In Situ Arsenic Speciation Using Surface-Enhanced Raman Spectroscopy And The Coffee Ring Effect, Valery Liamtsau
In Situ Arsenic Speciation Using Surface-Enhanced Raman Spectroscopy And The Coffee Ring Effect, Valery Liamtsau
FIU Electronic Theses and Dissertations
The coffee ring effect (CRE) phenomenon is originated from the nonuniform solvent evaporation of a sessile droplet deposited onto the flat substrate. Once the evaporation starts, the droplet suspended particles move outwards by the radial flow and concentrate in the edge region of the evaporating droplet, resulting in the formation of the so-called CRE stains. In this work we have expanded the applications of the CRE from separation of particles and macromolecules to small molecules, in particular, coupled to surface-enhanced Raman spectroscopy (SERS). Herein, we have developed a theoretical framework to describe the CRE-driven separation process of small molecules, using …
The Effects Of Copper Exposure On Fish Locomotion And Predator-Prey Interactions, Tiffany N. Yanez
The Effects Of Copper Exposure On Fish Locomotion And Predator-Prey Interactions, Tiffany N. Yanez
FIU Electronic Theses and Dissertations
This study determined the effects of the copper water quality criterion (WQC) by the EPA on [1] swimming performance and [2] predator-prey interactions of the Sailfin Molly, across a salinity gradient. Fish acclimated to FW (0 ppt) and 8-ppt saltwater were exposed to 11.3 and 8.44 µg/L Cu for 96 h, respectively. At the end of the exposures, fish swimming performance was determined by using the critical swimming speed, Ucrit, the speed at which a fish cannot longer maintain position in the water column. Ucritwas then measured again after a 4-week depuration period to determine if …
Aptamer-Based Assay For Detection Of Ochratoxin A, Amanda Nicole Bartley
Aptamer-Based Assay For Detection Of Ochratoxin A, Amanda Nicole Bartley
FIU Electronic Theses and Dissertations
Ochratoxin A (OTA) is a potent mycotoxin found in a wide range of agricultural products that has been linked to mitochondrial damage and renal disease. The standard methods for OTA analysis currently rely on the use of high-performance liquid chromatography (HPLC) coupled to fluorescence detection or mass spectrometry. Toward a highthroughput analysis of OTA, a single-stranded DNA aptamer, modified with a fluorophore, coupled to a complementary sequence, modified with a FRET-based quencher that dissociates in the presence of the target toxin, is proposed. In order to integrate “target trapping,” aptamer immobilization methods were explored to mediate interference issues. Assays were …
The Cyclodextrin-Perfluorinated Surfactant Host-Guest Complex: Fundamental Studies For Potential Environmental Remediation And Therapeutic Applications, Mary J. Errico
FIU Electronic Theses and Dissertations
Perfluoroalkyl substances (PFASs) are contaminants of emerging concern, and have been detected in drinking water, wildlife, humans, and the environment. Cyclodextrins (CDs), cyclic sugars composed of glucose monomers, are proposed as a potential remediation strategy. CDs can form host-guest complexes with hydrophobic molecules; this complexation could be capitalized on for PFAS removal and sequestration. These dissertation projects aim to study the fundamental host-guest interactions between a variety of PFASs and CDs for eventual applications in environmental and biological remediation. 1D and 2D Nuclear magnetic resonance (NMR) spectroscopic methods were employed to determine the strength, dynamics, and structure of the CD:PFAS …
Development And Application Of Aquatic Toxicology Studies For The Assessment Of Impacts Due To Chemical Stressors Using Non-Standard Indigenous Organisms, Abraham Jeffrey Smith
Development And Application Of Aquatic Toxicology Studies For The Assessment Of Impacts Due To Chemical Stressors Using Non-Standard Indigenous Organisms, Abraham Jeffrey Smith
FIU Electronic Theses and Dissertations
Research in the multidisciplinary science of ecotoxicology is crucial to assess injuries to ecosystem resources from chemical spills or other stressors used to support environmental decision-making. Established guidelines recommend the use of non-standard native species in toxicity investigations. This work focused on the use of native species for aquatic toxicity assessment to make more relevant conclusions on the potential for adverse biological effects to occur as a result to single chemical exposures or exposures to a complex mixture like oil. We apply these studies to investigate petroleum product impacts from the Deepwater Horizon incident and concerns for metal toxicity in …
The Effect Of Target-Specific Biomolecules In Breast Cancer, Mohannad Garoub
The Effect Of Target-Specific Biomolecules In Breast Cancer, Mohannad Garoub
FIU Electronic Theses and Dissertations
