Prolactin concentrations in avian plasma have been traditionally quantified using a radioimmunoassay (RIA) procedure. RIA is useful for such studies due to its sensitivity and specificity (e.g., Follett et al. 1972, Wingfield and Farner 1975, McNeilly et al. 1978, Burke and Papkoff 1980, Schwabl 1993). For example, the heterologous RIA for detecting prolactin in turkeys, developed by McNeilly et al. (1978), recovered 98% of a prolactin standard added to a turkey plasma sample and exhibited less than 0.1% cross reactivity with other plasma hormones. It was recently reported (H. Akutsu, J.-S. Park, and S. Sano, J. Am. Chem. Soc. 115:12185–12186, 1993) that in the strict anaerobe methyl groups from exogenous L-methionine are incorporated specifically into the 1 and 3 positions (Fischer numbering system) on the heme groups of cytochrome c3. It was suggested that under anaerobic conditions, protoporphyrin IX biosynthesis proceeds via a novel pathway that does not involve coproporphyrinogen III as a precursor but instead may use precorrin-2 (1,3-dimethyluroporphyrinogen III), a siroheme and vitamin B12 precursor which is known to be derived from uroporphyrinogen III via methyl transfer from S-adenosyl-L-methionine. In many filamentous cyanobacteria nitrogen fixation occurs in differentiated cells called heterocysts. Filamentous strains that do not form heterocysts may fix nitrogen in vegetative cells, primarily under anaerobic conditions. We describe here two functional Mo-dependent nitrogenases in a single organism, the cyanobacterium Anabaena variabilis. Using a lacZ reporter with a fluorescent beta-galactoside substrate for in situ localization of gene expression, we have shown that the two clusters of nif genes are expressed independently. One nitrogenase functions only in heterocysts under either aerobic or anaerobic growth conditions, whereas the second nitrogenase functions only under anaerobic conditions in vegetative cells and heterocysts. …

A nephritic condition was developed by infecting Swiss Webster albino mice with the malarial parasite Plasmodium berghei NK 65. These animals were tested for urinary protein and the presence of circulating immune complexes using reagent strips and a polyethylene glycol (PEG) precipitation assay. The circulating immune complexes were isolated from the sera using both affinity chromatography and PEG precipitation and from the kidney by acid elution. The isolated complexes were dissociated into their individual components and analyzed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). The components of the complexes were transferred to nitrocellulose sheets and probed for the presence of …

The Heterochronic Gene Lin-29 Encodes A Zinc Finger Protein That Controls A Terminal Differentiation Event In Caenorhabditis Elegans, Ann Rougvie, Victor Ambros

A hierarchy of heterochronic genes, lin-4, lin-14, lin-28 and lin-29, temporally restricts terminal differentiation of Caenorhabditis elegans hypodermal seam cells to the final molt. This terminal differentiation event involves cell cycle exit, cell fusion and the differential regulation of genes expressed in the larval versus adult hypodermis. lin-29 is the most downstream gene in the developmental timing pathway and thus it is the most direct known regulator of these diverse processes. We show that lin-29 encodes a protein with five zinc fingers of the (Cys)2-(His)2 class and thus likely controls these processes by regulating transcription in a stage-specific manner. Consistent …

In Caenorhabditis elegans, the terminal differentiation of the hypodermal cells occurs at the larval-to-adult molt, and is characterized in part by the formation of a morphologically distinct adult cuticle. The timing of this event is controlled by a pathway of heterochronic genes that includes the relatively direct regulatory gene, lin-29, and upstream genes lin-4, lin-14 and lin-28. Using northern analysis to detect endogenous collagen mRNA levels and collagen/lacZ reporter constructs to monitor collagen transcriptional activity, we show that the stage-specific switch from larval cuticle to adult cuticle correlates with the transcriptional activation of adult-specific collagen genes and repression of larval-specific …

Despite their similarity in structure, pyrrolizidine alkaloids (PAs) vary in their LD50s and in the organs in which toxicity is expressed. We have examined whether there are differences in the metabolism of certain PAs that are associated with these quantitative and qualitative differences in toxicity. Isolated rat livers were perfused with one of four PAs (seneciphylline, retrorsine, monocrotaline, and trichodesmine) at 0.5 mM for 1 hr, and the pyrrolic metabolites determined that were released into perfusate and bile or bound in the liver. Two emergence peaks were evident for adult Douglas-fir beetles (DFB), Dendroctonus pseudotsugae Hopkins, collected from caged brood trees in 1994. The June peak resembled emergence patterns observed in previous years; however, an August peak was detected for the first time since monitoring of the outbreak began. The second peak likely resulted from delayed development of brood of the '93-'94 generation that largely overwintered as larvae. A portion of the '92-'93 generation also emerged in 1994, confirming that DFB can extend its life cycle to two years under certain conditions. Visitations of flowers of the Crown Imperial by Great Tits at two different locations in Hessen, Germany, in 1990 and 1993 are described. The observations prove that flowers were visited because of the nectar; most likely pollination occurs during this visits. Similar reports from Europe are discussed. The significance of specific interactions including ecological generalists such as the Great Tit is considered in the context of the phylogenetic reconstruction of the development of co-adaptive syndroms.

Transient-gear aquaculture is a method of growing shellfish that is designed to minimize multiple-use conflicts in common use public trust waters, and avoiding the need for a conventional fixed aquaculture lease site. A pro-forma economic model of an oyster farm using transient gear in Rhode Island's Narragansett Bay is provided, and the advantages and disadvantages of transient gear aquaculture are discussed.
