Metachromatic leukodystrophy (MLD) is an inherited lysosomal storage disease caused by deficient arylsulfatase A (ARSA) activity, resulting in intra-lysosomal accumulation of sulfatide. Sulfatide is integral for proper maintenance of myelin in the central and peripheral nervous systems. This study characterized mitochondrial morphology, cytokine secretion and phagocytic activity in Schwann and mesenchymal cells isolated from ARSA-/- mice. Cells were treated with different, increasing concentrations of sulfatide for a period of 24 hours. ARSA-/- cells presented with persistent, increased fragmented mitochondrial structures suggestive of prolonged mitochondrial fission. Pathology reporting on radical prostatectomy (RP) specimens is essential to post-surgery patient care. However, current pathology interpretation of RP sections is typically qualitative and subject to intra- and inter-observer variability, which challenges quantitative and repeatable reporting of lesion grade, size, location, and spread. Therefore, we developed and validated a software platform that can automatically detect and grade cancerous regions on whole slide images (WSIs) of whole-mount RP sections to support quantitative and visual reporting. The delivery of antibiotics is an important component of therapy for orthopedic device-related infections (ODRI). In this study, we have investigated new techniques to characterize and enhance antibiotic delivery for ODRI. Characterization of small-molecule diffusion is essential to the development of drug-delivery systems. We have developed a quantitative, non-invasive, longitudinal, micro-CT technique to quantify the diffusion of small-molecules in an intact phantom. We employed a radio-opaque molecule (i.e., Iohexol) as a surrogate for commonly used antibiotics (e.g., Vancomycin). Transplantation is invariably associated with ischemia reperfusion injury (IRI) which causes organ dysfunction. IRI is also directly linked to several forms of programmed cell death including apoptosis and necroptosis, which increase kidney dysfunction, promote inflammation and may contribute to premature graft failure. The contribution of necroptosis and apoptosis following kidney IRI to cell-free DNA (cfDNA) generation and the potential of cfDNA to activate effectors such as NK cells involved in kidney IRI have not been defined.