Cancer is the second leading cause of mortality in the United States and the World, therefore, early effective prevention, diagnosis, and therapy is needed. Estrogens play a major role in the initiation and progression of breast cancer. Elevated lifetime exposure to estrogens is associated with an increased risk of developing breast cancer. Estrogens through influencing mitochondria contribute to estrogen induced breast carcinogenesis; however, the exact mitochondrial mechanisms underlying the estrogen carcinogenic effect in breast tissue are not clearly understood. For this dissertation, the mitotoxic and cytotoxic effects of triphenylphosphonium cation (TPP) and Origanum majorana organic extract (OME) as well as …
Label-Free Surface-Enhanced Raman Spectroscopy-Linked Immunosensor Assay (Slisa) For Environmental Surveillance, Vinay Bhardwaj
Label-Free Surface-Enhanced Raman Spectroscopy-Linked Immunosensor Assay (Slisa) For Environmental Surveillance, Vinay Bhardwaj
FIU Electronic Theses and Dissertations
The contamination of the environment, accidental or intentional, in particular with chemical toxins such as industrial chemicals and chemical warfare agents has increased public fear. There is a critical requirement for the continuous detection of toxins present at very low levels in the environment. Indeed, some ultra-sensitive analytical techniques already exist, for example chromatography and mass spectroscopy, which are approved by the US Environmental Protection Agency for the detection of toxins. However, these techniques are limited to the detection of known toxins. Cellular expression of genomic and proteomic biomarkers in response to toxins allows monitoring of known as well as …
Mangiferin As A Biomarker For Mango Anthracnose Resistance, Herma Pierre
Mangiferin As A Biomarker For Mango Anthracnose Resistance, Herma Pierre
FIU Electronic Theses and Dissertations
Mangos (Mangifera indica L.) are tropical/subtropical fruits belonging to the plant family Anacardiaceae. Anthracnose is the most deleterious disease of mango both in the field and during postharvest handling. It is most commonly caused by the Colletotrichum gloeosporioides complex. Mangiferin, a xanthanoid compound found in at least twelve plant families worldwide (Luo et al., 2012), is present in large amounts of the leaves and edible mangos. Even though this compound plays a pivotal role in the plant’s defense against biotic and abiotic stressors, no correlations been made between the compound and mango anthracnose resistance.
Mangos were collected, grouped …
Mechanisms Of Arsenic Detoxification And Resistance, Jitesh Kannan Pillai
Mechanisms Of Arsenic Detoxification And Resistance, Jitesh Kannan Pillai
FIU Electronic Theses and Dissertations
Arsenic is a ubiquitous environmental toxic substance. As a consequence of continual exposure to arsenic, nearly every organism, from Escherichia coli to humans have evolved arsenic detoxification pathways. One of the pathways is extrusion of arsenic from inside the cells, thereby conferring resistance. The R773 arsRDABC operon in E. coli encodes an ArsAB efflux pump that confers resistance to arsenite. ArsA is the catalytic subunit of the pump, while ArsB forms the oxyanion conducting pathway. ArsD is an arsenite metallochaperone that binds arsenite and transfers it to ArsA. The interaction of ArsA and ArsD allows for resistance to As(III) at …
Speciation, Metabolism, Toxicity, And Protein-Binding Of Different Arsenic Species In Human Cells, Szabina A. Stice
Speciation, Metabolism, Toxicity, And Protein-Binding Of Different Arsenic Species In Human Cells, Szabina A. Stice
FIU Electronic Theses and Dissertations
Despite of its known toxicity and potential to cause cancer, arsenic has been proven to be a very important tool for the treatment of various refractory neoplasms. One of the promising arsenic-containing chemotherapeutic agents in clinical trials is Darinaparsin (dimethylarsinous glutathione, DMAIII(GS)). In order to understand its toxicity and therapeutic efficacy, the metabolism of Darinaparsin in human cancer cells was evaluated. With the aim of detecting all potential intermediates and final products of the biotransformation of Darinaparsin and other arsenicals, an analytical method employing high performance liquid chromatography inductively coupled mass spectrometry (HPLC-ICP-MS) was developed. This method was …
Comprehensive Forensic Toxicological Analysis Of Designer Drugs, Madeleine Jean Swortwood
Comprehensive Forensic Toxicological Analysis Of Designer Drugs, Madeleine Jean Swortwood
FIU Electronic Theses and Dissertations
New designer drugs are constantly emerging onto the illicit drug market and it is often difficult to validate and maintain comprehensive analytical methods for accurate detection of these compounds. Generally, toxicology laboratories utilize a screening method, such as immunoassay, for the presumptive identification of drugs of abuse. When a positive result occurs, confirmatory methods, such as gas chromatography (GC) or liquid chromatography (LC) coupled with mass spectrometry (MS), are required for more sensitive and specific analyses. In recent years, the need to study the activities of these compounds in screening assays as well as to develop confirmatory techniques to detect …
