Trypanosoma brucei causes fatal human African trypanosomiasis and evades the host immune response by regularly switching its major surface antigen, VSG, which is expressed exclusively from subtelomeric loci. Telomere length and telomere proteins play important roles in regulating VSG silencing and switching. T. brucei telomerase plays a key role in maintaining telomere length, and T. brucei telomeres terminate in a single-stranded 3′ G-rich overhang. Understanding the detailed structure of the telomere G-overhang and its maintenance will contribute greatly to better understanding telomere maintenance mechanisms. Metabolomics can be described as a simultaneous and comprehensive analysis of small molecules in a biological sample. Recent technological and bioinformatics advances have facilitated large-scale metabolomic studies in many areas, including inborn errors of metabolism (IEMs). Despite significant improvements in the diagnosis and treatment of some IEMs, it is still challenging to understand how genetic variation affects disease progression and susceptibility. Food-borne diseases associated with fresh produce consistently cause serious public health issues. Although sanitization measures are utilized to enhance the safety of fresh produce, strategies that neglect the dynamic nature of commercial wash processes are limited, creating the potential for pathogen cross-contamination and major disease outbreaks. Abiraterone acetate (AA), the prodrug of abiraterone, is FDA-approved for the treatment of castration-resistant prostate cancer. Abiraterone is metabolized in patients to a more potent analogue, D4A. However, we have recently reported that this analogue is further metabolized to additional metabolites in patients treated with AA. Here, we present a liquid chromatography-tandem mass spectrometry method developed to resolve and detect abiraterone and its seven metabolites in human serum using an AB Sciex Qtrap 5500 mass analyzer coupled with a Shimadzu Nexera UPLC station. Translesion DNA synthesis (TLS) is the ability of DNA polymerases to incorporate nucleotides opposite and beyond damaged DNA. TLS activity is an important risk factor for the initiation and progression of genetic diseases such as cancer. In this study, we evaluate the ability of a high-fidelity DNA polymerase to perform TLS with 8-oxo-guanine (8-oxo-G), a highly pro-mutagenic DNA lesion formed by reactive oxygen species. Results of kinetic studies monitoring the incorporation of modified nucleotide analogs demonstrate that the binding affinity of the incoming dNTP is controlled by the overall hydrophobicity of the nucleobase. The National Bioengineered Food Disclosure Standard (the 'National Standard') was signed into law July 29, 2016. This Article analyzes the National Standard and posits that Vermont’s Act 120 was a more effective labeling law because it safeguarded consumer sovereignty. The State regulatory scheme in place prior to the passage of the National Standard satisfied consumer demand for disclosure while allowing for necessary experimentation with GMO labeling. Anti-cancer agents exert therapeutic effects by damaging DNA. Unfortunately, DNA polymerases can effectively replicate the formed DNA lesions to cause drug resistance and create more aggressive cancers. To understand this process at the cellular level, we developed an artificial nucleoside that visualizes the replication of damaged DNA to identify cells that acquire drug resistance through this mechanism. Visualization is achieved using "click" chemistry to covalently attach azide-containing fluorophores to the ethynyl group present on the nucleoside analog after its incorporation opposite damaged DNA. Trypanosoma brucei causes human African trypanosomiasis and regularly switches its major surface antigen, VSG, thereby evading the host's immune response. VSGs are monoallelically expressed from subtelomeric expression sites (ESs), and VSG switching exploits subtelomere plasticity. However, subtelomere integrity is essential for T. brucei viability. The telomeric transcript, TERRA, was detected in T. brucei previously. We now show that the active ES-adjacent telomere is transcribed. We find that TbRAP1, a telomere protein essential for VSG silencing, suppresses VSG gene conversionmediated switching. Importantly, TbRAP1 depletion increases the TERRA level, which appears to result from longer read-through into the telomere downstream of the …

One of the most heavily regulated aspects of the Health Care industry is the organ donation system (“Legislation and Policy”). Regulations in this area ensure the quality of the organs and morality of the process through which they were procured. This system, however, is failing in at least one sense; the number of patients requiring organ transplants is increasing, but the number of donors remains stagnant (Gordon, Patel, Sohn, Hippen, Sherman , 2014). Due to the lack of available transplant organs, a debate has been sparked on whether the United States government should allow for the purchasing of these organs.

