Glyphosate resistance among weed species has been an increasing problem in crop production over the past two decades. Glyphosate resistant Erigeron canadensis (horseweed) has been found on multiple continents and 20 U.S. states. It appears that the resistance is of non-target type, with mechanisms of action that have yet to be elucidated at the gene level. For the first portion of the study, I focused on reproducing the original indirect organogenesis protocol but discovered biotype variability can influence regeneration success. When mentioning the word hemp, especially in the local language of Madagascar, the literal translation does not set it apart from marijuana, as they are both called “rongony” - creating the stigma around hemp as the negative stereotype of marijuana. However, the material has been used by the ancestors of Madagascar, as well as across cultures, in its fibrous form to produce fabrication like textile goods and packaging. Summers are expected to continue to increase in heat/dryness in the Northeast, causing issues pertaining to forage production during the summer to worsen. Many pastures grow cool season grasses, even during the summer. These grasses enter a dormant period and slowdown in production during the months of July and August, leading to what is referred to as “summer slump”. Some farms grow corn silage during the summer, and while corn silage is a valuable crop, its cultivation often does not support soil biology. Diterpene Synthases Of The Rice Blast Fungus: Phylogenetic Analysis And Biochemical Characterization, Ayousha Shahi

Plant-pathogenic fungi harbor various specialized metabolites including diterpenoids that function as hormones and virulence factors. The fungus Magnaporthe oryzae is the causal agent of rice blast disease and can infect over 50 grass species. We demonstrate that the rice blast fungus encodes two diterpene synthases that produce normal pimara-8,15-diene and manoyl oxide scaffolds. Phylogenetic analysis of diterpene synthases among rice blast pathotypes showed functional conservation of the two core diterpene synthases amongst all pathotypes and suggests further expansion in select grass species. These insights into the blast fungal terpenome may inform efforts to counteract deleterious phytopathogens in crucial food crops.

Clarireedia spp. (formerly Sclerotinia homoeocarpaF.T. Bennett) is the causal agent dollar spot, the most economically important turfgrass disease impacting golf courses in North America. The most effective strategy for dollar spot control is repeated application of multiple classes of fungicides. However, reliance on chemical application has led to resistance to four classes of fungicides as well as multidrug resistance (MDR). Fungi are known to detoxify xenobiotics, like fungicides, through transcriptional regulation of three detoxification phases: modification, conjugation and secretion. Molecular chaperones are proteins found in virtually every organism and are essential to cell survival. When plants are heat stressed, they upregulate and downregulate multiple genes, many of which are associated with the heat shock response. Small heat shock proteins (sHSPs) are one class of molecular chaperones that are upregulated during heat shock. They are proposed to act as the first line of defense by binding to heat sensitive proteins and preventing their irreversible aggregation. Individuals within a plant species can differ greatly from one another, especially regarding the range of chemical compounds produced. However, the functions of many of these chemicals are unknown, but likely include defenses against herbivores, attractants for pollinators and seed dispersers, as well as mechanisms for resource competition. Traits that increase mean nutrient retention times are essential to plant performance in low resource environments, where multiple stressors (low water and nutrients) are present. Although physiological responses to either water or nutrient stress are well understood, fewer studies have investigated the interaction of these stressors from a phylogenetically-controlled, whole-plant perspective. This research focused on three, phylogenetically-controlled pairs of shrubby evergreen species from the California chaparral that either grow on or off serpentine soils, which differ in nutrient availability. While switchgrass (Panicum virgatum (L.)) has long been recognized as a viable bioenergy feedstock, it and other plants have cell walls with recalcitrance to processing. Recalcitrance is recognized as a major barrier to broad adoption of switchgrass and other feedstocks for cellulosic bioenergy. In an effort to reduce recalcitrance, transgenic plants have been generated with altered cell wall phenotypes such as reduced lignin. The purpose of this research was to increase protein, methionine, and cysteine content in Phaseolus vulgaris L., common bean in relation to the inhibitory compound tannin. Previous research has shown that sulfur fertilization increases total protein, methionine and cysteine content in various crops, but always in tandem with inhibitory compound increases. Fleshy fruits in different species can display large variation in color. A link between fruit color and seed dispersal success is suggested by previous studies showing that high intensity of fruit color increases the visitation rate in seed-dispersing birds. Wild tomato species (Solanum spp.) are excellent model organisms for research on genetic basis of differential fruit color development during the ripening process. Protoplasts are appropriate targets for genome editing, DNA functional screens and transgenesis. This project focuses on the comparison of inexpensive mesophyll protoplast isolation via the use of food-grade enzymes and transformation between diploid Panicum hallii Vasey (PAH) and polyploid cellulosic feedstock Panicum virgatum L (switchgrass), a relative of PAH. PAH has great potential as a C4 model species for crop and bioenergy research. Here an inexpensive switchgrass and PAH mesophyll protoplast isolation and transformation system was developed; the first protoplast system for PAH. Iron is among the essential micronutrients for all living organisms and is a cofactor for many cellular redox reactions. Although iron is a highly abundant metal element found in the earth’s crust, it is also a limiting factor in plant development when it is present as insoluble ferric oxides. Plants have evolved two strategies to acquire soluble iron referred to as Strategy I and Strategy II. Our lab has focused on the Arabidopsis thaliana double mutant ysl1ysl3. Leaf nutrient concentrations are an example of a functional trait, a trait that can be used to understand community dynamics by observing how plants interact with their environment. I explored how leaf nutrient concentrations were affected over successional time by environmental changes. The majority of prior research on functional traits focuses on either woody or herbaceous species, but not both life forms simultaneously. To address this, I also explored the successional changes to leaf nutrient concentrations, separating the plant community into woody and herbaceous species.

