Background/aim: It is believed that radiotherapy has important effects on oxidant/antioxidant systems. Oxidative stress occurs when the balance between oxidant formation and antioxidant defense is disrupted in favor of oxidants. The aim of this study was to determine the biochemical changes in saliva pre- and postradiotherapy in head-neck radiotherapy patients and to find out the effects of radiation on glutathione (GSH), glutathione peroxidase (GSH-Px), and malondialdehyde (MDA) levels in saliva. Materials and methods: This study included 16 patients undergoing head-neck radiotherapy in Atatürk University Research Hospital. The levels of GSH, GSH-Px, and MDA were measured in saliva samples taken from …

Asymmetric dimethylarginine (ADMA) is an inhibitor of nitric oxide synthase (NOS). Oxidative stress might be defined as an imbalance between protein oxidation and antioxidants. Our aim was to determine in vivo whether ADMA causes oxidative damage. Materials and methods: Thirty rats were divided into 3 equal groups: a control group, a group administered 1 mg/kg ADMA, and a group administered 2 mg/kg ADMA. ADMA was administered intraperitoneally for 7 days. Malondialdehyde (MDA), protein carbonyl (PC) content, and nitrate+nitrite concentrations were measured with serum samples. Superoxide dismutase (SOD), catalase, and glutathione peroxidase (GSH-Px) activities were analyzed with plasma samples. Results: A …

The Effect Of Alkali Burn On Corneal Glutathione Peroxidase Activities In Rabbits, İlknur Akyol Salman, Ahmet Kiziltunç, Orhan Baykal

Turkish Journal of Medical Sciences

Glutathione peroxidase (GPx) is an important corneal defense enzyme for protecting against hydrogen peroxides and lipid peroxides. In the present study, corneal GPx activities after alkali burn of the cornea were evaluated in rabbits. Materials and methods: Corneal alkali burn was created in the right eye of 10 rabbits by applying a 6 mm round filter paper soaked in 1 N NaOH onto the central cornea for 60 s. The right eyes of 10 healthy rabbits served as controls. The corneal GPx activities were determined by the method of Paglia and Valentina, 10 days after the burn. Results: GPx activities …

The Effects Of Sodium Selenite On The Antioxidative Defence Mechanism Of Human Hepatoma G_2 Cells, Kayahan Fişkin Jan 2000

The Effects Of Sodium Selenite On The Antioxidative Defence Mechanism Of Human Hepatoma G_2 Cells, Kayahan Fişkin

Turkish Journal of Medical Sciences

Human cells that live in biological harmony can become cancerous. As they do so, they engage in anaerobic respiration, undergo rapid cell growth and division according to nonmitotic rules, and important changes take place in their metabolism. We conducted several investigations in an effort to block or change the metabolic characteristics of cancer cells using a typical cancerous cell line. Specifically,we investigated the effects of sodium selenite (Na_2 SeO_ 3 ) on antioxidant enzyme activities and reduced glutathione levels in Hepatoma G_ 2 cells, cells that show rapid glutamine uptake and metabolism. A Hepatoma G_ 2 cell line was cultured …

The In Vitro Effects Of The Antimalarial Drug Primaquine, On The Activities Of Some Enzymes In Human Erythrocyte Lyzates, Meltem Müftüoğlu, Nazmi̇ Özer

Turkish Journal of Medical Sciences

The effect of primaquine on the enzymatic antioxidant defence mechanisms of erythrocytes were determined by measuring the activities of the below enzymes, before and after 30 minutes of incubation of hemolyzates with 1 mM primaquine at 37°C. Among the enzymes studied glucose-6- phosphate dehydrogenase seemed the most sensitive enzyme; 57 % and 78 % inhibition at zero (due to the binding of primaquine to NADP binding site) and 30 minutes of incubation (due to the reactive oxygen species), respectively. Catalase, superoxide dismutase, and glutathione-S-transferase were less sensitive to primaquine-derived reactive oxygen species: 33, 11, and 0 % inhibition at the …

For most infectious diseases, increased formation of reactive oxygen species is secondary to the primary disease process. Some microorganisms are highly susceptible to exogenous reactive oxygen species such as hydrogen peroxide. Glutathione peroxidase (GSH-Px) remove H 2 O 2 , and selenium is requird for the activity of GSH-Px. Therefore, to assess the status of GSH-Px and selenium in cutaneous leishmaniasis (CL), we determined serum selenium concentration and blood GSH-Px activity in 52 patients with CL and in 38 healthy controls. Selenium concentrations and GSH-Px activities were found to be lower in the patient group than in the controls (p
