Curcumin is a bright orange compound with myriad applications for human health and wellness. Curcumin occurs naturally in the plant Curcuma longa (commonly known as turmeric) but must be extracted from the roots in an environmentally unfriendly fashion to obtain commercially relevant amounts of the compound. In addition, extraction of curcumin from turmeric spice yields a mixture of various curcuminoids, presenting an issue for isolating it in its pure form and complicating its use in clinical settings.

A major economic obstacle in lignocellulosic ethanol production is the low sugar concentrations in the hydrolysate and subsequent fermentation to economically distillable ethanol concentrations. We have previously demonstrated a two-stage fermentation process that recycles xylose with xylose isomerase to increase ethanol productivity, where the low sugar concentrations in the hydrolysate limit the final ethanol concentrations. In this study, three approaches are combined to increase ethanol concentrations. First, the medium-additive requirements were investigated to reduce ethanol dilution. The majority of streams and rivers in the United States (U.S.) are ecologically impaired, or threatened by anthropogenic stressors. Recent reports have found atrazine in drinking water to be associated with increased birth defects and incidences of Non-Hodgkin’s Lymphoma, with higher levels of significance from exposure to both atrazine and nitrate-N. In contrast, recent illnesses from E. coli contaminating vegetables that originated from irrigation water has increased awareness of identifying sources of E. coli entering irrigation reservoirs.

Understanding the behaviors of bacteria near surfaces is crucial in many biological and ecological applications. This knowledge can be used to hinder undesired biofilm formation on medical instruments and wounds. On top of that, it could also provide further insights in biodegradation of dispersed oil. In this work, the behavior of Escherichia Coli near a surface was experimentally studied. We utilized an inverted microscope in the phase filed illumination mode and processed acquired images to track the motions of bacteria near surfaces with high accuracy and repeatability. The multidrug ATP-binding cassette (ABC) membrane transporters (efflux pumps) are found in both prokaryotes and eukaryotes and they can extrude diverse structurally unrelated substrates, such as antibiotics and chemotherapeutic agents out of the cells. The efflux pumps are responsible for multidrug resistance (MDR) and the failure of numerous treatments in infections and cancers. All ABC membrane transporters share a common modular topology containing two transmembrane domains (TMDs) and two nucleotide binding domains (NBDs). Fusion protein technologies can aid to improve solubility of recombinant protein from microorganisms and help recombinant protein purification. Elastin-like polypeptides (ELP) as a fusion tag can be utilized to facilitate the purification of recombinant proteins because ELP can provide its thermally responsive behavior to ELP tagged proteins. An ELP tag can be used as the purification carrier through inverse transition cycling (ITC), which is a simple and non chromatographic separation process. During infection or denitrification, bacteria encounter reactive nitrogen species. Although the molecular targets of and defensive response against nitric oxide (NO) in Escherichia coli are well studied, the response elements specific to S-nitrosothiols are less clear. Previously, we employed an integrated systems biology approach to unravel the E. coli NO-response network. Here we use a similar approach to confirm that S-nitrosoglutathione (GSNO) primarily impacts the metabolic and regulatory programs of E. coli in minimal medium by reaction with homocysteine and cysteine and subsequent disruption of the methionine biosynthesis pathway. Concentrations of human health-related microorganisms in runoff from agricultural plots (0.75m x 2m) treated with fresh and aged cattle manure, swine slurry and no manure (control) were determined. Three consecutive simulated rainfall events, producing 35 mm rainfall and separated by 24 h, were carried out for each plot. Fecal indicator (Escherichia coli, enterococci, Clostridium perfringens and coliphage) loads released in rainfall runoff from plots treated with fresh cattle manure, aged cattle manure and swine slurry treatments ranged from 5.52 x 105 to 4.36 x 109, 3.92 x 104 to 4.86 x 108, and 9.63 …
