Fluoridation is the addition of fluoride to a source of drinking water. In St. Louis’s public water supply, the amount of fluoride remains the same since 1953 at 5ppm. Dr. Ralph Rosen and his team of dentists and researchers advocated for the fluoridation of the water because it reduced the frequency of dental caries occurring in children and adults alike, but the public had opinions on the sudden addition of chemicals to their water. Vitamin D3 is an important chemical in the human body, however, many Americans have low levels of this vitamin. There are plenty of oral supplementations for Vitamin D3 deficiency, but those of older age and busy schedules may struggle to meet the minimum requirement. Chemoresistance is a discouraging trend that affects chemotherapeutic agents like 5-fluorouracil (5-FU) and leads to decreased efficacy of treatment. This has led to numerous studies into compound synergism. Glioblastomas are a type of lethal cancer that has shown chemoresistance to therapeutic agents such as 5-fluorouracil. Being the most common and lethal primary brain tumor, research on mechanisms of growth inhibition is vastly important (Lustig, 2018). Multiple Sclerosis (MS) is a neurodegenerative disease that particularly affects the nerves of the central nervous system (CNS). MS results in the degradation of the myelin sheath surrounding the axon, which is crucial for effective transmission of nerve signals. MS can be detected using lumbar punctures to look for elevated oligoclonal levels and using magnetic resonance imaging (MRI). The MRI can screen for plaques in the CNS, indicating the severity and probability of progression of the disease. Plaques are regions of nerves where the myelin has been removed from the nerves, preventing them from conducting the electrical signals necessary for …

Cancer remains as one of the leading causes of death in humans worldwide. Nanotechnology has made great strides in improving treatment for the disease. This work describes a simplistic approach to design self-assembled combination nanomedicines. A facile one-step ion exchange reaction is utilized to combine a chemotherapeutic (phosphonium) cation and photodynamic therapeutic (porphyrin) anion. An aqueous nanomedicine is prepared from the hydrophobic ionic combination drug via a single-step reprecipitation method. Upon conversion to ionic combination drug, improved photophysical properties of porphyrin were observed. These characteristics subsequently led to increased photodynamic therapeutic activity of nanomedicines—explained by greater singlet oxygen quantum yield. …

Chagas disease, classified as a neglected tropical disease (NTD), is caused by the parasite Trypanosoma cruzi and is an underfunded and under researched disease. The only FDA-approved drug in the United States is benznidazole which is expensive, harmful, and only effective in the illness’ acute phase. The goal is to create a more effective, lower costing and less harmful drug by using a condensation reaction to substitute different amines to diethyl squarate (DES) to make cycloalkylsquaramides. Opioid Use Disorder is patterns of opioid use leading to withdrawal, giving up important life events in order to use opioids, and excessive time spent using opioids, to name a few diagnostic criteria. The clinical progression of the disorder involves periods of acute exacerbation and remission that are cyclic in nature. Treatment is most effective when it includes both pharmacological and psychosocial modalities, referred to as medication assisted therapy (MAT). Three drugs used commonly in MAT-based treatment for OUD from oldest to newest include Methadone, Buprenorphine-naloxone, and Naltrexone. The mannose-6-phosphate/insulin-like growth factor 2 (M6P/IGF2) receptor is a transmembrane protein with the ability to sequester growth factors from the extracellular matrix. This behavior links the receptor to tumor suppression. On a structural level, the extracellular portion of the protein is segmented into 15 homologous repeats, which can be divided further into 5 triplet domains, labelled 1-3, 4-6, 7-9, 10-12, and 13-15. Each triplet receptor displays its own unique ligand binding affinity, including the ability to form dimers with triplets on a second M6P/IGF2 receptor. Introduction: Migraines are a leading cause of disability in many people worldwide. There are numerous postulations as to their etiology and various medications that treat them. As crippling as migraines are, it is critical that there are therapies to prevent them rather than just treat them acutely. Both OnabotulinumtoxinA (Botox) and Topiramate have been shown to be beneficial as prophylaxis for migraines. Introduction: Though levodopa-carbidopa therapy remains the gold standard of Parkinson’s disease (PD) treatment to this day, patients often experience increased dyskinetic activity as a result of long-term treatment; known as levodopa-induced dyskinesia (LID); the mechanism of which is not completely understood. In this review, the efficacy of two adjunct treatments for LID are compared: 1) the concomitant administration of amantadine, an antiviral medication with antidyskinetic properties; and 2) a new form of levodopa administration in the form of an intestinal gel delivered straight via a device much like an insulin pump.

