Recent studies provide strong evidence that the androgen receptor (AR) signaling pathway remains active in castration-resistant prostate cancer (CRPC). However, the underlying mechanisms are not well understood. In this study, we demonstrate that plant homeo domain finger protein 8 (PHF8 )interacts with and functions as an essential histone demethylase activity-dependent AR coactivator. Furthermore, we demonstrate that the expression of PHF8 is induced by hypoxia in various prostate cancer cell lines. Knockdown of either hypoxia-inducible factor HIF2α or HIF1α almost completely abolished hypoxia-induced PHF8 expression. BACKGROUND: Learning to perform pelvic and breast examinations produces anxiety for many medical students. Clerkship directors have long sought strategies to help students become comfortable with the sensitive nature of these examinations. Incorporating standardized patients, simulation and gynecologic teaching associates (GTAs) are approaches gaining widespread use. Obesity and its associated metabolic syndrome continue to be a health epidemic in westernized societies and is catching up in the developing world. Despite such increases, little headway has been made to reverse adverse weight gain in the global population. Few medical options exist for the treatment of obesity which points to the necessity for exploration of anti-obesity therapies including pharmaceutical and nutraceutical compounds. OBJECTIVE: The objective of this selective EBM review is to determine whether or not “Is Parathyroid Hormone (PTH) replacement therapy an effective treatment for patients with hypoparathyroidism?”

STUDY DESIGN: Review of a randomized control trial published in 2011, a prospective cohort study published in 2013, and a randomized crossover study published in 2012.

BACKGROUND: Cardiopulmonary resuscitation (CPR) training has traditionally involved classroom-based courses or, more recently, home-based video self-instruction. These methods typically require preparation and purchase fee; which can dissuade many potential bystanders from receiving training. This study aimed to evaluate the effectiveness of teaching compression-only CPR to previously untrained individuals using our 6-min online CPR training video and skills practice on a homemade mannequin, reproduced by viewers with commonly available items (towel, toilet paper roll, t-shirt).

Isoforms of the Fcγ receptor III (FcγRIII or CD16) are cell surface receptors for the Fc portion of IgG and important regulators of humoral immune responses. Different ligand binding kinetics of FcγRIII isoforms are obtained in three dimensions by surface plasmon resonance and in two dimensions by a micropipette adhesion frequency assay. We show that the anchor structure of CD16 isoforms isolated from the cell membrane affects their binding affinities in a ligand-specific manner. Changing the receptor anchor structure from full to partial to none decreases the ligand binding affinity for human IgG1 (hIgG1) but increases it for murine IgG2a …

Prostate cancer is one of the biggest threats to men’s health in the western world and it accounts for the second largest number of male cancer-related deaths in the United States. It is well established that prostate cancer cells depend on the androgen/androgen receptor pathway. Therefore, androgen deprivation therapy (ADT) has become the primary treatment option for prostate cancer. Patients with metastatic prostate cancer who receive (ADT) have shown increased quality of life. Mild Traumatic Brain Injury is a neurologic insult that impacts a child’s functioning across domains. Risk factors include previous concussion history, force of impact, and gender. Current research regarding return to school following a brain injury suggests the need for accommodations in a number of areas of learning including changes in schedule, reduced exposure to screens and text, limitations to classroom expectations for tasks such as reading and note-taking, and modified testing. A one-size-fits-all approach to accommodations is problematic as recovery rates vary widely. Studies have also suggested that mood symptoms are comorbid with concussion recovery. Mitochondria may be a principle source of oxidative stress causing MI/R injury. Coenzyme Q10 (CoQ10) is essential for electron transport in normal mitochondria, has antioxidant properties but its bioavailability is likely reduced due to oxidative stress during MI/R. Coenzyme Q1 (CoQ1) is a derivative of CoQ10, but is a more potent antioxidant than CoQ10 due to a shorter isoprene chain. We hypothesize that CoQ1 will exhibit better cardioprotective effects during MI/R. CoQ1 (MW=250 g/mol; 20 μM, n=5) and CoQ10 (MW=863 g/mol; 20 μM, n=5) were given at reperfusion in isolated rat hearts subjected to I (30 min)/R (45 min). We …

