Animal Sentience
Klein & Barron's review of recent insect neurobiology helps correct the impression that insect behavior is orchestrated without the benefit of central integrative mechanisms. Given their existence, the authors go on to ask whether these central mechanisms also feature the kind of integrative operations that support sentience, and propose that they do. Along the way they raise a number of conceptual and evidentiary issues of fundamental importance for the neuroscience of consciousness, allowing me to comment favorably on a number of them. Prenatal cocaine exposure causes profound changes in neurobehavior as well as synaptic function and structure with compromised glutamatergic transmission. Since synaptic health and glutamatergic activity are tightly regulated by brain-derived neurotrophic factor (BDNF) signaling through its cognate tyrosine receptor kinase B (TrkB), we hypothesized that prenatal cocaine exposure alters BDNF-TrkB signaling during brain development. Here we show prenatal cocaine exposure enhances BDNF-TrkB signaling in hippocampus and prefrontal cortex (PFCX) of 21-day-old rats without affecting the expression levels of TrkB, P75NTR, signaling molecules, NMDA receptor—NR1 subunit as well as proBDNF and BDNF. Spatial Memory In Black-Capped Chickadees: Studies Of Adult Hippocampal Neurogenesis And Win-Shift/Win-Stay Spatial Search, Nicole Ann Guitar

Two cognitive adaptations were studied in Black-capped chickadees through tests of adult hippocampal neurogenesis and Win-shift/Win-stay spatial search. Neurogenesis has been proposed to aid memory, therefore it was hypothesized that birds with decreased neurogenesis would perform poorer than controls in hippocampal-dependent spatial working and reference memory tasks followed by a reversal. Subjects with decreased neurogenesis, caused by the neurotoxin MAM, reversed slower than controls, suggesting that neurogenesis may contribute to differentiating similar memories, although this effect was nonsignificant. Win-shift/Win-stay foraging behavior is an adaptation to the replenishing and depleting nature of food. Since chickadees forage on food that depletes quickly …

The locus coeruleus (LC) is a small noradrenergic nucleus located in the midbrain that releases the neurotransmitter norepinephrine to diverse brain regions. Through release of norepinephrine, the LC plays a central role in modulating numerous physiological functions including attention, arousal, and mood and behavior. Although the LC projects to many brain region, there is limited information about the organization and the afferent projections to the LC that modulates its activity. Fragile X syndrome (FXS) is the leading genetic cause of autism. Individuals with Fragile X Syndrome (FXS) commonly display social, behavioral, and intellectual disabilities. Perceptual deficits and their underlying neural activity remain poorly characterized in FXS and other autism spectrum disorders (ASD’s). To explore visual perception in autism, we developed camera based pupil tracking software using OpenCV (an open-source computer vision library) capable of measuring visually evoked changes in pupil area and position in the FXS mouse model (Fmr1 KO). Hydrocephalus is a neurological disorder that typically requires a long-term implantation of a shunt system to manage its symptoms. These shunt systems are notorious for their extremely high failure rate. More than 40% of all implanted shunt systems fail within the first year of implantation. On average, 85% of all hydrocephalus patients with shunt systems undergo at least two shunt-revision surgeries within 10 years of implantation. A large portion of this high failure rate can be attributed to biofouling-related obstructions and infections. Probing the functions of human hippocampal subfields is a promising area of research in cognitive neuroscience. However, defining subfield borders in Magnetic Resonance Imaging (MRI) is challenging. Here, we present a user-guided, semi-automated protocol for segmenting hippocampal subfields on T2-weighted images obtained with 7-Tesla MRI. The protocol takes advantage of extant knowledge about regularities in hippocampal morphology and ontogeny that have not been systematically considered in prior related work. An image feature known as the hippocampal ‘dark band’ facilitates tracking of subfield continuities, allowing for unfolding and segmentation of convoluted hippocampal tissue. Amyotrophic lateral sclerosis (ALS) is a fatal neurodegenerative disease; survival in ALS is typically 3-5 years. No treatment extends patient survival by more than three months. Approximately 20% of familial ALS and 1-3% of sporadic ALS patients carry a mutation in the gene encoding superoxide dismutase 1 (SOD1). In a transgenic ALS mouse model expressing the mutant SOD1(G93A) protein, silencing the SOD1 gene prolongs survival. One study reports a therapeutic effect of silencing the SOD1 gene in systemically treated adult ALS mice; this was achieved with a short hairpin RNA, a silencing molecule that has raised multiple safety concerns, and …

Klein & Barron (2016) argue that insects have sentience because of functional similarities between the insect brain and vertebrate midbrain. Based on a recent theory of agency and consciousness, I argue that the functional similarities merely point to an advanced form of agency. Insects presumably lack the capacity for social bonding that may be required for subjective experiencing.

