Tattletales and T-Bow Update 20141018

http://works.bepress.com/gmcnamara/63

Tattletales is my concept to multiplex fluorescent protein biosensors ("Tattletales", which is also the overall concept name) and multicolor FP reporters ("T-Bow" for Rainbow T-cells and tumor cells) to study the physiology of live cells.

These are based on:

1. LacI-GFP :: LacO operator synthetic tandem repeat array (Robinett et al 1996 JCB, Fig 4A, http://jcb.rupress.org/content/135/6/1685.full.pdf )

2. the plethora of fluorescent protein biosensors - see page 1 of this "63" download site.

3. the many colors, and combinations of colors (CY11.5, CyPet-YPet 90% FRET, simple color combination fusions, Steven Vogel's "V6" and "V8" …

Mycoplasma Ovipneumoniae - A Primary Cause Of Severe Pneumonia Epizootics In The Norwegian Muskox (Ovibos Moschatus) Population, Torstein Tengs Dr.

Dr. Torstein Tengs

The Norwegian muskox (Ovibos moschatus) population lives on the high mountain plateau of Dovre and originates from animals introduced from Greenland. In the late summers of 2006 and 2012, severe outbreaks of pneumonia with mortality rates of 25-30% occurred. During the 2012 epidemic high quality samples from culled sick animals were obtained for microbiological and pathological examinations. High throughput sequencing (pyrosequencing) of pneumonic lung tissue revealed high concentrations of Mycoplasma ovipneumoniae in all six animals examined by this method and Pasteurella multocida subsp. multocida in four animals, whereas no virus sequences could be identified. Mycoplasma ovipneumoniae and P. multocida multocida …

Tattletales: multiplex fluorescent protein biosensors by spatial localization with TALE-FPs, Cas9-FPs, ZF-FPs, LacI-FPs, TetR-FPs, etc.

T-Bow: Rainbow T-cells and Tumor cells (and ES cells, iPS cells, other cells and organisms). You can think of this as "Brainbow meets TALENs/Cas9/ZFNs/other DNA sequence specific binding proteins".

If not familiar with Brainbow, see

Avian Influenza Virus Prevalence In Migratory Waterfowl In The United States, 2007-2009, Scott R. Groepper Mr., Thomas Deliberto, Mark Vrtiska, Kerri Pedersen, Seth Swafford, Scott Hyngstrom

Scott R Groepper

SUMMARY. We analyzed 155,535 samples collected for surveillance of avian influenza viruses (AIVs), in the United States from 2007 to 2009, from migratory waterfowl (ducks, geese, and swans). The goal was to elucidate patterns of prevalence by flyway and functional groups to determine targets for future surveillance. Apparent prevalence of AIV was highest in the Pacific Flyway in 2007–2008 (14.2% and 14.1%, respectively), in the Mississippi Flyway in 2009 (16.8%), and lowest each year in the Atlantic Flyway (range, 7.3%–8.9%). Dabbling ducks had higher apparent prevalence of AIV (12.8%–18.8%) than diving ducks (3.9%–6.0%) or geese and swans (3.6%–3.9%). We observed …

Changing Climate And The Altitudinal Range Of Avian Malaria In The Hawaiian Islands – An Ongoing Conservation Crisis On The Island Of Kaua`I, Carter T. Atkonson, Ruth B. Utzurrum, Dennis A. Lapointe, Richard J. Camp, Lisa H. Crampton, Jeffrey T. Foster, Thomas W. Giambelluca

Richard J. Camp

Transmission of avian malaria in the Hawaiian Islands varies across altitudinal gradients and is greatest at elevations below 1500 m where both temperature and moisture are favorable for the sole mosquito vector, Culex quinquefasciatus, and extrinsic sporogonic development of the parasite, Plasmodium relictum. Potential consequences of global warming on this system have been recognized for over a decade with concerns that increases in mean temperatures could lead to expansion of malaria into habitats where cool temperatures currently limit transmission to highly susceptible endemic forest birds. Recent declines in two endangered species on the island of Kaua’i, the ‘Akikiki (Oreomystis bairdi) …

Tattletales is my concept for spatial multiplexing many fluorescent protein (FP) biosensors in the same live cell. For example, there are excellent FP biosensors to Ca++ ions, pH, glucose, ribose, glutamine, glutamate, ATP, redox, ROS, pyruvate, cAMP, cGMP, IP3, PI(3,4,5)P3, cell cycle indicators (Fucci2), PKA, PKC, photsphatases, caspase(s) [1, 2]. However, these are typically used one biosensor per experiment, due in part to flooding …

The Hudson’S Bay Company Brigades Of 1832-33 And The Malaria Epidemic In California, David A. Bainbridge

David A Bainbridge

The ecological and cultural impacts of the Hudson’s Bay Company fur brigades to California were long term and important, but the expedition of 1832-33 caused a catastrophe by introducing the intermittent fever. The “intermittent fever” led to mortality rates from 50-90 percent or more, and it is likely more than 30,000 people died from the fever in the affected areas of California.

West Nile Virus West Nile Virus Infection In American Robins: New Insights On Dose Response., Larry Clark Jan 2013

West Nile Virus West Nile Virus Infection In American Robins: New Insights On Dose Response., Larry Clark

Larry Clark

West Nile virus (WNV) is a vector-borne pathogen that was first detected in the United States in 1999. The natural transmission cycle of WNV involves mosquito vectors and avian hosts, which vary in their competency to transmit the virus. American robins are an abundant backyard species in the United States and appear to have an important role in the amplification and dissemination of WNV. In this study we examine the response of American robins to infection with various WNV doses within the range of those administered by some natural mosquito vectors. Thirty American robins were assigned a WNV dosage treatment …

George McNamara

Cosmic ray particles image series acquired using a Hamamatsu ORCA-II ERG scientific grade CCD camera, cooled to -60 C. Each image is a consecutive 600 second (10 minute) exposure time with no light to the camera.

While processing the data, I discoverd that the background changed around planes 25 and 227 (see Excel file and jpeg screenshots), so I also processed only planes 025-227 (203 planes total, 2030 minutes, 33.83 hours). the CCD industry "rule of thumb" for a "typical" CCD sensor (i.e. 1/3" CCD) is that one cosmic ray particle strikes a sensor approximately every 30 seconds (assuming not …

The Internet is enabling greater access to spectral imaging publications, spectral graphs, and data than that was available a generation ago. The spectral imaging systems discussed in this issue of Cytometry work because reagent and hardware spectra are reproducible, reusable, and provide input to spectral unmixing and spectral components recognition algorithms. These spectra need to be readily available in order to determine what to purchase, how to use it, and what the output means. We refer to several commercially sponsored and academic spectral web sites and discuss our spectral graphing and data sites. Sites include fluorescent dye graph servers from …
