Pressurized Hot Water: An Alternative Method Of Nutrient Extraction And Subsequent Analysis For Use In Small-Scale Agriculture, Kristy Susanne Crane Jul 2004

Pressurized Hot Water: An Alternative Method Of Nutrient Extraction And Subsequent Analysis For Use In Small-Scale Agriculture, Kristy Susanne Crane

Theses and Dissertations

Soil analysis for small acreage farms in developing countries is often inconvenient and prohibitively expensive using current procedures, yet the information gained from these soil tests could have economical and environmental benefits. The pressurized hot-water (PHW) extraction coupled with colorimetric or turbidimetric analyses shows promise as an alternative to current procedures. Accepted methods of colorimetric analysis for NO3-N and P exist but an alternative method to atomic absorption spectrometry for K analysis is needed. Of the many possible methods to quantify K, tests performed in the Brigham Young University (BYU) laboratory found sodium tetraphenylborate to be unsatisfactory and sodium cobaltinitrite …

The Application Of Pedology, Stable Carbon Isotope Analyses And Geographic Information Systems To Ancient Soil Resource Investigations At Piedras Negras, Guatemala, Kristofer Dee Johnson

Theses and Dissertations

The ancient inhabitants of the Maya Lowlands enjoyed a long and fruitful period of growth which climaxed at around AD 800. At that time, millions of people successfully subsisted in a challenging environment that today only supports a population a fraction of that size. These facts, and the subsequent "Maya Collapse", are the impetus of many recent studies that utilize environmental data, in addition to conventional archaeology, to investigate this Maya mystery. Pedological studies and stable carbon isotope analysis of soil organic matter, combined with Geographic Information Systems (GIS) are three tools that can be used to answer crucial questions …

A Hydrothermal After-Ripening Time Model Of Seed Dormancy Loss In Bromus Tectorum, Necia Beck Bair Jul 2004

A Hydrothermal After-Ripening Time Model Of Seed Dormancy Loss In Bromus Tectorum, Necia Beck Bair

Theses and Dissertations

After-ripening, the process of seed dormancy loss in dry storage is associated with a decrease in the mean base water potential, one of the parameters of hydrothermal time. The rate of change of the mean base water potential is assumed to be a linear function of temperature above a specific base temperature and as a result can be described by a thermal after-ripening (TAR) time model, an extension of hydrothermal modelling. The thermal requirement for after-ripening is the thermal time necessary for the modelling base water potential of the seed to shift from its original value to its final value. …

Pressurized hot water and DTPA-Sorbitol are two relatively new soil boron (B) extraction methods with potential to replace the cumbersome hot water extraction. The objective of this research is to produce data in support of acceptance or rejection of these two alternative B extractions. The three soil tests were used to extract B from samples of calcareous sand and silt loam and limed, loamy fine sand treated with 10 levels of B and incubated for 7 and 28 d. As B application increased so did extractable B with each extraction method. High correlations (r of 0.977 to 0.999) were observed …

Mechanism Governing The Cellular Susceptibility To Secretory Phospholipase A2, Lauren Blackburn Jensen Jun 2004

Mechanism Governing The Cellular Susceptibility To Secretory Phospholipase A2, Lauren Blackburn Jensen

Theses and Dissertations

Secretory phospholipase A2 (sPLA2) is an important part of apoptosis and disposal of damaged and dying cells. However, healthy cells are not susceptible to attack by sPLA2. Recent studies have focused on membrane properties necessary to induce susceptibility in both artificial and biological membranes. Hydrolysis of phospholipids by sPLA2 requires at least two preliminary steps: first, adsorption of the enzyme to the cellular membrane, and second, movement of a phospholipid into the active site of the enzyme. We determined the effects of susceptibility on each of the two steps and determined the contributions changing the equilibrium constants have on susceptibility. …

The Impact Of Species Concept On Biodiversity Studies, Keith A. Crandall, Paul-Michael Agapow, Olaf R. P. Bininda-Emonds, John L. Gittleman, Georgina M. Mace, Jonathon C. Marshall, Andy Purvis

Faculty Publications

Species are defined using a variety of different operational techniques. While discussion of the various methodologies has previously been restricted mostly to taxonomists, the demarcation of species is also crucial for conservation biology. Unfortunately, different methods of diagnosing species can arrive at different entities. Most prominently, it is widely thought that use of a phylogenetic species concept may lead to recognition of a far greater number of much less inclusive units. As a result, studies of the same group of organisms can produce not only different species identities but also different species range and number of individuals. To assess the …

Velocity And Temperature Characterization Of The First Vacuum Stage Expansion In An Inductively Coupled Plasma - Mass Spectrometer, William Neil Radicic May 2004

Velocity And Temperature Characterization Of The First Vacuum Stage Expansion In An Inductively Coupled Plasma - Mass Spectrometer, William Neil Radicic

Theses and Dissertations

The inductively coupled plasma - mass spectrometer (ICP-MS) is the analytical instrument of choice for trace element detection and quantification. Despite the popularity of ICP-MS, significant degradation in sensitivity and precision occurs as the result of matrix and instrument-induced effects. The sources of these effects are not well understood, characterized, or correlated to particular plasma operating condition settings or matrix compositions and involve both neutral and charged species. The purpose of this study is to characterize the behavior of metastable Ar (I) atom and Ca (II) ion through the measurement of Doppler velocities and fluorescence line width "temperatures."

