Bacteriophage Populations In Wastewater Effluent, Guy William Lawrence

Effluent samples were taken at various stages of sewage treatment at a local wastewater treatment plant. Primary, secondary, tertiary and post-chlorinated effluents were sampled. The agar overlay method was used to determine the numbers of plaque-forming bacteriophage particles on thirty-six species of enteric bacteria from the Eastern Illinois University stock culture collection. Twenty-three pure culture phage isolates were prepared from selected plaques and their host range specificity was determined. For every host organism tested, a fifty percent or greater reduction of bacteriophage was evident through the sewage treatment process. Static renewal bioassays were performed with four polychlorinated biphenyls, including Capacitor 21 and Aroclors 1016, 1242, and 1254. Each compound was used to treat embryo-larval stages of the rainbow trout (Salmo gairdneri), channel catfish (Ictalurus punctatus), goldfish (Carassius auratus), redear sunfish (Lepomis microlophus), leopard frog (Rana pipiens), Fowler's toad (Bufo fowleri), and American toad (Bufo americanus). Developmental stages of the trout were the most sensitive, with LC50 's at 4 days posthatching of 0.3, 1.0, 1.1, and 1.6 μg/l for Aroclors 1254, 1242, 1016, and …

A protein similar to that previously demonstrated on poliovirus RNA and replicative intermediate RNA (VPg) was found on all sizes of nascent viral RNA molecules and on the polyuridylic acid isolated from negative-strand RNA. 32P-labeled nascent chains were released from their template RNA and fractionated by exclusion chromatography on agarose. Fingerprint analysis using two-dimensional polyacrylamide gels of RNase T1 oligonucleotides derived from nascent chains of different lengths showed that a size fractionation of nascent chains was achieved. The possible effects of ablastin, a reproduction-inhibiting antibody which is produced during the infection of the blood parasite, Trypanosoma musculi in the mouse, were investigated by fractionating the parasite into a supernatant fraction (designated as Group A test mice) and a pellet fraction (Group B test mice). These groups A and B mice were injected with the respective fractions, plus Freund’s Complete Adjuvant (FCA). Control mice were injected with FCA only. Human Herpes simplex virus type 2 (HSV 2)-transformed Balb/c mouse cells (H238 cells) are malignant in Balb/c mice. The immune stratus of mice bearing H238 cell-induced tumors was studied and changes in immune response with time and with altered diet of the mice were measured.

In the experiments dealing with the status of immunity with time after tumor cell injection, mice were fed Purina Laboratory Chow (P.L.C.), injected with H238 cells, and sacrificed three to eight weeks post-injection. Many new restriction-modification (R-M) system which are closely linked to serB and allelic to the hsdSB system of S. typimurium occur in different Salmonella serotypes. In PI transduction studies with these systems, recombinants were isolated which fell into one of three classes with reference to their specific restriction phenotype. Recombinants that restricted like the donor, recombinants that restricted like the recipient and recombinants that had lost the ability to restrict. This latter group having lost the recipient SB restriction without gaining the specific donor restriction and here called "zero recombinants,” were of special interest and were investigated.

Bvr-1, A Restriction Locus Of A Type C Rna Virus In The Feline Cellular Genome: Pleiotropic Restriction Of Endogenous Balb Virus In Cat X Mouse Somatic Cell Hybrids, Stephen J. O'Brien, Janice M. Simonson

