A variety of anthropogenic factors, including global climate change and eutrophication, are causing increases in cyanobacterial abundance. Increased prevalence of cyanobacteria can be detrimental, as some genera of cyanobacteria have the ability to produce toxic secondary metabolites known as cyanotoxins. The cyanotoxin microcystin- a hepatotoxin, is the most ubiquitous and toxic cyanotoxin in freshwater ecosystems, and has been quantified in a number of aquatic organisms. Hydrogen exerts thermodynamic control over the exclusion of methanogens by sulfate reducers in Cape Lookout Bight, NC, marine sediments. This has been demonstrated by previous in situ measurements, but has never been demonstrated in a batch incubation of unamended sediments and has never been combined with identification of the microorganisms involved in this process. We made triplicate anoxic incubations of sediments from the upper 3 cm of sediment over 122 days while taking weekly samples for DNA extraction, cell counts, and measurements of methane, sulfate, and hydrogen. At highly elevated temperatures, many biological reactions can proceed spontaneously from the ground state to the transition state. However, due to the long half-life of these reactions, catalysts are required to catalyze these reactions at modern day temperatures by lowering the activation energy. Wolfenden et al. has previously shown that catalysts enhance the rate of the reaction by reducing the enthalpy of activation. Determining an accurate estimation of the postmortem interval (PMI) of human remains is important for several reasons. First, it is used to determine whether the individual is recently deceased, and therefore of medicolegal concern. If so, the estimated PMI is used in narrowing the possible identities of the deceased. PMI can also be used in excluding, or convicting, a murder suspect.

INTRODUCTION: Consumer-based physical activity (PA) monitors are increasingly common, and must be validated against criterion measures to determine which models are accurate. Such studies will aid PA intervention research and individual consumers purchasing these devices. METHODS: Thirty participants (mean ±[plus or minus] SD; age, 25.5 ± 3.7 years; BMI, 24.9 ± 2.6 kg/m2[meters squared]) completed a structured PA routine including 11 activities ranging from sedentary behaviors to vigorous intensities. Accurate and unbiased estimates of current effective population size are of primary importance in making informed decisions for conservation purposes. One species of concern, the eastern red bat (Lasiurus borealis), is experiencing ongoing population losses due to wind turbine collisions. The proportion of the population being affected is currently unknown, although recent estimates of total fatalities at wind turbines over the time from 2000-2011 are in the hundreds of thousands for this species alone. Previous studies have found that addition of osmolytes weakens the binding of dihydrofolate (DHF) to R67 dihydrofolate reductase (DHFR), chromosomal DHFR from E. coli and a pteridine reductase. These results support the preferential interaction of DHF with osmolytes compared to water. Thus, a working model where interaction of DHF with osmolytes shifts the binding away from the protein-DHF complex towards the free species was proposed. As tetrahydrofolate and other folate redox states have similar structures to DHF, we predict osmotic stress will lower the catalytic efficiencies of other folate pathway enzymes. Throughout the thesis, I study mathematical models that can help explain the dependency of social phenomena in animals and humans on individual traits. The first chapter investigates consensus building in human groups through communication of individual preferences for a course of action. Individuals share and modify these preferences through speaker listener interactions. Personality traits, reputations, and social networks structures effect these modifications and eventually the group will reach a consensus. Enforcement is a key feature of the Clean Water Act. The Clean Water Act’s National Pollutant Discharge Elimination System (NPDES) permit program regulates effluent into streams that may impact aquatic life. Yet, authorities do not always enforce permits when violations occur. My research examines macroinvetebrate survey data near NPDES permitted facilities in Kentucky and Tennessee to determine the influence of enforcement actions on aquatic biodiversity. Pre-existing data from the Environmental Protection Agency and state agencies was used to make quantitative and spatial comparisons.