Hexamethylene bis-acetamide inducible protein 1 (HEXIM1) is identified as a novel inhibitor of estrogen stimulated breast cell growth, and it suppresses estrogen receptor-a transcriptional activity. HEXIM1 protein level has been found to be downregulated by estrogens. Recently, HEXIM1 has been found to inhibit androgen receptor transcriptional activity as well. Researchers have used Hexamethylene bisacetamide (HMBA) for decades to stimulate HEXIM1 expression, which also inhibit estrogen stimulated breast cancer cell gene activation and androgen stimulated prostate cancer gene activation. African Clawed Toads are a model organism for research on sensory integration due to their lateral line system that allows them to sense water movement. This study further investigates the choices made by toads when presented with two lateral line stimuli in the form of surface waves. When such stimuli are initiated simultaneously, toads tend to turn towards the more rostral stimulus or the nearer stimulus, waves from which arrive first. Ranunculus ficaria L., an ephemeral perennial invasive plant brought over from Europe, is becoming widespread throughout the Northeastern United States. This herbaceous buttercup is able to create extensive dense mats that limit native species growth. Taking advantage of an early growing season and rapid reproduction rates, this species can create dense monocultures, which threatens native communities and ecosystems. Elimination of native spring ephemerals results in decreased biodiversity. A better understanding of how R. ficaria responds to site characteristics is needed to prioritize management efforts toward high-risk sites.

Despite an array of improved treatment options over the past decade, prostate cancer remains the second leading cause of cancer mortality for men in the United States. Abiraterone and galeterone are oral steroidal compounds that are used to treat metastatic castration-resistant prostate cancer (CRPC). Abiraterone blocks 17a-hydroxylase/17,20-lyase (CYP17A1), an enzyme required for androgen synthesis. Galeteron inhibits CYP17A1, blocks the androgen receptor (AR), and decreases AR protein levels. Both drugs share the same structure with endogenous androgens such as dehydroepiandrosterone, which are substrates for the enzyme, 3ß-hydroxysteroid dehydrogenase (3ßHSD). Insulin-like growth factor (IGF) signaling plays an important role in cell growth and proliferation and is implicated in regulation of cancer, metabolism, and aging. Here we report that IGF-1 level in blood and IGF-1 signaling demonstrates circadian rhythms. Circadian control occurs through cryptochromes (CRYs)—transcriptional repressors and components of the circadian clock. IGF-1 rhythms are disrupted in Cry-deficient mice, and IGF-1 level is reduced by 80% in these mice, which leads to reduced IGF signaling. In agreement, Cry-deficient mice have reduced body (~30% reduction) and organ size. Down-regulation of IGF-1 upon Cry deficiency correlates with reduced Igf-1 mRNA expression in the …

Disturbance regimes, i.e. frequent fires, historically maintained oak barrens until European settlement patterns, and eventually, Smoky the Bear and the fire suppression campaign of the U.S. Forest Service snuffed out the periodic flames. In the absence of a disturbance regime, ground layer floral composition at many historical oak sand barrens will change predominantly because of a buildup of leaf litter and shading of the soils. Termed mesophication, this process of ecological succession will drive Black Oak Sand Barrens to an alternate steady state. The rhythms in the expression of circadian clock genes are affected by calorie restriction (CR), a dietary paradigm known to increase lifespan. In our current study, we show that circadian rhythms are influenced by sex and the effects of CR are different between males and females. In particular, we found a group of clock genes which showed a sex-dependent difference in expression, as well as in response to CR (Rev-Erb α, Ror γ and both Cryptochromes: Cry1 and Cry2 genes). The primary objective of a new survey of the Cuyahoga River was to assess species richness and population abundance of various mussel species in the family Unionidae throughout the Middle Cuyahoga River and upper parts of the Lower Cuyahoga River. Historically, few records existed for this river between Lake Rockwell and the Cuyahoga Valley National Park, a region in which four dams have been removed in the past 12 years. In this study we performed