The evolutionary genetics of haplodiploids and X-linked genes share many features and are different from diploid (autosomal) genes in many respects. For example, the conditions for a stable polymorphism, the amount of genetic load, and the effective population size are all expected to be quite different between haplodiploids or X-linked genes and diploids. From experimental data, the genetic load for X-linked genes is much less than autosomal genes and appears less for haplodiploids than for diploids. The observed amount of molecular variation for haplodiploids is much less than that for diploids, even more so than predicted from the differences in …

Ligation of B cell receptor (BCR) on BKS-2, an immature B cell lymphoma by anti-IgM antibodies (Ab) caused apoptosis. Here we report that signaling through B cell receptor in wild type BKS-2 cells down-regulated the expression of Egr-1, a zinc finger-containing transcription factor. A reduction in the level ofEgr-1 mRNA could be demonstrated as early as 30 min after the ligation of BCR on BKS-2 cells. Immunocytochemical and Western blot analysis revealed that the expression of EGR-1 protein was also inhibited by anti-IgM treatment. Antisense oligonucleotides to Egr-1 caused growth inhibition and apoptosis in BKS-2 cells, suggesting that …

The human hemizona has been demonstrated to select spermatozoa with good motility, normal morphology, and the capacity to undergo the zona-induced acrosomal reaction. The studies conducted here are directed at using the human hemizona to investigate two key questions: (1) whether human X-chromosome bearing spermatozoa (X-sperm) and Y-chromosome bearing spermatozoa (Y-sperm) differ in their functional survival time (assessed by their capacity to bind to the human zona pellucida) after prolonged in vitro incubation, and (2) whether hemizona-bound spermatozoa have a reduced aneuploidy rate compared to unbound (and therefore, unselected) spermatozoa?

Phyllostomid bats eat the fruits and disperse the seeds of a wide variety of neotropical plant species, particularly those typical of early successional stages (Fleming & Heithaus 1981, Heithaus 1982, Orozco-Segovia et al. 1985). In assessing the effects of frugivorous animals on plant populations and vice-versa, it is important to determine whether the animals are selective in the types of fruit they eat, or whether the proportional abundance of different fruit species in their diets simply reflects availability in the environment. The use of molecular techniques for the assessment of familial relationships among social species of mammals has become relatively commonplace. However, some species represent poor candidates for such studies due to naturally low levels of genetic diversity, leading to unacceptably large standard errors associated with estimates of relatedness. Here, we report on a preliminary study of genetic diversity within two populations of a social species of ground squirrel, Gunnison's prairie dog (Cynomys gunnisoni) using DNA fingerprinting. We observed low levels of diversity in the form of large mean coefficients of genetic similarity among individuals occupying the same population. …

The Legionnaire's disease bacterium, Legionella pneumophila, is a facultative intracellular pathogen which invades and replicates within two evolutionarily distant hosts, free-living protozoa and mammalian cells. Invasion and intracellular replication within protozoa are thought to be major factors in the transmission of Legionnaire's disease. Although attachment and invasion of human macrophages by L. pneumophila is mediated in part by the complement receptors CR1 and CR3, the protozoan receptor involved in bacterial attachment and invasion has not been identified. The coat proteins of the RNA phages MS2 and Qβ are structurally homologous, yet they specifically bind different RNA structures. In an effort to identify the basis of RNA binding specificity we sought to isolate mutants that convert MS2 coat protein to the RNA binding specificity of Qβ. A library of mutations was created which selectively substitutes amino acids within the RNA binding site. Genetic selection for the ability to repress translation from the Qβ translational operator led to the isolation of several MS2 mutants that acquired binding activity for Qβ RNA. Despite adequate hormonal stimulation, oocytes collected for the purpose of in vitro fertilization and embryo transfer display several levels of nuclear maturity. Preovulatory or mature oocytes, technically those that are Metaphase I or II, are inseminated shortly after aspiration and assessed for fertilization the following day. Prophase I oocytes, also called germinal vesicle-bearing or immature oocytes, require a 24-36 hour period in culture before being exposed to spermatozoa. During this time, the majority of Prophase I oocytes complete nuclear maturation in vitro, progressing from germinal vesicle breakdown through first polar body extrusion. Cancers of the oral cavity and pharynx have increased throughout the decades. Since oral cancer represents a small percentage of the total cancers in the United States, it is often overlooked. Though this is the case, the results of this cancer can be devastating. To address this issue, an Oral Cancer Screening was organized on August 14, 1996 at the West Michigan Cancer Center in Kalamazoo. The objective of this research was to determine the relationships between the morphological development and in situ ruminally degradable protein (RDP), ruminally undegradable protein (RUP), and microbial protein of two cool season grasses (intermediate wheatgrass and smooth bromegrass) and two warm season grasses (switchgrass and big bluestem). The initial growth of grass tillers grown near Mead, Nebraska was clipped at ground level six times during the 1992 growing season and morphologically classified. Mean stage was calculated. Forage was ground to pass a 2-mm screen and was incubated in ruminally fistulated steers for 16 h. The RUP was adjusted for microbial …