For the …

N9 Alkylation And Glycosylation Of Purines; A Practical Synthesis Of 2-Chloro-2'-Deoxyadenosine, Minghong Zhong May 2004

N9 Alkylation And Glycosylation Of Purines; A Practical Synthesis Of 2-Chloro-2'-Deoxyadenosine, Minghong Zhong

Theses and Dissertations

(a) The Robins reagent [2-acetamido-6-O-(diphenylcarbamoyl)purine] was utilized for glycosylation under Lewis acid conditions. Regioselectivity of glycosylation depends on the glycosyl donor and its 2-O- or 2-N-protecting group. Regioselective N9 glycosylation of 2-acetamido-6-O-(diphenylcarbamoyl)purine with problematic glucosamine has been accomplished by protecting the amino function as a phthalimido group with consequent stabilization of the oxocarbenium cation, and lowering the activation energy by introduction of trichloroacetimidate at the anomeric carbon.

(b) 6-Heteroaryl functions [6-(1,2,4-triazol-4-yl) and 6-(imidazol-1-yl)] were introduced into purine derivatives for regioselective N9 alkylation. The regiospecificity of alkylation mainly results from steric effects due to the coplanar conformation of the two linked …

Faculty Publications

Invariant natural killer T (iNKT) cells are an important source of both T helper type 1 (Th1) and Th2 cytokines, through which they can exert beneficial, as well as deleterious, effects in a variety of inflammatory diseases. This functional heterogeneity raises the question of how far phenotypically distinct subpopulations are responsible for such contrasting activities. In this study, we identify a particular set of iNKT cells that lack the NK1.1 marker (NK1.1neg) and secrete high amounts of interleukin (IL)-17 and low levels of interferon (IFN)-{gamma} and IL-4. NK1.1neg iNKT cells produce IL-17 upon synthetic ({alpha}-galactosylceramide [{alpha}-GalCer] or PBS-57), as well …

Fluorescence Detectors For Proteins And Toxic Heavy Metals, Uchenna Prince Paul Apr 2004

Fluorescence Detectors For Proteins And Toxic Heavy Metals, Uchenna Prince Paul

Theses and Dissertations

An inexpensive detector for proteins is described. The detection technique was based on two-photon excitation intrinsic protein fluorescence using a visible 532 nm diode-pumped nano laser as the excitation source. Proteins that exhibit intrinsic fluorescence must contain at least one tryptophan, tyrosine, or phenylalanine residue in their amino acid sequences. The detector was characterized and was found to have a detection limit of 4 micro-molar for tryptophan, 22 micro-molar for tyrosine and 500 micro-molar for phenylalanine. Bovine serum albumin, a serum protein with 3 tryptophan residues in its amino acid sequence was also used to characterize the detector. It was …

Analysis Of Clinically Important Compounds Using Electrophoretic Separation Techniques Coupled To Time-Of-Flight Mass Spectrometry, Zlatuse Durda Peterson Apr 2004

Analysis Of Clinically Important Compounds Using Electrophoretic Separation Techniques Coupled To Time-Of-Flight Mass Spectrometry, Zlatuse Durda Peterson

Theses and Dissertations

Capillary electrophoretic (CE) separations were successfully coupled to time-of-flight mass spectrometric (TOFMS) detection for the analysis of three families of biological compounds that act as mediators and/or indicators of disease, namely, catecholamines (dopamine, epinephrine, norepinephrine) and their O-methoxylated metabolites (3-methoxytyramine, norepinephrine, and normetanephrine), indolamines (serotonin, tryptophan, and 5-hydroxytryptophan), and angiotensin peptides. While electrophoretic separation techniques provided high separation efficiency, mass spectrometric detection afforded specificity unsurpassed by other types of detectors.

Both catecholamines and indolamines are present in body fluids at concentrations that make it possible for them to be determined by capillary zone electrophoresis coupled to TOFMS without employing any …

Phylogeny Of The Polyneopterous Insects With Emphasis On Plecoptera: Molecular And Morpological Evidence, Matthew Dana Terry

Theses and Dissertations

Polyneoptera is an assemblage of eleven insect orders comprising the "orthopteroid" insects. It includes familiar insects such as grasshoppers, roaches, termites, earwigs and preying mantises; as well as the more obscure web-spinners, angel insects and ice-crawlers. We present a phylogenetic analysis of the polyneopteran orders based on 18S rDNA, 28S rDNA, Histone 3, and a coded morphology matrix for an extensive sampling of taxa. We investigate the use of congruence between separate datasets as an a priori measure of alignment quality. Our results support the paraphyly of Polyneoptera, the monophyly of Dictyoptera, sister taxon relationships between Embiidina + Phasmatodea and …

