice to study this interaction between prenatal alcohol exposure and the postnatal environment. Alcohol exposure during synaptogenesis produces high levels of anxiety-like traits and decreased memory performance. Alcohol-exposed mice (and matched unexposed controls) were put in 'environmentally-enriched' conditions of voluntary exercise, physical activities and cognitive stimulation to ascertain the effects of a positive postnatal environment. The nonapeptide, oxytocin (OT), is implicated in a range of behavioural and physiological functions. However, OT's role in sickness behaviours remains unclear. This thesis examined effects of the OT agonist, carbetocin (CBT), and OT antagonist, L-368,899, on anxiety and locomotor sickness-related behaviours and pro-inflammatory cytokines, TNF-a and IL-6, in adult male CD-1 mice. Animals received 2 intraperitoneal treatment injections. The first treatment was carbetocin, L-368,899, or saline, while the second was lipopolysaccharide (LPS) or saline. Sedentary behavior (SB) has been associated with chronic diseases, such as Hypertension and Obesity; the risk appears greater the longer one engages in SB. University Students are a high-risk population for excessive SB due to academic responsibilities. Increasing frequency/length of breaks from sitting is an effective way to attenuate effects of SB. Cigarette smoking is a leading agent for premature morbidity and mortality among the global community. Most individuals surrender to tobacco use disorder due to the inability to cope with cravings and withdrawal symptoms. Exercise appears to provide acute relief. Currently, it is unclear how exercise attenuates these reductions. Furthermore, the available evidence has focused on acute outcomes besides smoking behaviour and is limited to simulated quit attempts. The primary aim of this thesis is to identify whether satisfaction with perinatal health care encounters is associated with lower postpartum weight retention (PPWR). PPWR is the difference between postpartum and pre-pregnancy weight. Principal components analysis confirmed the validity of a summed satisfaction score representing six dimensions: information, compassion, competency, privacy, respect and decision-making. Confounders were identified using a directed acyclic graph. Multivariable linear regression models were constructed using blocks, and backwards elimination. Results reflect a mean PPWR of 2.5 kg, and high satisfaction, with more than 90% of respondents being very satisfied or satisfied on each dimension. The multivariable …

Exclusive breastfeeding is associated with numerous health benefits for both mother and child, and is recommended for the first 6 months of an infant’s life. The purpose of this prospective study was to examine, using a survey-based design, the breastfeeding practices, self-efficacy, and perceived barriers and facilitators of primiparous mothers in London, Ontario. A total of 71 women (Mage = 30.0, SD = 4.3) participated in the study. Women (breastfeeding and non-breastfeeding) were asked to complete online surveys at three time points: < 4 weeks postpartum, 3 months postpartum, and 6 months postpartum. Results indicated that rates of exclusive breastfeeding decreased over time, whereas partial and non-breastfeeding rates increased. Women in the exclusive breastfeeding category reported the greatest levels of breastfeeding self-efficacy at all time points. Lastly, participants identified a number of breastfeeding-related facilitators (e.g., partner support, community services) and barriers (e.g., insufficient milk supply, latching difficulties).

Some blind humans use the reflected echoes from self-produced signals to perceive their silent surroundings. Although the use of echolocation is well documented in animals such as bats and dolphins, comparatively little is known about human echolocation. The overarching goal of the work presented in this thesis was to shed light on some of the basic functions of human echolocation, including the perception of the shape, size, and material. I addressed these aspects of echolocation using behavioural psychophysics and neuroimaging.

To thoroughly characterize any brain mechanism requires an appropriate animal model for invasive studies. An invaluable model system used toward a comprehension of cognitive neurophysiology is the macaque monkey. It is important to delineate similarities and limitations for this model in relation to the human brain and cognition. Associations between awareness and factors including age, BMI, gender, and obesity status were also examined. A total of 53 parent-child dyads participated in the study. Agreement between parent and child subjective measures (self-report) and objectively measured physical activity was examined to determine awareness. Most parents (87%) and children (83%) overestimated child moderate-vigorous physical activity (MVPA) beyond 30 minutes/day. Women diagnosed with Gestational Diabetes (GDM) are more likely to develop Type 2 Diabetes (T2DM). Despite this risk, few women are engaging in diabetes prevention behaviours. Methods: The Protection Motivation Theory (PMT) was employed to design and evaluate a post-GDM diabetes prevention information session. The utility of this theory in predicting intentions and diabetes prevention behaviours was assessed using regression analysis. Results: Two of the four PMT variables, response efficacy and self efficacy were found to be consistent predictors of intention across most of the recommended behaviours, accounting for between 25% and 77% of the variance in intention. We hypothesize that selective lesion of the septohippocampal GABAergic neurons suppresses the altered behaviors induced by an N-methyl-D-aspartate (NMDA) receptor antagonist, ketamine or MK-801. In addition, we hypothesize that septohippocampal GABAergic neurons generate an atropine-resistant theta rhythm that coexists with an atropine-sensitive theta rhythm in the hippocampus. Infusion of orexin-saporin (ore-SAP) into the medial septal area decreased parvalbumin-immunoreactive (GABAergic) neurons by ~80%, without significantly affecting choline-acetyltransferase-immunoreactive (cholinergic) neurons. The theta rhythm during walking, or the immobility-associated theta induced by pilocarpine, was not different between ore-SAP and sham-lesion rats. The current study uses diffusion tensor imaging to test whether individual differences in white matter predict performance on the math subtest of the preliminary Scholastic Aptitude Test (PSAT). Grade 10 and 11 PSAT scores were obtained from 30 young adults (ages 17- 18) with wide-ranging math achievement levels. Tract based spatial statistics was used to examine the correlation between PSAT math scores, fractional anisotropy (FA), radial diffusivity (RD) and axial diffusivity (AD). FA in left parietal white matter was positively correlated with math PSAT scores (specifically in the left superior longitudinal fasciculus, left superior corona radiata, and left corticospinal tract). …
