Investigation Of The Bactericidal Effects Of Vancomycin And Quinupristin/Dalfopristin On Staphylococcus Aureus Isolates, Mi̇ne Hoşgör Li̇moncu, Şafak Ermertcan, Güner Coşar
Turkish Journal of Medical Sciences
The present study aimed to determine the correlation between the bactericidal activity of vancomycin and quinupristin/dalfopristin (Q/D) on Staphylococcus aureus isolates and their minimal inhibition concentrations. The in-vitro susceptibilities of the 99 S. aureus isolates to vancomycin and Q/D were investigated by agar dilution. Thirty methicillin-resistant S. aureus (MRSA) and 30 methicillin-susceptible S. aureus (MSSA) vancomycin and Q/D susceptible isolates were involved in time-kill studies. While both MRSA and MSSA isolates were susceptible to vancomycin, 96% of both isolates were determined as susceptible to Q/D. In the time-kill test, after 6 h of incubation vancomycin exhibited a bactericidal activity of …

Comparative Cytotoxicity Of Fludarabine In Human Peripheral Blood Mononuclear Cells And In The Promyelocytic Leukemia Cell Line (Hl60), A. Lale Doğan
Turkish Journal of Medical Sciences
Fludarabine is an inhibitor of DNA/RNA synthesis with antitumor activity on lymphoproliferative malignancies. We investigated the dose-dependent cytotoxic effects of the drug on human peripheral blood mononuclear cells, the HL60 cell line and the differentiated HL60 cells. Dimethylformamide was used for the neutrophilic differentiation of HL60 cells and the MTT assay was used to estimate drug cytotoxicity. Undifferentiated tumor cells had higher proliferation rates compared to the differentiated subline. There was 57% cell death in HL60 cells at a dose of 5 µg/ml of the drug. On the other hand, in differentiated HL60 cells and nonmalignant cells, cell death did …

The Effects Of Melatonin On 6-Phosphogluconate Dehydrogenase: An In Vitro And In Vivo Study, İlhami̇ Gülçi̇n, Şükrü Beydemi̇r
Turkish Journal of Medical Sciences
Melatonin is known to influence a variety of biological processes including circadian rhythms, neuroendocrine, and cardiovascular and immune functions as well as thermoregulation. Melatonin is the chief secretory product of the pineal gland, although it is also produced in other organs. In this study, in vitro effects of melatonin on 6-phosphogluconate dehydrogenase from human erythrocytes and in vivo effects of melatonin on 6-phosphogluconate dehydrogenase from rat (Sprague-Dawley) erythrocytes were studied. Human erythrocyte 6-phosphogluconate dehydrogenase was purified in 3 steps, namely haemolysate preparation, ammonium sulphate fractionation (35-65%) and 2´,5´-ADP Sepharose-4B affinity gel chromatography. 6-Phosphogluconate dehydrogenase was purified with a recovery rate …

Typing Of Methicillin-Resistant Staphylococcus Aureus By Ribosome Spacer And Arbitrarily Primed Polymerase Chain Reaction, Tamer Essawi, Kamel Adwan, Nael Abu-Hassan, Galeb Adwan, Ahmed Saleh, Ghaleb Mohammad Adwan
Turkish Journal of Medical Sciences
Thirty-five methicillin-resistant Staphylococcus aureus (MRSA) clinical isolates were collected from 3 hospitals in the northern and southern parts of Palestine between February and May 1998. These isolates were typed by ribosome spacer PCR (RS-PCR) and arbitrarily primed PCR (AP-PCR). RS-PCR generated 9 different genotypes. The use of AP-PCR provided a high resolution typing method and allowed us to define 11 different clusters. Three major clusters, however, based on the combination of both typing methods, spread throughout the neonatal and intensive care units of Rafidya Hospital during the entire period.

Turkish Journal of Medical Sciences
All-trans retinoic acid (ATRA) sensitivity of acute promyelocytic leukaemia (APL) cells is strictly dependent on the presence of t(15;17), but the molecular background of this sensitivity remains obscure. We showed the down- regulation of DEK, AF4 and FLI1 genes in the vitamin D treatment of APL cell line HL-60 , using cDNA array technology in our previous study. This finding prompted us to investigate the expression of these genes in APL patient samples. The effect of ATRA was studied in 6 APL patients carrying t(15;17). Two samples from each patient were compared with a primary diagnostic sample and a sample …

Unsuccessful cadaveric kidney transplantation still remains as an important problem in organ transplantation. Cold storage solutions are not very efficient for kidney preservation. Vitamin E is an important dietary antioxidant, which may play a vital role in preventing free radical perfusion injury. We investigated the effect of vitamin E on kidney preservation with Euro-Collins (EC) solution using the isolated perfused dog kidney model. Recipients were randomly divided into 3 groups: group 1 (n = 6), immediately reperfused with EC solution, given laboratory pelleted diet and drinking water for 4 weeks; and group 2 (n = 6), flushed, and stored with …

Systemic Responses To Burn Injury, Bariş Çakir, Berrak C. Yeğen
Turkish Journal of Medical Sciences
The major causes of death in burn patients include multiple organ failure and infection. It is important for the clinician to understand the pathophysiology of burn injury and the effects it will have on the pharmacokinetics of a drug. The local and systemic inflammatory response to thermal injury is extremely complex, resulting in both local burn tissue damage and deleterious systemic effects on all other organ systems distant from the burn area itself. Thermal injury initiates systemic inflammatory reactions producing burn toxins and oxygen radicals and finally leads to peroxidation. The relationship between the amount of products of oxidative metabolism …
