Determination Of Antibiotic Resistance In Salmonella Typhimurium And Salmonella Kentucky Serotypes Of Animal Origin Using Conventional And Molecular Methods, Zeynep Şik, Mehmet Akan

The high incidence of multidrug-resistant Salmonella Typhimurium and Salmonella Kentucky isolates is a concern for human and animal health. This study aimed to investigate the antibiotic resistance determinants of a total of 150 S. Typhimurium and S. Kentucky isolates obtained from cows, calves, lambs, and poultry. Salmonella isolates were tested against 13 different antimicrobials using the Kirby-Bauer disc diffusion method, and genotypic antimicrobial resistance determinants were investigated by polymerase chain reaction. Significant differences were detected among serovars for gentamicin, streptomycin, ampicillin, sulfonamide, nalidixic acid, ciprofloxacin, and tetracycline resistance, with the rates of resistance to these antibiotics being determined to be …

Poultry industry is rapidly moving towards utilizing the best ingredients to precisely feed the birds for better performance, welfare, health, and profitability. One of the practices that evolved over time with research is better optimization of amino acid utilization in the diets thus changing the crude protein landscape. Historically, if we compare the modern diets to diets from 1980, significant changes are visible in the use of unbound or crystalline amino acids leading to reduced levels of CP and reduced unutilized nitrogen in the diets. Highly pathogenic avian influenza viruses (HPAIVs) of the A/goose/Guangdong/1/1996 lineage H5 clade 2.3.4.4b continue to have a devastating effect on domestic and wild birds. Full genome sequence analyses using 1369 H5N1 HPAIVs detected in the United States (U.S.) in wild birds, commercial poultry, and backyard flocks from December 2021 to April 2022, showed three phylogenetically distinct H5N1 virus introductions in the U.S. by wild birds. Unreassorted Eurasian genotypes A1 and A2 entered the Northeast Atlantic states, whereas a genetically distinct A3 genotype was detected in Alaska. In August 2021, the U.S. Environmental Protection Agency (EPA) released draft models to estimate daily NH3, H2S, PM10, PM2.5, and TSP emissions from egg-layer houses (high-rise and manure-belt) and manure storage using inputs of daily mean ambient temperature, relative humidity (RH), and hen inventory. These models were developed from refined datasets generated by the National Air Emissions Monitoring Study fieldwork completed in 2009. Notably, they do not include data for cage-free housing. Currently, 66% of U.S. laying hens are housed in cages; thus, these models, if adopted, will have a substantial impact …

Heat stress (HS) is a growing concern in broiler production. Little is known regarding the effect of HS on immune function. To examine the effects of HS on innate immunity, the local- and systemic-inflammatory responses to lipopolysaccharide (LPS) were examined in Cobb 500 male broiler chicks reared under thermoneutral (TN) or cyclic HS conditions. In August 2021, the U.S. Environmental Protection Agency( EPA) released draft models to estimate daily NH3, H2S, PM2.5, PM10, and total suspended particulate semissions from U.S. broiler operations using inputs of daily mean ambient temperature, relative humidity, and live animal weight. The EPA developed these models using data sets collected between 2005 and 2007. It is important to note that over 15 yr have passed since the original data were collected, and broiler genetics, nutrition, and management have improved consideably. Influenza A viruses are a diverse group of pathogens that have been responsible for millions of human and avian deaths throughout history. Here, we illustrate the transmission potential of H7N9 influenza A virus between Coturnix quail (Coturnix sp.), domestic ducks (Anas platyrhynchos domesticus), chickens (Gallus gallus domesticus), and house sparrows (Passer domesticus) co-housed in an artificial barnyard setting. In each of four replicates, individuals from a single species were infected with the virus. Quail shed virus orally and were a source of infection for both chickens and ducks. The Obese strain (OS) of chickens spontaneously develops autoimmune thyroiditis (SAT) and is a well-established biomedical model for Hashimoto’s thyroiditis in humans. Both conditions are characterized by the infiltration of thyroid glands with mononuclear immune cells resulting in the destruction of thyroid tissue and impairment of the thyroid’s endocrinological functions. Past studies described immune cell infiltration in thyroids of the OS chickens, but the time-course, cell composition, and relative amounts of the various immune cells infiltrating the thyroids have not been well defined. This dissertation aims to identify the bacterial communities responsible for meat spoilage, and the mechanisms that govern the behaviors of these organisms. The present dissertation contains a collection of studies that examine the genomic, proteomic, and metabolomic association to pulmonary hypertension or ascites phenotype in fast-growing broilers. Pulmonary hypertension is a multifactorial metabolic disease influenced by physiological, environmental, and nutritional factors. It is characterized by a number of structural changes including, thrombosis and adverse pulmonary vascular remodeling. Thus, the atrial pressure is increased, and the right ventricle becomes hypertrophied, resulting in heart failure and the death of the bird. This dissertation focused on the efficacy and validity of whole genome resequencing (WGR) for fine mapping genetic determinants of particular traits in a given organism. Previously, our research group used WGR to identify haplotype blocks of single nucleotide polymorphisms associated with ascites resistance with some as strong candidates for use in marker-assisted selection (MAS). Chapter 2 discusses the completion of a MAS project through evaluation of ascites incidence as well as production traits of economic value to poultry producers. Performance and efficiency of feed utilization in poultry is highly influenced by gut health, which is dependent on intestinal microbial balance. Probiotics are live microbial feed supplements or viable microorganisms that beneficially affect the host animal by improving its gastrointestinal tract (GIT) microbial balance. However, their mode of action and suitable GIT environment favoring their colonization of the GIT is obscure. The probiotic properties of Lactobacillus plantarum, Bifidobacterium longum, and Saccharomyces boulardii were evaluated. The effects of flock size of European starlings (Sturnus vulgaris) was experimentally manipulated to assess the potential of influenza A virus (IAV; H4N6) transmission from a flocking passerine to bobwhite quail (Colinus virginianus) through shared food and water resources to mimic starling intrusions into free-range and backyard poultry operations. Of the three starling flock sizes tested (n = 30, n = 20 and n = 10), all successfully transmitted the virus to all or most of the quail in each animal room (6/6, 6/6 and 5/6) by the end of the experimental period, as determined by seroconversion and/or viral RNAshedding. …

