In metabolic acidosis, the early increase in PEPCK mRNA and enzyme protein content contributes to the accelerated rates of ammonium and glucose formation. In situ hybridization demonstrated that expression of PEPCK was confined to medullary rays of rat kidney cortex in controls and spread throughout the cortex 10 hours following NH4Cl feeding (Am.J. Physiol., 267: F400, 1994). To identify the specific nephron segments expressing PEPCK in control and acidotic conditions, the mRNA for PEPCK along the nephron of the rat kidney was localized using the technique of reverse transcription and polymerase chain reaction (RT-PCR) in individual microdissected S1, S2 and …

The purpose of this study was to determine if terminally ill patients experience a change in perceived quality of life between admission to a hospice program and after having been in the hospice program for three weeks.

A repeated measures design with a convenience sample was used The sample consisted of 56 subjects who completed the Missoula-Vitas Quality of Life Index (MVQOLI). Twenty completed the second MVQOLL Data were obtained from the scores of this self-assessment tool. Data analysis included a comparison of the relationship of the first and repeated scores. The purpose of this study was to examine the effect of social support on an individual's psychological adaptation to an alternate form of nutrition support. Betty Neuman's Systems Model provided the framework for this study. A descriptive correlational design was chosen for this study and data were collected using a questionnaire methodology. Proposed data analyses measures were Pearson's product moment correlation, t- Test, and Mann-Whitney U. The purpose of this study was to examine the change in associate degree nursing (ADN) students’ attitudes toward the elderly following an initial clinical nursing experience in a nursing home. The Neuman Systems Model (Neuman, 1995) was the conceptual focus for the research. A convenience sample of ADN students in a basic nursing care clinical course at a community college was used. The Kogan Attitudes Toward Old People Scale (Kogan, 1961) was utilized to assess student attitudes toward the elderly. One-Year Prospective Prediction Of Violence Perpetration Among High Risk Youth From Personal And Social-Environmental Variables, Alan W. Stacy, Steve Sussman, Thomas R. Simon, Clyde W. Dent, Jill M. Steinberg

Objective: Measures of drug use, law-abidance beliefs, sensation seeking, fear of victimization, high-risk group identification, self- protection needs and behaviors, and demographics were investigated as longitudinal predictors of violence perpetration among 870 high-risk adolescents. Method: Self-reports from the same youth were obtained 1-year apart. Results: In addition to baseline violence perpetration, marijuana use, relatively youn1 age, male sex, high-risk group self-identification, low perceived efficacy of the police department, and nonavoidance of dangerous places predicted later perpetrated violence. Conclusion: Personal and social factors beyond baseline violent behavior predict risk for future violent behavior.

This booklet gives basic information on common parasites, diseases and injuries of fish harvested from fresh water in the Northwest Territories and Nunavut. It answers some of the more common questions asked by Aboriginal, domestic, sport and commercial harvesters, and by fisheries researchers. It is not an in depth treatment of fish health. Parasites that are uncommon or too small to be seen with the naked eye and diseases that are poorly known are not discussed. Relatively little research has been conducted on fish parasites in the region, less still on fish diseases and injuries.

The emerging tick-borne zoonoses human monocytic ehrlichiosis (HME) and human granulocytic ehrlichiosis (HGE) are underreported in the United States. From 1986 through 1997, 1,223 cases (742 HME, 449 HGE, and 32 not ascribed to a specific ehrlichial agent) were reported by state health departments. HME was most commonly reported from southeastern and southcentral states, while HGE was most often reported from northeastern and upper midwestern states. The annual number of reported cases increased sharply, from 69 in 1994 to 364 in 1997, coincident with an increase in the number of states making these conditions notifiable. A series of intensive, longitudinal, mark-recapture studies of hantavirus infection dynamics in reservoir populations in the southwestern United States indicates consistent patterns as well as important differences among sites and host-virus associations. All studies found a higher prevalence of infection in older (particularly male) mice; one study associated wounds with seropositivity. These findings are consistent with horizontal transmission and transmission through fighting between adult male rodents. Despite very low rodent densities at some sites, low-level hantavirus infection continued, perhaps because of persistent infection in a few long-lived rodents or periodic reintroduction of virus from neighboring populations. A PCR assay of 43 acute-phase serum samples was evaluated as a method for early detection of human granulocytic ehrlichiosis (HGE) and determination of etiology when serologic testing is inconclusive. Sequenceconfirmed products of the HGE agent were amplified from three individuals residing or having exposure history in Minnesota or Wisconsin, and similarly confirmed products from Ehrlichia chaffeensis were amplified from three individuals from Florida or Maryland. Etiology, as determined by PCR and serology, was the same whenever there was a fourfold difference between the maximum titers of antibodies to both antigens, indicating that presumptive determination of etiology may be based …