The synthesis of air-stable ligand-free nanopalladium hydride (PdNHx) in water was achieved and applied to various organic transformations, including (1) reduction of heteroaromatics such as (iso)quinolines, phenanthrolines, benzotriazole, and naphthyridines, (2) regio- and chemoselective reduction of nitroarenes, (3) alkyne reductions, (4) chemoselective dehalogenation, and (5) base-free detriflation. Both the synthesis of PdNHx and the reductions were carried out under mild conditions in a micellar medium using our designer surfactant PS-750-M, which eliminates the need for toxic organic solvents and extremely high hydrogen pressure and temperature. Notably, PS-750-M is also a replacement of hazardous polar-aprotic solvents. This newer approach for selective …

The purpose of this study is to investigate and determine the role of PEDF in the pathogenesis of diabetic retinopathy as induced by advanced glycation end-products (AGEs). Human retinal pericytes were isolated from donors and will be measured for cell proliferation under the influence of PEDF both in the presence and absence of AGEs. Expected outcomes involve both the maintenance and increased survival of HRPs after treatment with PEDF under stimulated hyperglycemic conditions.

Breast cancer 1, early onset (BRCA1) is a human tumor suppressor gene encoding the BRCA1 protein that maintains genomic integrity. Molecular events may contribute to the loss of BRCA1 function, contributing to site specific tumorigenesis. Loss of BRCA1 function may arise from mutation or epigenetic events. Germline mutations of BRCA1 have been thoroughly implicated in heritable breast and ovarian cancers. More recently, sporadic breast cancer has been shown to be driven by epigenetic alterations to the BRCA1 promoter region, specifically methylation. Previous work in our lab has found diphenyl ether benzylamines showed a successful response with a micromolar concentration of our lead compound to treat the deadly Trypanosamiosis rhodesience. Furthermore, mammalian cell lines saw promising resistance towards damages. The goal of this study was to synthesize a diphenyl thio benzylamine variant. This variant was more active toward T. b. rhodesience but showed more toxicity to both rat 10 (IC50 mM) and human cell lines (HFF, HC-04, U-2 OS, and HEK293). Immunotherapy strategies are very promising treatments for cancer patients. Specifically, Immune checkpoint inhibitor therapy focusing on the PD-1/PD-L1 pathway shows long-lasting positive results in many cancer patients. Unfortunately, not all the patients can benefit from this highly effective treatment. Hence, there is a great need for predictive biomarkers. Immunohistochemical (IHC) staining has been used as a way of predicting patient response, yet shows many problems. For example, IHC utilizes an invasive biopsy and sample fixing, which creates an incomplete and delayed picture of the patient’s biochemistry and the tumor microenvironment, consequently ignoring metastases.

Cisplatin is a chemotherapy compound effective against a variety of cancers. However, it can act as an ototoxin and cause hearing loss by promoting reactive oxygen species (ROS) production in auditory tissues. The antioxidant amino acid, L-serine has been hypothesized to lower levels of cisplatin-mediated ROS. In this project, we investigated whether L-serine can reduce cisplatin-mediated ROS production in auditory tissue and potentially act as an otoprotectant during cisplatin chemotherapy. We used a zebrafish utricular tissue culture system and fluorescent ROS indicator dye to spectrophotometrically measure if L-serine could decrease reactive oxygen species levels in cisplatin-treated tissues. One in eight women in the United States will be diagnosed with breast cancer. Approximately 25-30% of these breast tumors will overexpress Human Epidermal Growth Factor Receptor 2 (HER2), associated with a worse prognosis and decreased survival rate. Stimulation of HER2 results in activation and production of many signaling molecules, ultimately resulting in increased cell division. Two of the downstream molecules activated are PI3Kinase and, further downstream, the molecule Akt. Typically, overproduction and/or overactivation of PI3Kinase can lead to oncogenesis. Smoothened receptor (SMO) is a protein that in humans, is encoded by the SMO gene. A systemic mutation in its binding pocket helps predict the sensitivity of mutant proteins to different drugs. Known as a GPCR-like receptor, it is a component of the hedgehog signaling pathway; a pathway involved in body patterning and the regulation of adult stem cells. An uncontrolled or inappropriate activation of the Hedgehog pathway drives tumor progression in cancers and a number of birth defects.
