Comparing Low Salinity Transcriptomic Profiles Among Hard Clam, Mercenaria Mercenaria, Lines, Leslie Speight Youtsey

The hard clam is an important ecological and economic resource along the U.S. Eastern Seaboard. In Virginia alone, farm gate sales were estimated at $38.8 million in 2018 and $57.8 million in 2021, making Virginia the largest producer of hard clams in the U.S. This industry is primarily limited to higher salinity habitats on the seaside of the Eastern Shore of Virginia and lower Chesapeake Bay. Although the hard clam can be found in lower salinity habitats, they do not grow or survive at rates that are practical for productive aquaculture. Acetylation Controls Thyroid Hormone Receptor Intracellular Localization And Intranuclear Mobility, Cyril S. Anyetei-Anum

Thyroid hormone receptor (TR) is responsible for mediating the expression of genes involved in growth, development, and homeostatic regulation, in response to thyroid hormone. There are two main subtypes of TR, TRα1 and TRβ1, that mediate these physiological processes. Dysregulation of TR mediated processes is a contributing factor in disease pathology. Our prior studies show that TR is shuttled rapidly between the nucleus and cytosol, while localizing primarily to the nucleus. TRα1 contains two nuclear localization signals (NLSs) that act together to confer strong nuclear localization, while TRβ1 only contains one NLS (NLS-1). Assessing genetic variation within and among native populations and hatchery stocks of Crassostrea ariakensis using microsatellite markers In response to the dramatic decline of eastern oyster (Crassostrea virginica) populations in the Chesapeake Bay, introduction of the non-native Asian oyster Crassostrea ariakensis has been proposed. Currently several hatchery stocks of C. ariakensis, derived from a few wild populations along the coast of Japan and China, are being maintained in U.S. hatcheries in the Pacific northwest and on the east coast. Atlantic menhaden is an ecologically and economically important species along the U.S. east coast. As a filter-feeder and key prey fish, it provides a critical link between primary production, phytoplankton, and larger piscivorous predators, such as striped bass, bluefish, and weakfish. The species is also the target of one of the largest commercial fisheries in the country. Menhaden are assessed as a single, coastwide stock, and recent assessments indicate that it is not overfished. Teleost immunoglobulin M (IgM), an 800 kDa tetramer, possesses considerable structural diversity due to the non-uniform disulfide polymerization of its halfmeric or monomeric subunits. However, to date, no plausible functional role for this diversity has been demonstrated or proposed. This research was, therefore, designed to investigate the possible functional role(s) for this diversity using the trout model. The possible relationship between this structural diversity and affinity was specifically addressed. The relationship between high levels of disulfide polymerization and high affinity was demonstrated by selective immunoadsorption and analysis of antibodies isolated during the process of affinity maturation. In mammals, the natural resistance-associated macrophage protein 1 gene, Nramp1, plays a major role in resistance to mycobacterial infections. Chesapeake Bay (USA) striped bass, Morone saxatilis, are currently experiencing an epizootic of mycobacteriosis that threatens the health of this ecologically and economically important species. This dissertation characterizes an Nramp gene in this species (MsNramp ) and provide evidence for induction following Mycobacterium exposure. The striped bass MsNramp gene and 554 amino acid sequence contain all the signal features of the Nramp family, including a topology of 12 transmembrane domains (TM), the transport protein specific 'binding-protein-dependent transport system inner membrane component …

Snapping shrimp in the family Alpheidae are remarkably abundant in coral reef ecosystems worldwide. The second most speciose genus, Synalpheus , includes species usually small and reclusive, and their general morphology is notably uniform, yet the number of species is staggeringly high, as is the intraspecific variability. These features have contributed to a poorly resolved taxonomic status, as evidenced both by recurring lumping and splitting of species and by lack of proper definition of several common species. The amphipods of Lake Baikal, Russia, are a taxonomically and morphologically diverse group, and have been cited as a classic example of adaptive radiation. An unfortunate consequence of this diversity has been an instability of Baikal amphipod classification, with frequent revisions spurred by differing opinions about the phylogenetic utility of different characters. Classifications have never been based on a rigorous phylogenetic analysis of morphological data and only recently has the phylogeny of Lake Baikal's amphipods been examined using molecular tools. The results of life history studies involving the sailfish, Istiophorus platypterus, from the Atlantic, Pacific, and Indian oceans were reviewed. Results of studies agree on several aspects of sailfish biology; sailfish exhibit opportunistic feeding behavior, follow the 28??C isotherm, spawn in summer mature at approximately age 3, and are the least migratory of the billfishes. However, results are in conflict concerning sex ratio and whether sailfish are single or multiple spawners. Although many researchers have reported that sailfish are sexually dimorphic, their fragile otoliths make them difficult to age, aging studies have not been validated, and sexual dimorphism has not …

Perkinsus marinus is the causative agent of the oyster disease Dermo in the eastern oyster, Crassostrea virginica. In vitro propagation of the parasite has led to the establishment of multiple isolates by several investigators. Little work, however, has been done to characterize different isolates. In this study multiple isolates were examined for genetic and biochemical diversity. at two loci, the ITS region and the ATAN region, there was as much intra-isolate genetic variation among DNA sequences of some isolates as there was inter-isolate variation. The study of antibody responses in prominent aquaculture species such as the rainbow trout, Oncorhynchus mykiss, can facilitate vaccine development and contribute to producing useful paradigms of adaptive immunity in lower vertebrates. Thus, it is essential to identify genes responsible for antibody responses. In the mouse model, hybridoma technology allows for the association of monoclonal antibodies possessing various affinities for antigen with specific VH sequences, gene family utilization, and other molecular events (i.e. somatic hypermutation) that occur during the specific immune response.
