Alzheimer’s disease (AD) is a progressive neurodegenerative brain disorder that leads to a progressive decline in a person’s memory and ability to communicate and carry out daily activities. The brain pathology in AD is characterized by extensive neuronal loss, particularly of cholinergic neurons, intracellular neurofibrillary tangles composed of the tau protein (NFTs) and extracellular deposition of plaques composed of β-amyloid (Aβ), a cleavage product of the amyloid precursor protein (APP). Direct heating of a metal-organic framework provides a simple, controllable way of effecting a covalent post-synthetic modification. Herein we report that an allyloxy-tagged zinc metal-organic framework undergoes a thermally-promoted aromatic Claisen rearrangement through which the framework connectivity and porosity are maintained.

The paper describes the bioassay-guided isolation, structure elucidation and anticancer evaluation of five flavonoids (-)-liquiritigenin (1), (-)-neoliquiritin (2), isoliquiritigenin (3), isoliquiritin (4) and formononetin (5) from the twigs of Jacaranda obtusifolia H. B. K. ssp. rhombifolia (G. F. W. Meijer) Gentry. The structures were elucidated based on 1H, 13C NMR, comprehensive 2D NMR, MS analyses and comparison with previously reported spectral data. Compounds 1 and 3 were demonstrated to be inhibitory in vitro against NCI-H187 (small cell lung cancer) with IC50 values of 30.1 and 16.6 μg mL-1, respectively. Translocating birds to a new area of habitat to restore or supplement depleted populations may pose a significant threat to the translocated individuals. While for many species, translocated individuals appear to move larger distances than resident animals, species with poor dispersal capacity may be restricted in movements and translocation methods may need to accommodate differences in movements to ensure success. [1] The goal of this study is to determine how H2O and HDO measurements in water vapor can be used to detect and diagnose biases in the representation of processes controlling tropospheric humidity in atmospheric general circulation models (GCMs). We analyze a large number of isotopic data sets (four satellite, sixteen ground-based remote-sensing, five surface in situ and three aircraft data sets) that are sensitive to different altitudes throughout the free troposphere. Streptococcus pyogenes is a major human pathogen responsible for numerous diseases ranging from uncomplicated skin and throat infections to severe, life threatening invasive disease such as necrotising fasciitis and streptococcal toxic shock syndrome. These severe invasive infections progress rapidly and produce high rates of morbidity and mortality despite the implementation of aggressive treatment plans. The activation of plasminogen and the acquisition of plasmin activity at the bacterial cell surface is critical for the invasive pathogenesis of this organism. "The strong mechanistic relationship between climatic factors and seed dormancy and germination suggests that forecast climatic changes will significantly affect seed bank persistence. This review focuses on the potential impact of changing temperature, rainfall and fire regimes on the longevity of long-term persistent seed-banks. Extracellular protein misfolding and aggregation underlie many of the most serious amyloidoses including Alzheimer's disease, spongiform encephalopathies and type II diabetes. Despite this, protein homeostasis (proteostasis) research has largely focussed on characterising systems that function to monitor protein conformation and concentration within cells. We are now starting to identify elements of corresponding systems, including an expanding family of secreted chaperones, which exist in the extracellular space. In recent genome-wide association studies, the extracellular chaperone protein, clusterin, has been identified as a newly-discovered risk factor in Alzheimer's disease. We have examined the interactions between human clusterin and the Alzheimer's disease-associated amyloid-β 1-40 peptide (Aβ 1-40), which is prone to aggregate into an ensemble of oligomeric intermediates implicated in both the proliferation of amyloid fibrils and in neuronal toxicity. Using highly sensitive single-molecule fluorescence methods, we have found that Aβ 1-40 forms a heterogeneous distribution of small oligomers (from dimers to 50-mers), all of which interact with clusterin to form long-lived, stable complexes. Mobilization of arsenic from freshwater and estuarine sediments during the transition from oxic to anoxic conditions was investigated using recently developed diffusive sampling techniques. Arsenic speciation and Fe(II) concentrations were measured at high resolution (1−3 mm) with in situ diffusive gradients in thin films (DGT) and diffusive equilibration in thin films (DET) techniques. Water column anoxia induced Fe(II) and As(III) fluxes from the sediment. A correlation between water column Fe(II) and As(III) concentrations was observed in both freshwater (rs = 0.896, p < 0.001) and estuarine (rs = 0.557, p < 0.001) mesocosms. Porewater sampling by DGT and DET techniques confirmed that arsenic mobilization was associated with the reductive dissolution of Fe(III) (hydr)oxides in the suboxic zone of the sediment; a relationship that was visible because of the ability to measure the coincident profiles of these species using combined DGT and DET samplers. The selective measurement of As(III) and total inorganic arsenic by separate DGT samplers indicated that As(III) was the primary species mobilized from the solid phase to the porewater. This measurement approach effectively ruled out substantial As(V) mobilization from the freshwater and estuarine sediments in this experiment. This study demonstrates the capabilities of the DGT and DET techniques for investigating arsenic speciation and mobilization over a range of sediment conditions.

