Soybeans are grown on approximately 1.3 million ha in Arkansas generating an estimated $1.7 billion annually. Foliar diseases on soybean can result in economic losses. Growers spend significant time and money on disease scouting via crop consultants and often (subsequent) fungicide applications. Fungicide trials are often arranged in small plots designs. In these scenarios, spatial variability of foliar disease is minimized. While it is advantageous to minimize variance outside of treatment differences, another limitation with many small plot trials is ample disease pressure or having only lower severity. This study examined the effects that water table depth and soil characteristics have on plant species richness and species composition within pitcher plant bogs across seasons. Eight piezometers were installed at random distances to monitor long-term water table depth and pressure fluctuations along a ~710-meter line transect traversing upland and bog habitats. Vegetation sampling quadrats (n=128) were set up near each piezometer. Cover data and water table depths were collected in spring and late summer. Wheat (Triticum aestivum L.) is a self-pollinating species that is most often bred as a recombinant inbred line. When two inbred individuals are crossed, they produce progeny which experience heterosis; heterosis is the increased robustness experienced due to the reintroduction of heterozygosity. Hybrid wheat may be produced through the use of chemical hybridization agents, yet the structure of wheat flowers decreases the efficiency of outcrossing. Crop improvement through biotechnology is an integrated effort, incorporating multiple approaches like integration of genes, editing of native genes, and removal of selection marker genes. Before streamlining the protocols, the efficiency and feasibility of the individual approach and their components must be tested. I investigated the lasting impacts of a management plan designed to improve oak regeneration and benefit wildlife in the Ozark Highlands in Madison, Co., AR. To assess the efficacy of the management plan, I used variables relevant to the success and establishment of oak trees. Controlled burns and selective logging were used to thin the canopy, increase ground level productivity, and increase the abundance of small mammals. I used measurements of overstory and understory densities, light availability, and the density of mice in the genus Peromyscus across time to look at the lasting impacts of management. Rising CO2 may warm northern latitudes up to 10 °C by the end of the century. However, responses of plant physiological processes (such as photosynthesis and respiration) and growth to climate change remain uncertain. Seedlings and mature trees of tamarack (a deciduous species) and black spruce (an evergreen species), North America dominant conifers, were exposed to combined warming (up to +9 ˚C) and elevated CO2 (up to +300 ppm). The Polycomb group (PcG) proteins form two protein complexes, Polycomb repressive complex 1 (PRC1) and PRC2, which are key epigenetic regulators in eukaryotes. PRC2 represses gene expression by catalyzing trimethylation of histone H3 lysine 27 (H3K27me3). In Arabidopsis thaliana, CURLY LEAF (CLF) and SWINGER (SWN) are two major H3K27 methyltransferases, playing essential roles in plant growth and development. Despite their importance, genome-wide occupancy profiles of CLF and SWN have not yet been determined and compared. Isoëtes (Isoëtaceae, Isoetales, Lycopodiophyta) is a cosmopolitan genus of aquatic lycophytes, occurring on every continent except Antarctica. Of approximately 200 total taxa, about half are in a clade of species mostly occurring in North and South America. Eastern North America accounts for 22% of global taxonomic diversity, containing 32 fertile taxa and 16 named hybrids. This taxonomic diversity is built upon relatively little morphological difference, and even combined with phylogenetic analysis using several nuclear and chloroplast DNA markers, no well-resolved systematic treatment within this clade exists.

For most of the recent evolutionary history of plants, atmospheric CO2 concentrations have been far below modern values. However, little is known about how plants grown in a low CO2 environment balance their carbon demand for growth while meeting their requirement for nitrogen acquisition, the establishment of mycorrhizal fungal associations, and the production of defense compounds. Here, I investigated how low CO2 affects Elymus canadensis and Picea mariana by comparing their growth at low and current CO2 concentrations. Phosphorus (P) is an essential plant macronutrient vital to fundamental metabolic processes. Plant-available P is low in most soils, making it a frequent limiter of growth. Declining P reserves for fertilizer production exasperates this agricultural challenge. Plants modulate complex responses to fluctuating P levels via global transcriptional regulatory networks. Although chromatin structure plays a substantial role in controlling gene expression, the chromatin-level mechanisms involved in regulating P homeostasis have not been determined.

Current evidence has revealed the involvement of epigenetic mechanisms, including histone deacetylases (HDACs), in plant stress responses. In Arabidopsis thaliana, HDA19, belonging to the RPD3/HDA1 class, interacts with transcription factors to form repressor complexes. HDAC research mainly exists for dicotyledons, whereas research on monocotyledons is limited. Brachypodium distachyon is used as a model plant to investigate questions unique to monocot crops. BdHD1 is the closest homologous gene to HDA19 in B. distachyon. This study investigated potential protein-protein interactions between BdHD1 and each of BdMYB22, BdWRKY24, and BdHOS15. Plant cell walls are complex structures that contain a matrix of cellulose, lignin and hemicellulose. The regulation of the biosynthesis of these components has been well-studied in the eudicot plant Arabidopsis thaliana, and a transcriptional network has been elucidated. Several NAC and MYB family transcription factors are key regulators of secondary cell wall biosynthesis, and their functional characterization provides significant insight into the complex underlying transcriptional network. Despite growing interest in flowering phenology among ecologists, as reflected by an increase in the number of papers, there is little information on how studies typically measure and describe a plant’s flowering phenology. The focus of this study was the literature on flowering phenology and the approaches researchers have taken to quantify flowering phenology. We explored the seasonal factors that might play a role in triggering germination of Dendromecon rigidaand Ehrendorferia chrysantha.D. rigida and E. chrysantha have been found difficult to germinate using common greenhouse techniques, Keeley and Fotheringham successfully germinated both species after storing their seeds in the field over a year and treating them with smoke. Identifying the specific seasonally changing factors that stimulated germination could have implications for understanding germination requirements for these and other hard-to-germinate chaparral species.