In chapter one, a molecular phylogenetic study, based on six …

Androgen Receptor Expression In The Rat Prostate Is Down-Regulated By Dietary Phytoestrogens, Edwin D. Lephart, Trent D. Lund, Daniel J. Munson, Herman Aldercreutz, Robert J. Handa Jan 2004

Androgen Receptor Expression In The Rat Prostate Is Down-Regulated By Dietary Phytoestrogens, Edwin D. Lephart, Trent D. Lund, Daniel J. Munson, Herman Aldercreutz, Robert J. Handa

Faculty Publications

Background- It is well established that the growth of the prostate gland is a hormone-dependent phenomenon involving both androgenic and estrogenic control. Proliferation of prostate cells is, at least in part, under control of estrogen receptor beta (ER-beta). Phytoestrogens bind ER-beta with high affinity and therefore may have antiproliferative effects in the prostate. Methods- The prostates of male Long-Evans rats fed a diet high in phytoestrogens (Phyto-600) or very low levels of phytoestrogens (Phyto-free) were analyzed to determine the impact of dietary phytoestrogens on prostate weight and androgen receptor (AR) expression in the prostate. Results- Dietary phytoestrogens significantly decreased post-pubertal …

Multiple Domains Of The Co-Chaperone Hop Are Important For Hsp70 Binding, Gregory M. Nelson, Patricia E. Carrigan, Patricia J. Roberts, Jha'nae Stoffer, Daniel L. Riggs, David F. Smith Jan 2004

Multiple Domains Of The Co-Chaperone Hop Are Important For Hsp70 Binding, Gregory M. Nelson, Patricia E. Carrigan, Patricia J. Roberts, Jha'nae Stoffer, Daniel L. Riggs, David F. Smith

Faculty Publications

The Hop/Sti1 co-chaperone binds to both Hsp70 and Hsp90. Biochemical and co-crystallographic studies have suggested that the EEVD-containing C terminus of Hsp70 or Hsp90 binds specifically to one of the Hop tetratricopeptide repeat domains, TPR1 or TPR2a, respectively. Mutational analyses of Hsp70 and Hop were undertaken to better characterize interactions between the C terminus of Hsp70 and Hop domains. Surprisingly, truncation of EEVD plus as many as 34 additional amino acids from the Hsp70 C terminus did not reduce the ability of Hsp70 mutants to co-immunoprecipitate with Hop, although further truncation eliminated Hop binding. Hop point mutations targeting a carboxylate …

Botanical And Chemical Composition Of Diets Selected By Alpacas (Lama Pacos) Pastured In The Ulla Ulla Wetlands, Lino Constancio López López Jan 2004

Botanical And Chemical Composition Of Diets Selected By Alpacas (Lama Pacos) Pastured In The Ulla Ulla Wetlands, Lino Constancio López López

Theses and Dissertations

This study was done in the subtropical high Andes of Ulla Ulla. Its objectives were to determine the botanical composition, nutritional value, and preferred forages in the wetlands (hydromorphic and mesico) diet, by season (dry and wet), and by botanical family (12 families). Three-hundred sixty samples were collected from 6 alpacas with esophageal fistulations, and from them nutritional value and botanical composition were determined; the latter was compared with the botanical composition of the wetlands to determine preference. The results are highly significant (p<0.01) for botanical families, interaction between season and family, interaction of wetland type and family, and interaction of the season with wetland type and family. Species from the family Juncaceae were the most consumed with 27.78%, followed by moderate consumption of Cyperaceae (21.03%), Gramineae (15.02%), and Rosaceae (12.39%). Lower consumption occurred with Umbeliferaceae and Asteraceae with 6.61% and 5.64%, respectively, and the lowest consumption was of Plantaginaceae, Scrophulariaceae, minor plants, Gentianiaceae, unidentified plants, and Ranunculaceae with 2.86%, 2.50%, 2.17%, 2.08%, 1.03%, and 0.89%, respectively. The botanical families with the greatest preference value (PV) were Gramineae (PV=6.48), Scrophulariaceae (PV=8.75), Gentianiaceae (PV=7.55), and Ranunculaceae (PV=8.16). Proportional preference was observed in Juncaceae (PV=1.49), Cyperaceae (PV= -1.72), Rosaceae (PV=1.82), and Umbeliferaceae (PV= -0.82). The Asteraceae family was classified as a forced preference (PV= -6.29). The average nutritional value of the diet shows high values: 14.69% of raw protein, 14.58% ash, 85.42% organic matter, 21.43% raw fiber, and 31.00% neutral detergent fiber. It was concluded that alpacas efficiently select their diet for nutritional value, however, consumption is not appropriate, which results in negative live weight gain, especially in the dry season. On the other hand, the plants' preference values correspond to the lowest and one of the highest is a forced preference, according to the botanical composition of the wetland, which suggests that the preferred plants are repeatedly consumed in current continual pastures, and that alpacas are forced to consume unpalatable plants. This reflects on bad CANAPAS wetlands management in Ulla Ulla. Consequently, reversing this situation implies changes in pasture management and a reduction of the animal load.
