Chlorine has effectively reduced the prevalence of waterborne diseases, however there are secondary consequences to this public health advancement. Disinfection byproducts (DBPs) are chemicals formed when chlorine reacts with natural organic matter (NOM) in water. A new class of DBPs, halobenzoquinones (HBQs), has recently been identified and data suggests it could be potentially carcinogenic and up to 1000 times more toxic than some regulated DBPs. The use of antibiotics in animal feed is a large cause of concern due to bacterial resistance, which occurs when bacteria change after exposure to antibiotics and become less affected by the drug. Consequently, the desire to find a replacement for these antibiotics has garnered interest in both farmers and consumers. The ideal substance would display antimicrobial activity without promoting bacterial resistance and would still function as a growth promoter in animals. Receptor kinases (RKs) are transmembrane proteins that have been shown to regulate an array of important processes in A. thaliana, including polar cell growth, plant reproduction, and many other plant growth processes. In this thesis, I examine RECEPTOR KINASE 7 (RK7) and FERONIA (FER), two closely related transmembrane RKs, and their effects on plant reproduction. The RK7 gene when knocked out (rk7) in conjunction with FER resulted in delayed plant growth, decreased seed yield, and a lower percentage of the seeds germinating as compared to the single FER knockout. The developing pancreas has been identified as a potentially sensitive target for oxidative damage caused by environmental toxicant exposure in the developing zebrafish, Danio rerio. The objectives of this study were to 1) investigate the effects of exposure to a common pro-oxidizing toxicant, Mono(2-ethylhexyl) phthalate (MEHP), on pancreatic development, and 2) elucidate redox-sensitive temporal windows of pancreatic organogenesis in the zebrafish. Declines in several pollinator species are due to a variety of factors, including pathogens. Incorporating pollinator-friendly plant species into wild and agricultural habitats could reduce the stress of pathogens if food sources act medicinally against pathogens. Previous research demonstrated one domesticated sunflower cultivar (Helianthus annuus) can dramatically reduce a gut pathogen (Crithidia bombi) of the common eastern bumble bee, Bombus impatiens. The T cell-specific kinase, Protein Kinase C theta (PKCq) is essential to T cell activation and differentiation. PKCq integrates T cell receptor (TCR) and CD28 signaling, and ultimately activates transcription factors necessary for full T cell activation, proliferation, survival, and differentiation into T helper (Th) subsets. Th1, Th2, Th17 and Treg cells compose the four major lineages of T helper cells, differentiated from CD4 T cells, and each have different requirements for PKCq.

Double strand breaks (DSB) are a common source of DNA damage in both prokaryotes and eukaryotes. If they are not repaired or are repaired incorrectly, they can lead to cell death (bacteria) or cancer (humans). In Escherichia coli, repair of DSB are typically accomplished via homologous recombination and mediated by RecA. This repair pathway, among others, is associated with activation of the SOS response. DNA adenine methyltransferase (dam) mutants have an increased number of DSB and, therefore, are notorious for being RecA-dependent for viability. Trypanosomatid parasites such as Trypanosoma brucei have unusual mechanisms of gene expression including polycistronic transcription, mitochondrial RNA editing and trans-splicing. Additionally, these protists rely mainly on post-transcriptional regulation where RNA-binding proteins (RBP) have shown to play a major role. RBP6 and RBP10 are two examples of RBPs that play crucial roles in procyclic and bloodstream form parasites differentiation respectively, by post-transcriptional regulation. The accessory olfactory bulb (AOB) processes pheromonal signals which in turn drive social behaviors. Here we identify a tract of aromatase-expressing (arom+) fibers in the dorsal lateral olfactory tract (dLOT) which terminate in the granule cell layer (GCL) of the AOB. We utilized a retrograde tracer in aromatase reporter animals to delineate the source of these fibers. We show that these input fibers emerge almost exclusively from a contiguous population of arom+ neurons that spans the bed nucleus of the accessory olfactory tract (BAOT) and posterioventral subnucleus of the medial amygdala (MeApv). The neuropeptide hormone arginine-vasopressin (AVP) is produced in both the supraoptic and paraventricular nuclei of the hypothalamus. Expression within these nuclei is conserved across species, and species differences in the expression of AVP and its cognate receptors correlate with differences in social behavior. The Food Safety Modernization Act (FSMA) has a regulation stating that there can be no detectable generic Escherichia coli in agricultural water. The objective of this study was to optimize the use of peracetic acid (PAA) as a mitigating strategy in postharvest processing. This project began by investigating the impact of storage temperatures on stability of a commercially available PAA sanitizer. Regression analysis showed a quadratic relationship in the concentration of PAA over 12 weeks when storing the product at 4, 20 and 37°C. Gypsophila paniculata is an invasive species in Michigan’s northern lower peninsula and a problem invasive in much of the northern United States and Canada. Gypsophila paniculata readily outcompetes native plants in sandy, well-drained soils due to its deep taproot, which allows access to scarce resources. It reproduces and disperses mainly by seed, but the phenology of seed maturation is poorly understood. In animal and yeast cells, cell division (cytokinesis) is facilitated by the formation of a contractile acto-myosin ring (CAR). Proper CAR formation and constriction is heavily reliant on the temporal regulation, phosphorylation, and localization of key proteins. In the fission yeast S. pombe, Mid1 is an important dimeric CAR scaffolding protein that connects the contractile apparatus to the plasma membrane at the right place and time during cytokinesis. Sediment pollution is a major cause of stream degradation. Our objectives were threefold. First, we assessed the impacts of environmental stressors on the structure of fish and macroinvertebrate communities across a gradient of agricultural to urban land cover in a coldwater tributary of the Grand River, Michigan, USA called Indian Mill Creek. We found that instream woody debris, streambed substrate, riffle and pool habitat variability, and riparian conditions affected aquatic macroinvertebrates along an agricultural to urban land cover gradient. Stream ecosystem metabolism is commonly measured in stream ecology studies in order to understand the functioning of the stream ecosystem and as an indicator of stream health. One common method for gathering the time series data required to estimate stream metabolism is the free water dissolved oxygen method, which involves measuring dissolved oxygen in freely moving water. This is accomplished by taking measurements at a single location (one-station monitoring method) or at two locations (two-station monitoring method). Invasive species, including the non-native forb Centaurea stoebe (spotted knapweed), constitute an imminent threat to degraded and restored native prairies. Considering the major threat that C. stoebe poses to imperiled prairie ecosystems, I examined the effectiveness of fire as a control agent of C. stoebe and (±)-catechin. I conducted a 2-year experiment in part of a restored tallgrass prairie community at Pierce Cedar Creek Institute in Barry County, Michigan between May and August of 2016 and 2017. Wetlands in dune landscapes provide important breeding habitat for amphibians along the Lake Michigan Coast. Unfortunately, these unique habitats and the corresponding amphibian metapopulations are understudied and threatened. We assessed amphibian species richness and terrestrial habitat type in 16 permanent and ephemeral wetlands along the coast of Lake Michigan in Grand Haven, MI. Wetland area, terrestrial habitat type, depth, hydroperiod, shade, and degree of isolation were measured from April to September of 2017. Fusion of cancer cells has been observed in tumors for more than a century and is thought to contribute to tumor development and drug resistance. The low frequency of cell fusion events and the instability of fused cells have hindered our ability to understand the molecular mechanisms that govern cell fusion. We have developed a patterned gel system that can isolate cell fusion events and we demonstrate that several breast cancer cell lines can fuse into multinucleated giant cells in vitro, and the initiation and longevity of fused cells can be regulated solely by biophysical factors.