We describe the concordance among results from various laboratory tests using samples derived from nine culture-proven cases of human monocytic ehrlichiosis (HME) caused by Ehrlichia chaffeensis. A class-specific indirect immunofluorescence assay for immunoglobulin M (IgM) and IgG, using E. chaffeensis antigen, identified 44 and 33% of the isolation-confirmed HME patients on the basis of samples obtained at initial clinical presentation, respectively; detection of morulae in blood smears was similarly insensitive (22% positive). An indirect immunofluorescence assay (IFA) was used to identify patients with antibodies reactive to the human granulocytic ehrlichiosis (HGE) agent. Serum samples collected from clinically ill individuals were submitted to the Centers for Disease Control and Prevention by physicians via state health departments from throughout the United States and tested against a panel of ehrlichial and rickettsial pathogens. Antibodies reactive to the HGE agent were detected in 142 (8.9%) of 1,602 individuals tested. A 3-month ecologic investigation was done to identify the reservoir of Ebola virus following the 1995 outbreak in Kikwit, Democratic Republic of the Congo. Efforts focused on the fields where the putative primary case had worked but included other habitats near Kikwit. Samples were collected from 3066 vertebrates and tested for the presence of antibodies to Ebola (subtype Zaire) virus: All tests were negative, and attempts to isolate Ebola virus were unsuccessful. Mule deer fawns (Odocoileus hemionus) were inoculated orally with a brain homogenate prepared from mule deer with naturally occurring chronic wasting disease (CWD), a prion-induced transmissible spongiform encephalopathy. Fawns were necropsied and examined for PrPres, the abnormal prion protein isoform, at 10, 42, 53, 77, 78 and 80 days post-inoculation (p.i.) using an immunohistochemistry assay modified to enhance sensitivity. Conjunctivitis Caused By A Swine Chlamydia Trachomatis-Like Organism In Gnotobiotic Pigs, Douglas G. Rogers, Arthur A. Andersen

The objective of this study was to determine whether a chlamydial strain recovered from growing and finishing swine with conjunctivitis or keratoconjunctivitis could cause the same infections in gnotobiotic pigs. The strain shares biological characteristics with Chlamydia trachomatis. After propagation in Vero cells and preparation of the inoculum (107 inclusion-forming units/ml), chlamydial strain H7 was instilled into the ventral conjunctival sac (0.15 ml/sac) of 12 anesthetized 3-day-old gnotobiotic piglets. Four age-matched gnotobiotic piglets were anesthetized and sham infected with uninfected cell culture lysates. A plasmid bearing a cDNA copy of the genome of bovine viral diarrhea virus (BVDV), chimeric derivatives of the plasmid and a method of producing an infectious bovine viral diarrhea virus using the plasmid are disclosed. The invention relates to a plasmid DNA molecule that replicates easily in E. coli and contains a sufficient portion of the genome of BVDV, cloned as cDNA, to be a suitable template to produce RNA in vitro which, upon transfection into bovine cells, gives rise to infectious BVDV. Many emerging and reemerging bacterial pathogens synthesize toxins that serve as primary virulence factors. We highlight seven bacterial toxins produced by well-established or newly emergent pathogenic microbes. These toxins, which affect eukaryotic cells by a variety of means, include Staphylococcus aureus α-toxin, Shiga toxin, cytotoxic necrotizing factor type 1, Escherichia coli heat-stable toxin, botulinum and tetanus neurotoxins, and S. aureus toxic-shock syndrome toxin. For each, we discuss the information available on its synthesis and structure, mode of action, and contribution to virulence. We also review the role certain toxins have played in unraveling signal pathways in eukaryotic cells and summarize …

Postoperative clinical, histopathological and radiological findings were evaluated for disc fenestration, open discolysis (chy-mopapain) and percutan discolysis which had been performed on three groups of dogs, each group containing 6 healthy mongrels of different ages, sexes and breeds. The degrees of intervertebral space narrowing were classified as mild, medium and severe according to postoperative radiological examinations. Sections which were prepared from intervertebral areas which had been operated on, were investigated histopathologically. This study was conducted to compare performances of some important honeybee (A. mellifera L.) races and ecotypes in Turkey. Experimental colonies were collected from 6 different regions such as Central Anatolia Region (Beypazary), North Eastern Anatolia Region (Posof), Marmara Region (Gökçeada); Thrace Region (Saray), Aegean Region (Fethiye) and Mediterranean Region (Erdemli).