It is considered that the implementation of this policy will protect the natural environment and native crayfish species. The complex mechanisms of heredity are little appreciated by nonspecialists, in some measure, because of misunderstandings that are perpetuated when words used for technical terms have other, more widely understood, folk meanings. When a word has both technical and folk meanings, it is the responsibility of the specialist to avoid promoting confusion by either using extremely cautious and precise language when using the term or, in cases when confusion is inevitable, abandoning the term in favor of one without a widely understood folk meaning. Staphylococcus aureus chromosomal DNA from 125 methicillin-resistant and 10 methicillin-sensitive clinical isolates was digested with rare-cutting restriction endonucleases and subjected to pulsed-field gel electrophoresis (PFGE). Thirty-nine distinct genomic macrorestriction patterns (GPs) were identified using SmaI; 29 of these patterns are from methicillin-resistant S. aureus (MRSA).

A dendrogram showing percent similarity among the patterns was constructed which revealed the considerable genomic diversity of the collection even though the isolates had been obtained during outbreaks. The majority of the MRSA formed one broad group with two outbreak subclasses. However, 8 of the 29 MRSA GPs (28%) were diverse.

Tat and Rev, small proteins encoded by the Human Immunodeficiency Virus type 1 (HIV-1), regulate its pattern of gene expression within infected cells. Tat increases transcription from the integrated pro virus by approximately a thousand-fold; Rev effects a shift in the pattern of splicing of the viral mRNA. Both are likely to interact with cellular proteins in executing their respective functions. Tat has additional activities (including neurotoxicity and inhibition of lymphocyte activation) which may be extraneous to its viral functions.

Homology Among The Autosomal Chromosomes Of Boophilus Annulatus (Say) And B. Microplus (Canestrini), Bonnie S. Gunn

Autosomes of Boophilus annulatus and B. microplus were compared using C- and G-bands to establish the degree of homology. Karyotypes of both species consisted of 20 autosomes and an XX:XO sex determination system with the X being the largest chromosome. All chromosomes of B. annulatus were acrocentric with heterochromatin limited to centromeric regions. The B. microplus karyotype was acrocentric with one band of noncentromeric heterochromatin occurring in three chromosome pairs. Interspecific comparisons indicated seven pairs of G-band homologous autosomes and three G-band homologous pairs when added interstitial heterochromatin was considered. Previously, all of the major fruit fly, Drosophila melanogaster, chromosomes (I, II, and III) have been shown to be associated with geotaxis, but the Y chromosome has not. Using two methods (back-crossing and chromosome substitution), Y chromosomes from lines that have evolved stable, extreme expressions of geotaxis were placed into different genetic and cytoplasmic backgrounds to test the resulting males for geotaxis. The results of the back-crossing do not support the interpretation of Y-chromosome effects on geotaxis. These tests do not have sufficient statistical power, however, to detect small genetic effects.
