The coat proteins of the RNA bacteriophages Qβ and MS2 are specific RNA binding proteins. Although they possess common tertiary structures, they bind different RNA stem loops and thus provide useful models of specific protein-RNA recognition. Although the RNA-binding site of MS2 coat protein has been extensively characterized previously, little is known about Qβ. Here we describe the isolation of mutants that define the RNA-binding site of Qβ coat protein, showing that, as with MS2, it resides on the surface of a large β-sheet. Mutations are also described that convert Qβ coat protein to the RNA binding specificity of MS2. …

A diverse group of xenobiotics has a high binding affinity to the estrogen receptor (ER), suggesting that it can accommodate large variability in ligand structure. Relationships between xenobiotic structure, binding affinity, and estrogenic response have been suggested to be dependent on the conformational structures of the ligands. To explore the influence of conformational flexibility on ER binding affinity, a quantitative structure—activity relationship (QSAR) study was undertaken with estradiol, diethylstilbestrol, and a set of polychlorinated hydroxybiphenyls (PCHBs) of environmental concern. Cryptotis avia G. M. Allen 1823, from the Eastern Andean Cordillera of Colombia, is a small-eared shrew previously known from, at most, five specimens. Review of the holotype, consisting of an understuffed skin and a partial skull, and three potential topotypes indicates that they are refereable to Cryptotis thomasi Merriam, 1897, and C. avia should be considered ajunior synonym of that name. Cryptotis thomasi is restricted to highlands around Bogotá in the Eastern Cordillera. An experiment was conducted to characterize the effects of postruminal administration of casein, glutamine, cornstarch, and water on protein turnover and in vitro muscle cell proliferation. Four MARC III steers (205 kg) were fed a protein-restricted bromegrass hay-based diet (2.86 Mcal of DE/kg and 13.6 g of N/kg). Using a 4 x 5 Latin square arrangement balanced for residual effects, casein and glutamine, equal to 50% of basal dietary nitrogen intake, cornstarch, isocaloric with casein infusion, or an equal volume of water was continuously infused into the abomasum of steers. ABP-1 had long been thought to be localized exclusively in epidermal cells in coleoptiles (Löbler and Klämbt, 1985), and thus had fitted nicely the “epidermal-growth-control-hypothesis” (Kutschera, 1987, 1992). Emergence patterns and population levels of Douglas-fir beetles (DFB), Dendroctonus pseudotsugae Hopkins, in the seventh year of infestation following the Clover Mist Fire were evaluated on the Clarks Fork Ranger District of the Shoshone National Forest. Adult emergence in 1995 returned to a single peak beginning in mid-June, about a week later than normal likely due to cold, wet spring weather. Adults of both the '93-'94 and '94-'95 generations emerged, demonstrating for the second year in a row that DFB can extend its life cycle up to two years. In North America, the Colorado potato beetle, Leptinotarsa decemlineata, is often infected with the host-specific, gut-inhabiting Colorado potato beetle spiroplasma (CPBS). CPBS is apparently a commensal, but it may be useful in biocontrol if it can be transformed to express an insect-lethal gene. Difficulty in cultivating the organism, however, has hindered the development of a suitable transformation system. In this study, we eliminated the need for coculturing CPBS with insect cells. CPBS was reliably isolated with the BBL Anaerobic GasPak Jar system (low redox, enhanced CO(inf2)), which was easier to use and less expensive than insect cell coculture methods. A …

Studies of mating patterns of tropical trees, typically involving common species, have revealed that most species are outcrossed and that, in some cases, a significant reaction of outcross pollen moves long distances. We evaluated mating systems and effective pollen dispersal for three hermaphroditic insect-pollinated Neotropical tree species, Calophyllum longifolium, Spondias mombin and Turpinia occidentalis, all of which occurred at low adult densities at the study site. In Caenorhabditis elegans, the fates of the multipotent vulval precursor cells (VPCs) are specified by intercellular signals. The VPCs divide in the third larval stage (L3) of the wild type, producing progeny of determined cell types. In lin-28 mutants, vulva development is similar to wild-type vulva development except that it occurs precociously, in the second larval stage (L2). Consequently, when lin-28 hermaphrodites temporarily arrest development at the end of L2 in the dauer larva stage, they have partially developed vulvae consisting of VPC progeny. During post-dauer development, these otherwise determined VPC progeny become reprogrammed back to the multipotent, signal-sensitive state …

The NF-kappaB/c-Rel proteins are a family of evolutionarily conserved transcription factors activated during development that in the adult, mediate many processes including the immune response. A high degree of sequence similarity is shared between the NF-kappaB/c-Rel family of transcription factors and the Drosophila Dorsal protein as well as between its cytoplasmic inhibitor, IkappaBalpha, and the Drosophila Cactus protein. Genetic analyses of Dorsal have defined components of a signaling pathway for Dorsal activation, including a serine/threonine kinase, Pelle, placed upstream of Dorsal and Cactus. Lung injury in bacterial infection is a multifactorial phenomenon that involves bacterial metabolites and host factors. Primary isolates of type II pneumocytes and established cultures of Madin-Darby canine kidney (MDCK) cells were used to study effects of Pseudomonas aeruginosa exoproducts on epithelial paracellular permeability. The results indicate that elastase (PE) and exotoxin A (Exo A) have different, but complementary, actions that diminish epithelial barrier function. We measured transepithelial electrical resistance (TER) and permeability coefficient for mannitol (Pm) across cell monolayers plated on tissue culture membranes. In the present study we reconstruct older models of mechanical inhomogenities in growing plant organs, in order to establish an accurate historical background for the current discussion. We focus on the iatromechanic model developed in Stephen Hales' Vegetable Staticks, Wilhelm Hofmeister's mechanical model of negative geotropism, Julius Sachs' explanation of the development of tissue tension, and the differential-auxin-response-hypothesis by Kenneth Thimann and Charles Schneider. We have applied a high-speed counter-current chromatography (CCC) technique to the separation and purification of pyrrolizidine alkaloids from Amsinckia tessellata, Symphytum spp., Trichodesma incanum (Boraginaceae), and Senecio douglasii var. longilobus (Asteraceae). Alkaloidal fractions were separated in a solvent system composed of a chloroform mobile phase and 0.2 M potassium phosphate buffer, of an optimum pH, as the stationary phase. The implementation of site-specific crop management is dependent on the variations in yield and yield potential within a field. Crop yield maps are important for both the implementation and evaluation of site-specific crop management strategies. Management decisions and evaluations based on yield maps must take into consideration the accuracy and resolution of the maps.

An impact-based yield monitor and a volumetric yield monitor were compared. The effect of different dynamic models of combine grain flow on the calculated instantaneous yields were investigated. Both simple time delay models and first order models could be used to model the grain flow. In …

A study was conducted to maxmuze plant regeneration frequencies from cultured anthers of' Chandler', 'Honeoye', and 'Redchief strawberries (Fragaria x ananassa Duch.). A comparison of auxins (IAA, NAA), cytokinins (BA, BPA, KIN) and carbohydrates (sucrose, glucose, maltose) in MS medium showed that the highest shoot regeneration across cultivars (8%) occurred when using a medium containing 2 mgtl IAA, I mgtl BA, and 0.2 M glucose. A comparison ofMS, NN, and HI inorganic medium (a new formulation based on the anther culture literature) solidified with either agar or gellan gum and containing IAA, BA, and glucose, showed the highest shoot regeneration …