The in vitro propagation is a promising method for the production of plants in species that are endangered, commercially valuable, and/or otherwise difficult to propagate through traditional horticultural methods. Leaf and petiole explants of critically endangered African violets (Saintpaulia rupicola B.L. Gray leaf spot is a globally important foliar disease of maize caused by the fungus Cercospora zeae-maydis. In C. zeae-maydis, light is a critical environmental signal linked to pathogenesis and secondary metabolism. However, the mechanisms by which the fungus senses and responds to light are not fully understood. Thus, the overarching goal of this project was to unravel the connection between light and pathogenesis in C. zeae-maydis. The conserved fungal protein White Collar-1 (WC-1), designated Crp1 in C. zeae-maydis, senses light and initiates physiological responses. Prenylation has important roles in the cell: targeting and localizing proteins to subcellular compartments and promoting protein-protein interactions. The purpose of this study was to identify historical trends of neglect in plant biology education, their continuation today in Michigan middle school classrooms, and to uncover reasons for this neglect. A content analysis of state and national science standards and an online survey of Michigan middle school life science teachers were conducted. Findings showed a trend of increasing references in national standards from animals or plants to general organisms, as well as a decrease in the overall number of specific references in the standards. A field study was conducted in 2017 and 2018 at the LSU Agricultural Center H. Rouse Caffey Rice Research Station (RRS) near Crowley, LA. to evaluate the influence of different adjuvants in overcoming the antagonism of quizalofop when mixed with bispyribac in Louisiana rice production. The antagonism of quizalofop mixed with bispyribac plus HCOC at 14 DAIT was overcome with a neutral interaction observed at 28 DAIT for barnyardgrass control with an observed control of 91%, compared with an expected control of 97%. Background: Many plants must balance the need for pollination services with mediating the risk of pollinator-vectored pathogens. Vaccinium corymbosum, highbush blueberry, is negatively affected by an insect-vectored, fungal plant pathogen, Monilinia vaccinii-corymosi (MVC), the cause of mummy berry disease, in which the asexual spore mimics pollen grains and is transferred from blighted tissue to flowers via pollinators, resulting in inedible, hardened fruits. Highbush blueberry plants require outcrossed pollen for maximum yield and fecundity. Therefore, yield of blueberry plants rely on a balance between adequate pollination service and disease avoidance. Losses caused by pests remain an important limitation to achieving high rice yields in the United States. Arbuscular mycorrhizal fungi (AM fungi) are able to modify plant physiology by increasing plant growth or inducing defense responses against insect herbivores. However, studies of the role of AM fungi in agroecological factors, including natural occurrence, plant resistance, soil dependency, and plant tolerance, with specific regards to pests that feed on rice plants have not been conducted before. Soybean (Glycine max [L.] Merr.) is a globally cultivated crop that is important to the sustainability of many industries. However, like all plants, optimal cultivation of soybean is threatened by detrimental environmental factors. For example, high yield of soybean is threatened by soil-borne pathogens like Phytophthora sojae. Resistance against P. sojae was previously positively correlated with aliphatic suberin deposition in soybean. As such, a deeper understanding of the biosynthesis of suberin may assist in engineering a resistant form of soybean, based on enhanced suberin content. Medicago sativa is one of the most produced perennial forage crops in Canada. Drought stress is a major form of abiotic stress affecting the productivity and annual yield of alfalfa. A small noncoding RNA, miR156 has a role in abiotic stress tolerance, while regulating downstream SPL genes. In this study, whole leaf proteome and amino acid concentrations of miR156OE (A8) genotype of alfalfa under drought stress were quantified. In results, 3,000 proteins groups were identified using MaxQuant when searched against Medicago truncatula protein sequence database. Freshwater ecosystems are some of the most biologically diverse environments on Earth. Billions of humans rely on functioning freshwater ecosystems for drinking water and many other services. These ecosystems are increasingly threatened by human impacts including nutrient pollution, invasive species, and climate change. Here I contribute four research chapters that investigate freshwater diversity and ecosystem threats using the Characeae, a family of freshwater green macroalgae, as a study system. Short Tandem Target Mimic (STTM) is a commonly technique used for functional studies of a number of genes in several plant model systems. However, very little is known about application of STTM technique in tree species. In this study, STTM was applied to knock down microRNA396 (miR396) in transgenic poplar trees for the first time. STTM396 expression resulted in dramatic decrease in miR396 expression levels leading to taller plants with larger leaves and larger leaf cell size.