light, immunofluorescent and transmission electron microscopy of Colpodella trophozoites to characterize trophozoite morphology and protein distribution. The use of Giemsa staining and antibodies to distinguish Colpodella life cycle stages has not been performed previously. Rhoptry and β-tubulin antibodies were used in immunofluorescent assays (IFA) to identify protein localization and distribution in the trophozoite stage of Colpodella (ATCC 50594). We report novel data identifying “doughnut-shaped” vesicles in the cytoplasm and apical end of Colpodella trophozoites reactive with antibodies specific to Plasmodium merozoite rhoptry proteins. African clawed toads (Xenopus laevis) use their lateral line system to detect prey. The African clawed frog can determine the distance of the origin of a surface wave. This allows the frog to differentiate which stimulus is more important for prey capture. The African clawed toads were put into a glass basin which was filled with water. This study was undertaken to determine the changes in freshwater bivalve population since the survey conducted in 2001 in the northern region of the Rocky River, Ohio, USA. During our study, we discovered 69 live specimens and 58 shells most of which were found in two distinct locations near the top and bottom of the area surveyed. Our finds consisted of eight species in comparison to the nine that were seen in the same area of the 2001 study. Its vector, the tsetse fly, feeds on an infected host and passes the parasites into the bloodstream of other mammalian hosts. The unique challenge for elimination is its complex process of antigenic variation, where the parasite regularly switches its major surface antigen, VSG. Plasmodium falciparum, the causative agent of severe human malaria shares the presence of apical complex organelles with the free-living predatory alveolate, Colpodella sp. In this study we investigated morphological, biochemical and molecular characteristics of Colpodella sp. in a diprotist culture containing Bodo caudatus as prey. Colpodella attaches to its prey using the apical end. Attachment lasted for approximately 20 minutes while the cytoplasmic contents of the prey were aspirated into the posterior food vacuole of Colpodella in a process known as myzocytosis. We will use the roundworm Caenorhabditis elegans to study the process of meiosis, which is involved in the formation of gametes (sperm and eggs). Its small size, rapid life cycle, transparency and well-annotated genome allows researchers to track the effects of mutations that disrupt gametogenesis. Our preliminary data demonstrated that a genetic screen for mutations that increase the viability of embryos produced by worms lacking SPO-11, a critical factor regulating meiotic chromosomal inheritance, can identify genes required for the accurate transmission of the genome from one generation to the next. Programmed cell death (apoptosis) is induced by the same culture conditions as differentiation in skeletal myoblasts, yet these processes result in mutually exclusive physiological endpoints. Dissecting this coordinate regulation could enable selective manipulation relevant to the alleviation of muscle degeneration, the effectiveness of regeneration, or treatment utilizing skeletal myoblast transfer. Analysis of asynchronous cultures of myoblasts in growth media (GM) were determined to have 50% of cells in G1 phase, 30% of cells in S phase and 20% of cells in G2/M phase. As a recurring symptom of eutrophication in Lake Erie, massive blooms of harmful algae pose a threat to safe drinking water supply and recreational water use. Causes of the recent re-eutrophication in the lake include changes in the tributary phosphorus loading and/or increases in the internal nutrient loading potentially mediated by the colonization of zebra and quagga mussels. Dihydroorotase is the enzyme that catalyzes the third step of the de novo biosynthesis of pyrimidines. M. jannaschii is a hyperthermophillic archaeon that can serve as a model organism for research purposes. This experiment is a first step toward elucidating the structure of the dihydroorotase in M. jannaschii. The enzyme was purified by salting out and heating the solution and then putting the supernatant through cation exchange chromatography and hydrophobic interaction chromatography. Twenty-four conditions were tested to determine if a crystal of dihydroorotase could be formed. Two of these conditions led to preliminary crystal formation. These findings can be utilized …
