Measured as elapsed time from first use to dependence syndrome onset, the estimated 'induction interval' for cocaine clearly is short relative to the cannabis interval, but little is known about risk of becoming dependent when use persists. Published estimates for this facet of drug dependence epidemiology are from life histories elicited years after first use. To improve estimation, we turn to new data from nationally representative samples of newly incident drug users identified via probability sampling and confidential computer-assisted self-interviews for the National Surveys on Drug Use and Health, 2004-2013. “Cardinal rules” and best practice approaches have guided governmental risk communication efforts at chronic risk sites for more than two decades, playing an important role in how those most affected by contamination make sense of risk. In addition to providing information, however, communication approaches themselves can affect community perceptions indirectly, through stakeholder interpretations of the processes by which risk information is shared. It is increasingly necessary to evaluate not only whether risk communication approaches have been effective for increasing knowledge but if, in fact, the ways in which information is shared has had unintended consequences that change how stakeholders perceive …

Roughly 25 years ago, the United States National Institute on Drug Abuse (US NIDA) initiated the creation of public use datasets for its National Household Survey on Drug Abuse, since re-named the National Survey on Drug Use and Health (NSDUH). The Substance Abuse and Mental Health Services Administration (SAMHSA), which assumed responsibility for the survey in 1992, has continued and expanded this effort to make the survey data available to researchers. During 2012, SAMHSA created a “Restricted-Use Data Analysis System” (R-DAS) to provide researchers with the capability to create tabulations using restricted NSDUH variables not otherwise available on the …

One of the hallmarks of a drug dependence (DD) process is an escalation in rate of drug self-administration (DSA). We seek to extend current biostatistical approaches for epidemiological research on drug dependence processes via an investigation of a four-parameter dose-effect curve (DEC).

Healthcare and public health research ultimately seek to improve patient and population health. Unfortunately, more than a decade often passes before research findings become routinized in practice. Improving translational speed, reach, and efficacy requires partnerships among researchers, practitioners, community stakeholders, and communication scholars. The specific methodological aims of our research are: 1) to clarify statistical inference procedures used by R-DAS to produce confidence interval estimates; 2) to review a simple solution to some of the research questions not currently addressed by R-DAS (i.e., with respect to tests of significance); and 3) to describe how in certain situations the percentages and the corresponding standard errors, which are suppressed from the R-DAS output due to the confidentiality concerns can be estimated “by hand”.

The persistence of pain after surgery increases the recovery interval from surgery to a normal quality of life. AYX1 is a DNA-decoy drug candidate designed to prevent post-surgical pain following a single intrathecal injection. Tissue injury causes a transient activation of the transcription factor EGR1 in the dorsal root ganglia–dorsal horn network, which then triggers changes in gene expression that induce neuronal hypersensitivity. AYX1 is a potent, specific inhibitor of EGR1 activity that mimics the genomic EGR1- binding sequence. Administered in the peri-operative period, AYX1 dose dependently prevents mechanical hypersensitivity in models of acute incisional (plantar), inflammatory (CFA), and chronic …

This report updates surveillance results for COPD in the United States. For 1999 to 2011, data from national data systems for adults aged ≥ 25 years were analyzed. In 2011, 6.5% of adults (approximately 13.7 million) reported having been diagnosed with COPD. From 1999 to 2011, the overall age-adjusted prevalence of having been diagnosed with COPD declined (P = .019). In 2010, there were 10.3 million (494.8 per 10,000) physician office visits, 1.5 million (72.0 per 10,000) ED visits, and 699,000 (32.2 per 10,000) hospital discharges for COPD. From 1999 to 2010, no significant overall trends were noted for physician …

Opioid receptor antagonists increase hyperalgesia in humans and animals, which indicates that endogenous activation of opioid receptors provides relief from acute pain; however, the mechanisms of long-term opioid inhibition of pathological pain have remained elusive. We found that tissue injury produced m-opioid receptor (MOR) constitutive activity (MORCA) that repressed spinal nociceptive signaling for months. Pharmacological blockade during the posthyperalgesia state with MOR inverse agonists reinstated central pain sensitization and precipitated hallmarks of opioid withdrawal (including adenosine 3′,5′-monophosphate overshoot and hyperalgesia) that required N-methyl-D-aspartate receptor activation of adenylyl cyclase type 1.
