Melanoma skin cancer is the abnormal growth of the melanocytes – the pigmented cells located in the epidermis. The current gold standard diagnostic technique for determining whether a lesion is cancerous involves subjectively examining suspicious lesions and performing an invasive biopsy to confirm melanoma. This method may neglect some lesions or cause scarring from biopsies that turn out to be benign. Thus, impedance-based detection using a multi-electrode device was investigated as a noninvasive technique to diagnose melanoma skin cancer. Peripheral Artery Disease is a very serious disease characterized by an arterial occlusion due to atherosclerotic plaques. In response to an arterial occlusion, arteriogenesis occurs, causing smooth muscle cells to transition from a contractile to synthetic state. Also following an arterial occlusion, functional impairment was seen in the collateral circuit. An immunofluorescence protocol was developed in order to assess the impact of collateral enlargement (arteriogenesis) on smooth muscle phenotype at various time points. Organophosphates (OP’s) have been used in chemical warfare for years and tend to lead to death due to an attack on the nervous system. This work focuses on modeling a demyelinated Hodgkin and Huxley (HH) neuron with Simulated Program with Integrated Circuit Emphasis (SPICE) platform. Demyelinating disorders affect over 350,000 people in the U.S and understanding the demyelination process at the cellular level is necessary to find safe ways to treat the diseases [9]. Utilizing a previous SPICE model of an electrically small cell neuron developed by Szlavik [32], an extended core conductor myelinated neuron was produced in this work. In some instances neuropathies can be diagnosed through a conduction velocity test. However, not all neuropathies can be classified using this method. Gaining an understanding of how the stimulus level varies for different fiber sizes at different fiber depths within a conductive medium will provide useful information for simulation studies.
Following a two-step approach using COMSOL and MATLAB, a simulation was implemented to investigate the stimulus necessary to activate different sized fibers at different depths. Sleep apnea is a condition where people pause while breathing in their sleep; this can be of great concern for infants and premature babies. Current monitoring systems either require physical attachment to a user or may be unreliable. Microfluidic devices are diagnostic tools that are small, portable, and inexpensive. This makes them ideally suited for resource-limited settings. To construct a 3D device, each layer of patterned paper is stacked using double-sided tape with holes corresponding to the pattern. It is necessary to have a paste to fill the holes in the tape and provide a medium for the fluid to wick through. This project encompasses the design and development of a pretreatment microfluidic device for samples of physiological conductivity, namely a saline solution. The conductivity was reduced through the combination of dilution and ion removal using electric fields to enable downstream electro kinetic manipulations. The two major parts of this project include (1) designing a pretreatment protocol to reduce the conductivity of the sample solution to an acceptable level and (2) designing /fabricating a microchip that will effectively allow aim (1) to be performed on chip.

Functional vasodilation in arterioles is impaired with chronic ischemia. We sought to examine the impact of chronic ischemia and age on skeletal muscle resistance artery function. To examine the impact of chronic ischemia, the femoral artery was resected from young (2-3mo) and adult (6-7mo) mice and the profunda femoris artery diameter was measured at rest and following gracilis muscle contraction 14 days later using intravital microscopy. Functional vasodilation was significantly impaired in ischemic mice (14.4±4.6% vs. 137.8±14.3%, p<0.0001 n=8) and non-ischemic adult mice (103.0±9.4% vs. 137.8±14.3%, p=0.05 n=10). In order to analyze the cellular mechanisms of the impairment, a protocol was developed to apply pharmacological agents to the experimental preparation while maintaining tissue homeostasis. Endothelial and smooth muscle dependent vasodilation were impaired with ischemia, 39.6 ± 13.6% vs. 80.5 ± 11.4% and 43.0 ± 11.7% vs. 85.1 ± 10.5%, respectively. From this data, it can be supported that smooth muscle dysfunction is the reason for the observed impairment in arterial vasodilation.

This study examines the nanoindentation (energy inclusive) properties of 0.2 mg/kg alendronate treated ribs at one and three years against a vehicle treated control in a fresh-frozen, non-cold-mounted, condition. This was to verify if the tissue-level properties for 0.2 ALN treated beagles would increase because of an increased level of mineralization despite a microdamage increase.

The kinetics of milk coagulation are complex and still not well understood. A deeper understanding of coagulation and the impact of the relevant factors would aid in both cheese manufacturing and also in determining the nutritional benefits of dairy products. A method using confocal microscopy was developed to follow the movement of milk fat globules and the formation of a milk protein network during the enzyme-induced coagulation of milk. Every 34 seconds, someone in the United States suffers from a heart attack. Most heart attacks are caused by atherosclerotic build up in the coronary arteries, occluding normal blood flow. Balloon angioplasty procedures in combination with a metal stent often result in successful restoration of normal blood flow. However, bare metal stents often lead to restenosis and other complications. Mammary epithelial cells are highly efficient secreting cells. With genetic engineering, the uses of these cells could be endless. Research is being conducted on these cells to determine their full potential to the biotech industry.
This paper investigates whether bovine epithelial mammary cells can survive in glutaraldehyde-treated gelatin tubes in an operating bioreactor. Many bioreactors were developed and tested to suit the needs of the cells. Procedures were created and carried out to ensure sterility of the bioreactors. Bovine mammary epithelial cells were implanted in the bioreactors and samples of their growth were taken over time.

In previous studies it was determined that the fiber diameter distribution in a peripheral nerve could be estimated by a simulation technique known as group delay. These results could be further improved using a combinatorial optimization algorithm called simulated annealing. This paper explores the structure and behavior of simulated annealing for the application of optimizing the group delay estimated fiber diameter distribution. Specifically, a set of parameters known as the cooling schedule is investigated to determine its effectiveness in the optimization process.