Lucinid clams and their sulfur-oxidizing endosymbionts comprise two compartments of a three-stage, biogeochemical relationship among the clams, seagrasses, and microbial communities in marine sediments. A population of the lucinid clam, Stewartia floridana, was sampled from a subtidal seagrass bed at Bokeelia Island Seaport in Florida to test the hypotheses: (1) S. floridana, like other lucinids, are more abundant in seagrass beds than bare sediments; (2) S. floridana gill microbiomes are dominated by one bacterial operational taxonomic unit (OTU) at a sequence similarity threshold level of 97% (a common cutoff for species level taxonomy) from 16S rRNA genes; …

Plant invertases are a class of proteins that have enzymatic function in cleaving sucrose to fructose and glucose. Cell wall invertase, located on the exterior of the cell wall of plant cells, plays a key role in the unloading of sucrose from the apoplast to the sink tissues. Cell wall invertase interacts with an inhibitor, cell wall invertase inhibitor, post-transcriptionally to regulate its activity. The inhibitor is constitutively expressed in pollen development, early developing seeds, and senescing leaves: indicative of sucrose allocation being a limiting factor at these stages of development. The ATP-binding cassette (ABC) transporter gene superfamily is ubiquitous among extant organisms. ABC transporters act to transport compounds across cellular membranes and are involved in a diverse range of biological processes and functions including cancer resistance in humans, drug resistance among vertebrates, and herbicide resistance in weeds. This superfamily of genes appears to be larger and more diverse in the plant kingdom—yet, we know relatively less about ABC transporter function in plants compared with mammals and bacteria. Eastern hemlock hosts the hemlock woolly adelgid, an introduced sap-feeding insect that causes rapid deterioration of the host. Like most conifers, eastern hemlock produces a variety of constitutive and induced defenses, primarily terpenoids. To explore the relationship of terpenoid defenses with adelgid infestations, we artificially infested hemlocks at a forest site and a plantation site, and compared their terpenoid concentrations to those in control trees. Infested trees showed lower terpenoid concentrations than control trees, suggesting that eastern hemlock not only fails to induce production of terpenoids in response to adelgid infestation, but becomes less able to produce carbon-based defenses due …

The work presented here focuses on the molecular mechanism of phytosiderophore secretion in graminaceous plants. In maize, yellow stripe3 (ys3) is a mutant that is deficient in its ability to secrete iron-chelating compounds of the mugineic acid family known as phytosiderophores. Phytosiderophores are specific to grasses and are used for the acquisition of iron. Genetic linkage mapping of the ys3 locus lead to a region of interest on chromosome 3 defined by marker UMC1773. Pityopsis ruthii (Small) Small, also known as Ruth’s golden aster, is a federally endangered herbaceous perennial, endemic to two river systems, the Hiwassee and the Ocoee, within the Cherokee National Forest, Polk County, Tennessee. There are approximately 13,000 individuals that may be at high risk of short-term extirpation (Thompson and Schwartz, 2006). Little is known of the basic reproduction and life history of P. ruthii. Several experiments were conducted to further develop capacitance sensor-based automated irrigation systems. The first experiment tested whether the photosynthetic response to decreasing volumetric water content (VWC) differed among four species tested. A sigmoidal curve best described the relationship for all species (r2 [r-squared]>0.86). The VWC that maintained maximum photosynthesis at 90% was selected as a potential conservative irrigation set point and values were not different between species, nor were 100% container capacity values.