Bvr-1 is a dominant X-linked feline gene which restricts the replication of B-tropic murineleukemia virus (B-MuLV) in somatic cell hybrids between murine BALB/c-RAG cells and FL-74 feline cells. Since the hybrids were originally derived by the hypoxanthine aminopterin thymidine selection scheme, counter selection experiments on 6-thioguanine result in preferential survival of hybrid cells which have spontaneously lost the feline X-chromosome on which is located the structural gene for hypoxanthine guanine phosphoribosyl transferase (IMP: pyrophosphate phosphoribosyl transferase, E.C. 2.4.2.8) and Bvr-1. Biological nitrogen fixation, a means of alleviating our increasing dependence on synthesized nitrogen fertilizers, is being studied with renewed interest. Production of nitrogen fertilizer requires large amounts of natural gas and other non-renewable energy sources, while the energy for biological nitrogen fixation is derived either directly or indirectly from photosynthesis. With only limited supplies of natural gas and an increasing population, and thus increasing demand for food, the advantages and even necessity of understanding and enhancing biological nitrogen is obvious. Colibacillosis (CB) in livestock is an economically important disease caused by the bacterium Escherichia coli. It is estimated that nearly one third of all baby pigs born in South Dakota die prior to weaning, and one fourth of these deaths is believed to be caused by CB (62). Similarly, it has been stated that 6 to 20% of calves born alive die from this disease (3). The intestinal bacterial flora of an animal has a considerable influence on the physiology of that animal. Some bacteria of the intestine are pathogenic and can initiate infections if they gain access to a susceptible individual. Some of the intestinal bacteria can produce potent carcinogens in vitro and can presumably do this also in vivo. Food passing through the alimentary tract of an animal is also chemically modified through the enzymatic action of the bacterial flora. The survival of fecal coliforms, fecal streptococci, and heterotrophs after exposure to chlorine were compared in receiving waters of the Mattoon and Charleston, Illinois waste treatment facilities employing membrane filter techniques and Standard Plate Counts. Water samples were taken from receiving waters upstream from the treatment plant, from secondary effluent and chlorinated secondary effluent within the plant, and from downstream receiving waters. Stream flow contribution and plant effluent contribution required the calculation of dilution factors to determine expected numbers of the various microflora found downstream assuming no residual chlorine effect. An Immunochemical Study Of The Combining Sites Of The Second Lectin Isolated From Bandeiraea Simplicifolia (Bs Ii), Charles Wood, E. A. Kabat, S. Ebisu, Irwin J. Goldstein

Two lectins with different binding specificities have been isolated from extracts of seeds of Bandeiraea simplicifolia. The first, Bandeiraea lectin I [11] was specific for terminal non-reducing αDGalactosyl residues. It reacted with B substances from human ovarian cysts and with several galactomannans to form precipitin lines in agar gels. Polysaccharides with terminal αDGalactosyl residues, such as larch galactan, did not react. The lectin agglutinated B erythrocytes strongly but also reacted to a lower titre with A1 and very weakly with A2 erythrocytes [15, 28] indicating that terminal non-reducing αDGalNAc [24] can …

Bovine abortion continues to cause economic loss to farmers and ranchers. In South Dakota, monetary loss from bovine abortion has been estimated at 4 to 4.8 million dollars annually. (31) Brucellosis was once considered the major cause of bovine abortion. The lambdoid bacteriophages are ideal subjects for the study of populational interactions at the molecular level because they do not mutually exclude one another in mixed infection of the same host cell. Previous work done in this laboratory using the lambdoid phage pairs λ-434 hy mi and λ-80 have demonstrated a density-dependent reduction in progeny yield per infected cell (burst size) as a function of increasing multiplicity of infection (m.o.i.). The λ cI gene which codes for the repress or molecule has been demonstrated to be the cause of the interaction in the λ-434 hy mi phage pair. …

Administration of Corynebacterium parvum in combination with localized irradiation has been shown to be more efficient than radiation treatment alone in prolonging the survival time of methylethrane induced tumor in C3H mice. Fractionated low dose total body irradiation has been demonstrated to be a promising therapy for patients with chronic lymphocytic leukemia. The ampullariid Pomacea urceus uses its ctenidium and lung in respiration. The snails reach the surface with their siphon and ventilate their lung by withdrawing their head-foot in a pumping action. The lung gas also serves to change the buoyancy of the snails. After a ventilation a snail does not necessarily return to the same buoyancy level.