In this study, Phytophthora infestans and Peronospora farinosa f. sp. spinaciae populations were analyzed using single nucleotide polymorphisms (SNPs). In Ecuador and Colombia, Phytophthora infestans causes significant damage to potato and tomato and the epidemiology is known to be highly clonal. Our objective was to measure population structure within the context of this clonal epidemiology using both synonymous and nonsynonymous markers. Candidate SNP sites were selected by comparing the draft genomes of the Ecuadorian isolates EC1-3527 and EC1-3626. Background: Exposure to various types of food cues via television commercials may enhance consumption of different foods, particularly in females who engage in cognitive efforts to control food intake (i.e., restrained eaters). However, it is unknown if food-related commercials differing in health perceptions impact on intake of foods with differing health perceptions. The purpose of this study was to determine the thermal inactivation kinetics (D and z-values) of Lactobacillus bulgaricus ATCC 11842, Lactobacillus fructivorans ATCC 8288, Lactobacillus plantarum ATCC 4008, Listeria innocua ATCC 33091, ATCC 51742, and 11/97, and Staphylococcus carnosus CS-299 as potential surrogates for hepatitis A virus (HAV; D72°C = 0.9 min). Freshwater mussels (Family Unionidae and Margaritiferidae) are a widely threatened group of bivalve molluscs, particularly in the Southeastern United States. Translocation of freshwater mussels is an increasingly common conservation method. However, there are relatively few studies that quantitatively investigate the factors influencing translocation success or failure. In October 2013, hundreds of Medionidus conradicus and Lampsilis fasciola were translocated to the Pigeon and Nolichucky Rivers in Tennessee, with an interim partial survey (June 2014) and a full survey (October 2014). Background: It is suggested that increasing intake of fruits and vegetables may lead to decreased energy intake via substitution of higher energy-dense foods, such as snack foods (SF; i.e. candy, chips, cookies). This study investigated the impact of increasing fruit (F; grapes) intake, via increased portion size, on SF (potato chips) intake during a meal and whether increases in F intake affected total meal energy intake, via reducing SF intake (substitutive eating), in normal-weight adults. Compared to rodents, broiler chickens, those reared for meat, are an attractive model for studies of adipose biology, and obesity development in children. The broiler chicken lacks the gene for uncoupling protein 1, the hallmark for brown adipose tissue making them a useful model to study lipid metabolism in white adipocytes. The release of fly ash at the Tennessee Valley Authority (TVA) Kingston Fossil Plant (KIF) on 22 December 2008 discharged approximately 4.1 million cubic meters of coal ash into the adjacent aquatic and terrestrial systems. Previous benthic invertebrate investigations conducted by TVA and collaborative researchers concluded that benthic invertebrates in the Emory River were at moderate risk from ash-related constituents, primarily arsenic, in ash-contaminated sediment that remained in the Emory River following extensive dredging efforts. This master’s thesis investigates the effects of prescribed fire and overstory thinning on bats and their insect prey in hardwood forest stands of Tennessee. Chapter 1 is a review of literature that emphasizes the importance of this research and outlines the objectives and study area for this project. Chapter 2 examines the effect of prescribed fire and overstory thinning on the abundance and biomass of nocturnal flying insects important in the diet of bats. Objective: The objectives of this research were to describe the proportion of packaged foods with front-­‐of package nutrition marketing or marketing to children that were high in saturated fat, sodium, and/or sugar content, and to delineate changes in the proportions of these foods from 2007 to 2014.

Methods: Front-­‐of-­‐pack nutrition marketing, child marketing, and nutrient criteria from the Nutrition Facts Label on all packaged food items in a large Midwestern supermarket were recorded in 2007 (n = 5,500) and again in 2014 (n = 6,324). Background: A few studies have shown that art–based nutrition education (primarily in the form of theater) can increase nutrition knowledge and change behavior. Little research exists on the use of visual arts in nutrition education programs. The purpose of this study was to determine the impact on children’s nutrition knowledge, self-efficacy, and behavior, of using visual art and differing levels of intensity of art activities in nutrition education.

Asian earthworms (Amynthas spp.) are invading North American forests and consuming the vital detrital layer that forest floor biota (including the Eastern Red-backed Salamander, Plethodon cinereus) rely on for protection, food, and habitat. Salamander population decline has been associated with European earthworm-mediated leaf litter loss, but there have been few studies on the interactions between Amynthas spp. and P. cinereus. Since the large, active Amynthas spp. earthworms spatially overlap with salamanders beneath natural cover objects and in detritus, they may compound the negative consequences of resource degradation by physically disturbing important salamander activities (foraging, mating, and egg brooding). Islands have long fascinated ecologists and evolutionary biologists because they provide replicated systems with which to test hypotheses regarding evolutionary and ecological theory. Because islands are relatively simple systems, they enable researchers to isolate specific factors responsible for observed phenomena. Many current questions in ecology and evolutionary biology have been addressed in island systems. For example, factors involved in the genetic divergence of populations are easier to discern in island populations because confounding effects of gene flow from adjacent populations are limited.