The pelleting and extrusion processes use both thermal and mechanical energies to alter the crystalline structure of the starch granule, which makes it more digestible than raw starch. The particle size of the ground corn particle in the diet also affects the rate at which gelatinization occurs in the extrusion process. This endeavor is multifaceted, as many different factors play a part in influencing the productivity of birds, including environmental conditions, overall health, and genetic potential. Spirulina (Arthrospira platensis) is a cyanobacteria microalgae that has potential as an animal feed ingredient. Five experiments were conducted to further assess its ability to serve as a protein source in broiler diets. Experiment 1 consisted of an amino acid digestibility and apparent metabolizable energy assay. The results indicated that the standardized ileal amino acid levels of spirulina are comparable with typical soybean meal. Experiment 2 assessed the performance and processing parameters of broilers fed 10% spirulina in low crude protein diets, using published digestibility coefficients. Formaldehyde fumigation in poultry hatch cabinets has been utilized for sanitation of hatching eggs for over a century. Formaldehyde is utilized to minimize pathogenic microbes on the surface of the egg as well as the microbial bloom during the hatching process. While formaldehyde is effective, its use is regulated in the United States and Europe due to its carcinogenic nature. Formaldehyde has been shown to damage the cuticle of the egg, cause embryonic death, and damage the epithelial lining of the respiratory tract of freshly hatched chicks, predisposing them for respiratory infection. The poultry industry has dominated the dynamic livestock sector in Latin America during the last decades, doubling production in the last 20 years (Williams and Anderson, 2019). In the future, poultry production in the region is expected to grow at almost twice the annual global rate: 2.5% and 4.0%, respectively. This study was developed to assess whether microencapsulation of amino acids (AA) improves their absorption to increase broiler growth performance and reduce nitrogen excretion compared to standard feed-grade AA. Five hundred and sixty Ross 708 male broilers were randomly distributed to 70 battery cages and reared for 21 days. Reported data is grouped into periods: 0-7 days, 0-14 days, and 0-21 days. A total of 5 treatment diets were fed: an industry-based control and 4 diets with decreased levels of methionine, lysine, and threonine in either encapsulated or free forms, with or without a botanical feed additive. Wooden or woody breast (WB) is a myopathy of the pectoralis major in fast-growing broilers that influences the quality of breast meat and causes an economic loss in the poultry industry. The objective of this study was to evaluate growth and proteome differences between 5 genetic strains of broilers that yield WB and normal breast (NB) meat. Eight-week-old broilers were evaluated for the WB myopathy and divided into NB and WB groups. Here, we report three detections of H7N1 low pathogenicity avian influenza viruses (LPAIV) from poultry in Missouri (n¼2) and Texas (n¼1) during February and March 2018. Complete genome sequencing and comparative phylogenetic analysis suggest that the H7 LPAIV precursor viruses were circulating in wild birds in North America during the fall and winter of 2017 and spilled over into domestic poultry in Texas and Missouri independently during the spring of 2018.

While price typically drives consumers’ food-purchasing decisions in low-income countries, religious attributes associated with food production and corporate branding could influence buying patterns. In Mozambique, more than 46% of people were living below the poverty line of ($0.31 USD) per day in 2018. That being said, in the Nampula Province (the location of this study), which is the second poorest province in the country, over 25% of the population is Muslim and may be willing-to-pay (WTP) a premium for Halal meat products to uphold Islamic beliefs.