Almost all living organisms have circadian clocks coordinating physiology and behavior, and an innate molecular clock drives rhythmic changes by integrating environmental and metabolic stimuli to generate 24 hour timing. Drosophila melanogaster has proved to be an excellent model organism with a well-characterized circadian clock and the neural circuits underlying clock have been intensely investigated. The neuropeptide pigment-dispersing factor (PDF) plays an essential role in maintaining circadian rhythmicity and synchronizes circadian clock neurons. Dispositional anxiety is an important risk factor for the development of anxiety and other psychological disorders. Symptoms commonly expressed by highly anxious individuals include intrusive memories, uncertainty, and worry — all occurring in the absence of immediate threat. This raises the possibility that anxious individuals have difficulty governing threat’s access to working memory, the mental workspace where goal-related information is actively retained for guiding on-going behavior. Using functional magnetic resonance imaging (fMRI) while 81 subjects completed a well-validated working memory task, I show that threat-related and neutral distracters unnecessarily gain access to working memory, as evidenced by increased neural activity …

The importance of social behaviour is evident in human society, but there are both costs and benefits associated with cooperation and sociality throughout the animal kingdom. At what point do the benefits outweigh the costs, and when do selective pressures favour sociality and colonization over solitude and independence? Brain homeostasis can be affected in a number of ways that lead to gross anatomical, cellular, and molecular disturbances giving rise to diseases like Alzheimer’s disease (AD) and related dementias. Unfortunately, the mechanistic pathoetiology of AD’s hallmark features of cerebral amyloid plaque buildup and neuronal death are still disputed. Using human brain AD sections, immunohistochemistry experiments revealed internalized surface proteins, co-localized to an expanded lysosomal compartment. Other stains for amyloid-β1-42 (Aβ42) and various immunoglobulin (Ig) species displayed them leaking out of the cerebrovasculature through a dysfunctional blood-brain barrier (BBB), binding to neurons in the vicinity, and localizing to intracellular vesicles …

Tumor necrosis factor (TNF) signaling is required for inflammatory nociceptive sensitization in both Drosophila and vertebrates. In Drosophila larval model of nociceptive sensitization, UV irradiation in results in epidermal apoptosis and thermal allodynia. TNF/Eiger is produced from dying epidermal cells and acts its receptor in nociceptive sensory neurons to induce thermal allodynia. Inhibition of TNF signaling results in attenuation of nociceptive sensitization whereas epidermal apoptosis still occurs in the absence of TNF. Major gaps in this model are the precise relationship between apoptotic cell death and production of TNF/Eiger, downstream signaling mediators for TNFR/Wengen, and target genes that alter nociceptive …