Thirty two new stemofoline analogues were prepared from didehydrostemofoline for studies as AChE inhibitors. C-3 side-chain modified amino, carbamate, triazole and oxazole stemofoline derivatives were prepared. In general the amine derivatives were found to be stronger inhibitors of AChE than their alcohol analogues that we previously reported. Compounds 5 and 26, with small C-3 side chain substituents, were two of the most active inhibitors. Preliminary molecular docking studies suggested that these compounds may inhibit AChE by binding horizontally along the passage of the active-site gorge and block access to acetylcholine.

Highly transparent polyethylene terephthalate (PET) mesh fabrics were dip coated with carbon nanotubes (CNT) followed by inkjet printing with poly(3,4-ethylenedioxythiophene):poly(styrene sulfonate) (PEDOT:PSS) to develop textile-based CNT/PEDOT:PSS composite conductors. These coated fabrics show a combination of conductivity, transparency and flexibility suitable for use in flexible displays. Sequential CNT and PEDOT:PSS coating greatly enhances the conductivity at high tensile strains without reducing the optical transparency. Captive breeding and release is a tool used by conservation biologists to re-establish populations of endangered or locally extinct species. Reintroduced animals that have been bred in captivity must learn to meet the challenges posed by free living, and to adjust to local environmental conditions, food and water sources. Gelapin hydrogels were comprehensively characterised through chemical, mechanical and physical analysis techniques. It was found that the hydrogels could be cross-linked to up to 90% using a genipin concentration of 24.4% (w/ w). The hydrogels reach a stable swollen state and cease leaching of residual starting materials after 72 h in phosphate buffered saline solution at 37 uC. Members of the Calliphoridae (blowflies) are significant for medical and veterinary management, due to the ability of some species to consume living flesh as larvae, and for forensic investigations due to the ability of others to develop in corpses. We retrieved column-averaged dry air mole fractions of atmospheric carbon dioxide (XCO 2) from backscattered short-wave infrared (SWIR) sunlight measured by the Japanese Greenhouse gases Observing SATellite (GOSAT). Over two years of XCO 2 retrieved from GOSAT is compared with XCO 2 inferred from collocated SWIR measurements by seven ground-based Total Carbon Column Observing Network (TCCON) stations. The average difference between GOSAT and TCCON XCO 2 for individual TCCON sites ranges from -0.87 ppm to 0.77 ppm with a mean value of 0.1 ppm and standard deviation of 0.56 ppm. We present a comparison of techniques for estimating atmospheric emissions from fires using Australia's 2009 "Black Saturday" wildfires as a case study. Most of the fires started on Saturday the 7th of February 2009 (a date now known as "Black Saturday") and then spread rapidly, fanned by gale force winds, creating several firestorms and killing 173 people. The essential oils from the leaves and fruits of Bridelia retusa (L.) A.Juss. were isolated by hydrodistillation. The essential oils were obtained in 0.0013% yield as a pale yellow liquid and 0.0026% yield as a violet-light brown liquid for the leaf oil and fruit oil respectively. The composition of each essential oil was analysed by means of GC-(FID) and GC-MS. Eleven constituents accounting for 48.77% of total leaves oil were identified. The Arabian Peninsula is a key region for understanding hominin dispersals and the effect of climate change on prehistoric demography, although little information on these topics is presently available owing to the poor preservation of archaeological sites in this desert environment. Here, we describe the discovery of three stratified and buried archaeological sites in the Nefud Desert, which includes the oldest dated occupation for the region. The stone tool assemblages are identified as a Middle Palaeolithic industry that includes Levallois manufacturing methods and the production of tools on flakes. The Kimberley region of northwest Australia contains one of the World's largest collections of rock art characterised by two distinct art forms; the fine featured anthropomorphic figures of the Gwion Gwion or Bradshaw paintings, and broad stroke Wandjina figures. Luminescence dating of mud wasp nests overlying Gwion Gwion paintings has confirmed an age of at least 17,000 yrs B.P. with the most recent dates for these paintings from around the mid-Holocene (5000 to 7000 yrs B.P.).
