Deep wounds in the gingiva caused bytrauma or surgery require a rapid and robust healing of connective tissues. Wepropose utilizing gas-brushed nanofibers coated with collagen and fibrin for that purpose. Our hypotheses are that protein-coated nanofibers will: (i) attract and mobilize cells in various spatial orientations, and (ii) regulate the expression levels ofspecific extracellular matrix (ECM)-associated proteins, determining the initial conformational nature ofdense and soft connective tissues. Gingival fibroblast monolayers and3D spheroids were cultured onECMsubstrate and covered with gas-blown poly-(DL-lactide-co-glycolide)(PLGA) nanofibers (uncoated/coated with collagen and fibrin). The field of tissue engineering focuses on delivering patient-derived stem cells to the body through the use of degradable biomaterials, such as hydrogels, which are infused into engineered scaffolds. Hydrogels act as templates to support and guide cells towards the regeneration of new tissue. In this study, we introduce a completely intraoperative procedure for obtaining a fibrin hydrogel for tissue engineering applications. In the past, fibrin hydrogel has been commonly formed by combining fibrinogen protein with animal-derived thrombin. Instead, we have developed an automated, and therefore reproducible, protocol to isolate and form fibrin hydrogel without the use of animal-derived thrombin. …

Specific objective: Guided tissue regeneration (GTR) aims to regenerate the lost attachment apparatus caused by periodontal disease through the use of a barrier membrane. For the GTR procedures to be successful, barrier membranes are required to be present at the surgical site for an extended period of time (weeks to months). Synthetic membranes have the advantage of prolonged presence in a wound site; however, they do not actively contribute to wound healing.