It’s known that PMA is a broad-spectrum PKC activator that augments SO release in PMNs viaNADPH oxidase activation. It was shown that PKC delta (δ) negatively regulates PMN elastase release which suggests that PKC δ may also negatively regulate PMN NADPH oxidase SO release. To test the hypothesis, we altered PKC δ activity using selective cell permeable PKC δ activator, inhibitor peptides, and rottlerin, another type of PKC δ inhibitor. We predict that PKC δ activation would attenuate PMA-induced PMN SO release, whereas PKC δ inhibition would augment the response. Osteopathic Manipulative Treatment (OMT) is a category of several types of manual techniques used by osteopathic physicians. According to the osteopathic tradition, structure and function are intimately related. This project examined the relationship between shoulder structure and function in an osteopathic framework, with a specific goal to understand how the structure of the acromion affects the development of shoulder injuries. For the first part of this project, cadaveric dissections were performed and muscle diameters were taken of structures within the subacromial space. Suicide is a leading cause of death for adolescents and represents a major national health concern (American Foundation for Suicide Prevention, [AFSP], 2015; (Centers for Disease Control [CDC], 2013). Hyperglycemia has been associated with vascular endothelial dysfunction in part by a reduction in nitric oxide (NO) production and increased oxidative stress (e.g., increased superoxide (SO) and hydrogen peroxide (H2O2). Endothelial-derived NO can be significantly reduced by increased SO/H2O2 in part by the activation of NADPH oxidase during hyperglycemia. Of the 7 NADPH oxidase isoforms, NADPH oxidase isoform 1 (NOX1) is mainly expressed in the vasculature and may play a major role in hyperglycemia induced oxidative stress and vascular endothelial dysfunction. Alzheimer's' Disease (AD) is a progressive neurodegenerative disease that affects millions of elderly individuals in the western world. There are two forms of AD, sporadic late-onset AD and Familial AD. The neuropathy seen in both cases of AD is the same; neurofibrillary tangles (NFTs) that are composed of modified tau protein, and neuritic senile plaques (NSPs) composed of deposited beta amyloid peptide. Matrix metalloproteinases (MMPs) are a family of zinc-dependent endopeptidases with the unique ability to breakdown virtually the entire extracellular matrix (ECM). Through ECM remodeling, MMPs play an important role in normal development, tissue repair, angiogenesis, and apoptosis. Studies have shown that unregulated MMP expression plays a role in many cancers and chronic inflammatory diseases. Previous research has used gene sequence analysis ofthe MMP-1 promoter to identify a putative ZBP-89 binding site at -1969 bp. Prostate cancer, or prostatic carcinoma (PrCa), is the second leading cause of cancer-related deaths in men in the Western world. One in seven men are expected to be diagnosed with the disease in the United States. Suppressing androgen/androgen receptor pathway is a first-line therapeutic treatment known as Androgen Deprivation Therapy (ADT). Most patients respond well to ADT, but an overwhelming majority of PrCa progress to an advanced stage of PrCa independent of hormone, known as Castration Resistant Prostate Cancer (CRPC). OBJECTIVE: The objective of this selective EBM was to investigate the question, “Does routine use of medical cannabis (marijuana) decrease the intensity of neuropathic chronic pain in adults?

STUDY DESIGN: systematic review of 3 English language primary studies, published between 2005 and 2009.

DATA SOURCES: Three Randomized Controlled Trials (RTC’s) published on or after 2005 were selected based on their relevance to the proposed question via PubMed. All three RTC’s compared cannabis use vs. placebo for chronic neuropathic pain, with two RTC’s focusing on HIV neuropathic pain and the third focusing on MS.

OBJECTIVE: The objective of this selective EBM review is to determine whether or not viscosupplementation is effective in reducing osteoarthritis knee pain

STUDY DESIGN: Review of three randomized, double blind, placebo control trials between 2009- 2010

DATA SOURCES: Three randomized, double blind, placebo control trials were found via PubMed and NCBI

OUTCOMES MEASURED: Each randomized control trial measured knee pain and/or patient satisfaction following their intervention (placebo or hyaluronic acid) in patients with knee osteoarthritis, which was measured via WOMAC pain scale and patient global assessment forms.

RESULTS: (P<0.05) for change in pain at 14-26 weeks after injection (NNT=6- 8 for Chevalier et al and 4 for Kul-Panza & Berker). Patient satisfaction was higher in the experimental groups as compared to the control with conclusions of NNT between 11 and 20 for Chevalier et al and Kul-Panza & Berker respectively. Chavelier et al found adverse events such as arthralgias and joint effusion in <10% of their study population (NNH=39). No dichotomous data was presented in Diacoglu et al; however an ANOVA was deemed statistically significant (p<0.01).

CONCLUSION: Review of these articles concludes that the use of hyaluronic …