Harboring the apolipoprotein E4 (APOE4) allele is the greatest genetic risk factor associated with late-onset (sporadic) Alzheimer’s disease (AD), however the mechanism by which apoE4 contributes to the pathology of AD is unknown. The proteolysis of apoE4 has been suggested to contribute to AD pathology due to a possible toxic gain, or loss of function. In order to determine if apoE4 is being cleaved, we designed and characterized a site-directed cleavage antibody directed at position D151 of full length human apoE4. Acute and chronic pain can be extremely debilitating conditions, and a better understanding of their underlying pathology is necessary to provide effective treatment. C-Fibers are responsible for transmission of the majority of nociceptive signals, with TRPV1+ C-Fibers being specific to noxious heat. Formalin, an inflammatory agent, acts through TRPA1+ C-Fibers, which have a high degree of co-localization with TRPV1. Using a neonatal capsaicin preparation, which irreversibly ablates the majority of TRPV1+ C-Fibers, formalin- induced inflammation and subsequent ATF3 expression was investigated. Glyceollins (Glys), produced by soy plants in response to stress, have anti-estrogenic activity in breast and ovarian cancer cell lines in vitro and in vivo. In addition to known anti-estrogenic effects, Glys exhibit mechanisms of action not involving estrogen receptor (ER) signaling. To date, effects of Glys on brain physiology and function are unknown. Drug addiction is characterized by compulsive drug use and chronic relapse despite negative consequences. Drug-induced structural and functional changes in the brain are thought to underlie these characteristics. One mechanism that may mediate these characteristics are growth factors, such as basic fibroblast growth factor (bFGF or FGF2), as they are necessary for cellular growth, survival, differentiation, and have roles in memory, mood, and anxiety disorders. bFGF mRNA and protein expression is increased following stimulant administration and is necessary for stimulant-induced changes in dendrites and behavioral sensitization. Vascular pathologies represent the leading causes of mortality worldwide, accounting for 31% of all deaths in 2012. Cerebral hypoxia is a condition that often manifests as a result of these medical conditions. Remarkably, the nervous system has evolved mechanisms to compensate for oxygen deprivation. The dilation of existing vessels and the growth of new blood vessels are two prominent physiological responses to hypoxia, both of which play a critical role in maintaining cerebral homeostasis. Anxiety is a significant public health problem characterized by substantial psychological, physical, and economic burden. A key feature of anxiety is the inability to regulate fear. Face recognition is a problem that has theoretical and applied value. However, the fact of facial aging is rarely addressed in research and unmentioned in the major theories of face recognition. Facial aging also has ramifications for missing persons and fugitive cases, confounding attempts by law enforcement to recover these people whose last known images are years or decades out of date. This dissertation reports three studies aimed at measuring baseline age-gap recognition ability and testing various training regimens designed to increase accuracy rates for this unique kind of recognition task.

Previous research has shown that sensory neurons are not exclusively influenced by external stimuli but rather reflect interactions between sensory inputs and the ongoing activity of the brain. Transcranial magnetic stimulation (TMS) produces a transient magnetic field that activates underlying cortical tissue by eliciting an electrical discharge of the neurons in the targeted area. Repetitive TMS (rTMS) uses patterns of repetitive TMS pulses and has been reliably shown to produce changes in the state of cortical excitability outlasting the time of stimulation. One such protocol that has demonstrated states of increased excitability is intermittent theta burst stimulation (iTBS). This method applies high-frequency bursts (50Hz) of pulses every 200 ms in trains of ten bursts. Autism spectrum disorder (ASD) is associated with functional abnormalities of the anterior cingulate cortex (ACC), a brain area that mediates social behavior. Given evidence of a role of inflammation in ASD, markers of pro-inflammatory and anti-inflammatory microglia were studied using postmortem ACC tissues from ASD and age-matched typically developed control donors. Gene expression levels of pro-inflammatory (CD68, HLA-DRA, IL1B, NOS2, PTGS2) and anti-inflammatory (ARG1, IGF1, MRC1, PPARG) microglial genes were measured using quantitative real-time PCR. Infertility is one of the major problems of obesity. Studies have shown that administration of leptin reversed obesity-induced infertility in rats and mice. Coenzyme Q10 (CoQ10) is an antioxidant and also supplies the energy needed for ovulation and embryo development. We hypothesized that leptin when combined with CoQ10 could greatly improve obesity-induced infertility. The results showed a significant decrease in food intake, body weight, and the regular estrous cycle was restored after treatment with leptin+CoQ10. A previous study has found that perceiving degraded speech requires attention, with compromised behavioral and neurological measures of speech processing for degraded speech, but not clear speech, when participants are distracted (Wild et al., 2012b). We extended these findings by examining behavioral and neural correlates of speech perception under different levels of cognitive load using multiple object tracking. We also investigated the role of attention in perceiving degraded speech that was as intelligible as clear speech, in order to separate perceptual outcomes (i.e., intelligibility) from the requisite processing demands. A novel framework for the origins of consciousness and mind, the Cellular Basis of Consciousness (CBC), is presented. The model is based on a simple, perhaps radical axiom: subjectivity is an inherent feature of particular kinds of organic form. Experiential states, including those denoted as "mind" and "consciousness," are present in the most primitive species. The model has several conceptual and empirical virtues, among them: (a) it (re)solves the problem of how minds are created by brains ─ also known as the "Hard Problem" (Chalmers 1995) ─ by revealing that the apparent difficulty results from a category error, (b) it …
