The self-assembly of charged macromolecules forms the basis of all life on earth. From the synthesis and replication of nucleic acids, to the association of DNA to chromatin, to the targeting of RNA to various cellular compartments, to the astonishingly consistent folding of proteins, all life depends on the physics of the organization and dynamics of charged polymers. In this dissertation, I address several of the newest challenges in the assembly of these types of materials. Hemp is considered as one of the sustainable agricultural fiber materials. Degumming or surface modification of hemp bast is needed to produce single fibers for ensuing textile and industrial applications. The traditional degumming process necessitates a high amount of alkali, which causes detrimental environmental pollution. This study offers a new method to degum hemp fibers with reduced use of harmful alkali and precious water resources. In this work, hemp bast fibers were degummed by using combined microwave energy and deep eutectic solvent (DES). Localized chemical delivery plays an essential role in the fundamental information transfers within biological systems. Thus, the ability to mimic the natural chemical signal modulation would provide significant contributions to understand the functional signaling pathway of biological cells and develop new prosthetic devices for neurological disorders. In this paper, we demonstrate a light-controlled hydrogel platform that can be used for localized chemical delivery in a high spatial resolution. By utilizing the photothermal behavior of graphene-hydrogel composites confined within micron-sized fluidic channels, patterned light illumination creates the parallel and independent actuation of chemical release in a group of fluidic ports. Gas-liquid-solid fluidized beds have been widely applied in wastewater treatment, however, the current method of wastewater process has several limitations. Hence, an improved method is in demand. A 3.5 height and 0.1534m inner diameter column was used to study the hydrodynamic characteristics of a bubble-induced three-phase inverse fluidized bed. Air, water and three types of low-density particles were employed as gas, liquid and solid phases.

The goal of this dissertation was to parse the roles of physical, mechanical and chemical cues in the phenotype plasticity of smooth muscle cells (SMCs) in atherosclerosis. We first developed and characterized a novel synthetic hydrogel with desirable traits for studying mechanotransduction in vitro. This hydrogel, PEG-PC, is a co-polymer of poly(ethylene glycol) and phosphorylcholine with an incredible range of Young’s moduli (~1 kPa - 9 MPa) that enables reproduction of nearly any tissue stiffness, exceptional optical and anti-fouling properties, and support for covalent attachment of extracellular matrix (ECM) proteins. Bone defects are a prevalent problem in orthopedics and dentistry. Calcium phosphate-based coatings and nanocomposites offer unique solutions towards producing scaffolds with suitable physical, mechanical and biological properties for bone regeneration.
We developed a novel method to synthesize hydroxyapatite (HA) particles with high aspect ratio using sol-gel chemistry and hydrothermal treatment. We obtained tunable pure-phase carbonated-HA in the form of micro/nanorods and nanowires (diameters 25-800 nm). To mimic the structure of bone, HA nanowires were homogenously mixed within poly(ε-caprolactone) (PCL) to produce nanocomposites with improved mechanical properties as determined by uniaxial tensile testing.
