Evidence For Proximal Control Of Ligand Specificity In Hemeproteins: Absorption And Raman Studies Of Cryogenically Trapped Photoproducts Of Ligand Bound Myoglobins, Mark R. Chance Dec 1991

Evidence For Proximal Control Of Ligand Specificity In Hemeproteins: Absorption And Raman Studies Of Cryogenically Trapped Photoproducts Of Ligand Bound Myoglobins, Mark R. Chance

Faculty Scholarship

The absorption and resonance Raman spectra of cryogenically trapped photoproducts of oxy and carboxy derivatives of myoglobin (Mb) are compared and analyzed in an attempt to understand the structural basis for ligand specificity in hemeproteins. Pulsed and cw excitations over a wide temperature range are used in order to differentiate between kinetic hole burning (KHB), optical pumping of structural relaxation, and spontaneous relaxation effects. Using these techniques, we are able to correlate changes in the absorption spectrum (band III at ≈ 760 nm) with low-frequency Raman bands. Based on these correlations we are able to determine which proximal heme pocket …

Expression And Function Of Zebra: The Epstein-Barr Viral Replication Activator, Naomi Taylor Dec 1991

Expression And Function Of Zebra: The Epstein-Barr Viral Replication Activator, Naomi Taylor

Yale Medicine Thesis Digital Library

Epstein-Barr virus (EBV) encodes a protein, ZEBRA, which enables the virus to switch from a latent to a lytic life cycle. ZEBRA expression was studied in lymphoid cells harboring either standard virus or a mixture of standard and defective viruses. ZEBRA protein could not be detected in cells latently infected with standard EBV but was constitutively expressed in cells containing both defective and standard EBV genomes. Experiments indicated that regulation of transcription of the ZEBRA gene (BZLF1) is a pivotal event in the control of EBV replication. ZEBRA specific BZLF1 transcripts were spontaneously expressed in cells harboring defective virus but …

Preliminary Epidemiological Interpretation Of The Intrinsic And Extrinsic Factors Contributing To The Coverage And Participation Rates Of The Vaccination Program In Arssi, Ethiopia, Muchie Kidanu Dec 1991

Preliminary Epidemiological Interpretation Of The Intrinsic And Extrinsic Factors Contributing To The Coverage And Participation Rates Of The Vaccination Program In Arssi, Ethiopia, Muchie Kidanu

Electronic Theses and Dissertations

Many immunization programs in developing countries arr now undergoing a period of rapid utilization. Innovative strategies, such as immunization holidays, are being tried. Political commitment and enthusiasm are high. There is a strong desire by health professionals to protect the lives of many children from the untimely death and/or disability resulting from infectious diseases preventable by vaccination.

Evaluation of an immunization program can benefit other elements of the health system, particularly those that are not amenable to objective evaluation as immunization. In this case, immunization can serve as an indicator of the overall performance of the health system. The problems …

The Effects Of Two Types Of Hypnotic Suggestions On Analgesic Responding In Moderately Hypnotizable Subjects, Gloria M. Haddad Dec 1991

The Effects Of Two Types Of Hypnotic Suggestions On Analgesic Responding In Moderately Hypnotizable Subjects, Gloria M. Haddad

Masters Theses

This study assessed the effects of two hypnotic suggestions on hypnotic performance in medium hypnotizable subjects. Subjects were exposed at separate intervals to either an hypnotic suggestion patter containing specific imagery or alternatively to a suggestion patter that provided only general problem-solving direction. Hypnotic performance was assessed using tolerance and threshold measures on a cold-pressor test.

The problem-solving suggestions were shown to increase both threshhold and tolerance of noxious stimulation. The imagery suggestions proved to be effective only for the threshhold measure.

These findings are discussed in terms of their implications for the use of hypnosis with subjects who do …

Comparison Of A Quantitative Microtiter Method, A Quantitative Automated Method, And The Plate-Count Method For Determining Microbial Complement Resistance, Margie D. Lee, Richard E. Wooley, John Brown, Kathy R. Spears, Lisa K. Nolan, Emmett B. Shotts Jr.

Lisa K. Nolan

A quantitative microtiter method for determining the degree of complement resistance or sensitivity of microorganisms is described. The microtiter method is compared with a quantitative automated system and the standard plate-count technique. Data were accumulated from 30 avian Escherichia coli isolates incubated at 35 C with either chicken plasma or heat-inactivated chicken plasma. Analysis of data generated by the automated system and plate-count techniques resulted in a classification of the microorganisms into three groups: those sensitive to the action of complement; those of intermediate sensitivity to the action of complement; and those resistant to the action of complement. Although the …

Lisa K. Nolan

Y. A quantitative microtiter method using chicken plasma is described for determining the degree of complement resistance or sensitivity of avian Escherichia coli isolates. Results obtained with the microtiter method using chicken plasma were compared with results obtained using commercially available standardized guinea pig serum as the source of complement. The test organisms consisted of five isolates of E. coli isolated from chickens. Three isolates were from flocks with colisepticemia; one was from a flock with omphalitis; and one isolate was a non-pathogenic control. Data were accumulated from the five avian E. coli isolates incubated at 35 C with either …

The Landmark Citation Method: Analysis Of A Citation Pattern As A Collection Assessment Method, Catherine B. Soehner, S Tanner Wray, Daniel T. Richards

Dartmouth Scholarship

The landmark citation method is a new collection assessment method based on the citation record of a single landmark article. This citation record is developed by identifying sources which cite the landmark article. A bibliography, extracted from the citation record, is then used to complete an assessment of the collection. This method was developed and used to assess the biotechnology collection of the National Library of Medicine. The information gained from this study, in addition to demonstrating the technique, also provided insight into the evolution of the biotechnology literature.

The Ability Of Simian Virus 40 Large T Antigen To Immortalize Primary Mouse Embryo Fibroblasts Cosegregates With Its Ability To Bind To P53., Jiyue Y. Zhu, Marina Abate, Philip W. Rice, Charles N. Cole Dec 1991

The Ability Of Simian Virus 40 Large T Antigen To Immortalize Primary Mouse Embryo Fibroblasts Cosegregates With Its Ability To Bind To P53., Jiyue Y. Zhu, Marina Abate, Philip W. Rice, Charles N. Cole

Dartmouth Scholarship

The large T antigen encoded by simian virus 40 (SV40) plays essential roles in the infection of permissive cells, leading to production of progeny virions, and in the infection of nonpermissive cells, leading to malignant transformation. Primary mouse embryo fibroblasts (MEFs) are nonpermissive for SV40, and infection by wild-type SV40 leads to immortalization and transformation of a small percentage of infected cells. We examined the ability of an extensive set of mutants whose lesions affect SV40 large T antigen to immortalize MEFs. We found that immortalization activity was retained by all mutants whose lesions are located upstream of codon 346. …

A Novel Transposon Trap For Mycobacteria: Isolation And Characterization Of Is1096, Jeffrey D. Cirillo, Raul G. Barletta, Barry R. Bloom, William R. Jacobs Jr.

School of Veterinary and Biomedical Sciences: Faculty Publications

In the course of developing strategies to obtain a mutation in the aspartate semialdehyde dehydrogenase (asd) gene of Mycobacterium smegmatis, an efficient transposon trap was constructed which may be generally useful for the identification of transposable elements in mycobacteria. A DNA fragment containing the asd gene was replaced with an aminoglycoside phosphotransferase gene (aph) to generate a Δasd::aph allele. Attempts to replace the wild-type asd gene with the Δasd::aph allele were unsuccessful, suggesting that this deletion was lethal to the growth of M. smegmatis. The plasmid, pYUB215, which …
