After amputation, muscles in the residual limb are detached from their insertion points and no longer span the missing joints. Our objective was to quantify the effect of amputation-induced disuse on residual muscle structure, an indirect indicator of muscle force-generating capacity. One hind paw was surgically removed at the ankle joint of ten rabbits. At two weeks (n=5) and 4 weeks (n=5) post-amputation and for select muscles (gastrocnemius, soleus, tibialis cranialis, extensor digitorum, and flexor digitorum superficialis), we measured and computed several muscle structure properties. Surgical site infections and chronic wounds, especially those caused by antibiotic-resistant microorganisms, result in hospitalization and fatalities each year. Methods to prevent these infections, such as cleaning and preparing medical tools, have had minimal success in preventing infections. Further, antibiotic treatments have become less successful in treating infections and wounds because of antibiotic-resistant bacteria. Antimicrobial peptides (AMP) are a possible treatment solution for wound infections. Current orthopedic implants are overwhelmingly composed from metallic materials. These implants show superior mechanical properties, but this can additionally result in stress shielding due to a modulus mismatch between the bone tissue and implanted device. Polymeric implants reduce this stress shielding effect but have much lower mechanical properties, limiting their use. Polylactic acid (PLA) is a widely used biodegradable thermoplastic polymer, however, its use has been limited by the polymer’s mechanical properties and rapid loss of strength during degradation in vivo. Many are currently FDA approved as magnetic resonance imaging contrast agents and tracers for magnetic particle imaging applications. Magnetic properties of such materials are crucial to obtain good contrast and resolution. Laparoscopes are prone to fogging which can lead to a limited field of view during surgical procedures. Current methods of mitigating fogging issues are not efficient or can require costly modification to the laparoscope. Previous studies of diamond-like carbon (DLC) coatings found doping the films improved hydrophilic qualities, suggesting their possible use as antifogging coatings for laparoscopes. For this work, two series of DLC films, doped with either SiO and Al2O3 were investigated.

This thesis presents the use of diblock copolymers, poly(butadiene)-b-poly(ethylene oxide) (PBm PEOn) and poly(isoprene)-b-poly(ethylene oxide) (PImPEOn), as amphiphilic molecular building blocks for the formation of synthetic polymer bilayer membranes using the droplet interface bilayer (DIB) technique. The DIB technique makes use of the self-assembly of amphiphilic macromolecules along oil-water droplet interfaces that can then be physically connected for the construction of liquid supported macromolecular bilayers at the droplet interface. The objective of this project was to develop a wireless, noncontact monitoring system that measures multiple physiological parameters in human faces from a distance using a camera. Compared to traditional sensors, this monitoring system does not use wires or adhesives, providing a safer, more user-friendly application. This thesis describes the initial design process for an application of continuum robotics to endoscopic surgical procedures, specifically dissection of the colon. We first introduce the long-term vision for a benchtop dual-instrument endoscopic system with intuitive haptic controllers and then narrow our focus to the design and testing of the instrument manipulator itself, which must be actuated through the long, winding channel of a standard colonoscope.

Heat treating fluorochlorozirconate (FCZ) glasses nucleates nanocrystals in the glass matrix, resulting in a glass ceramic that has optical properties suitable for use as a medical imaging plate. As the temperature of heat treatment rises, the resulting FCZ glass-ceramic becomes increasingly more opaque as the size of the orthorhombic phase BaCl2[barium chloride] nanocrystals grow within the glass matrix. This opaqueness negatively affects imaging. The effect of adding Fe3+[iron] on the valence state of zirconium and overall glass quality was investigated.

Fe3O4 [Magnetite] nanoparticles have magnetism that differs greatly from their bulk counterparts. Whereas bulk Fe3O4 is a ferrimagnet, single-domain Fe3O4 nanoparticles have been found to be superparamagnetic. This allows for increased magnetization of the nanoparticles compared to the bulk when in a magnetic field. For most paramagnets, magnetization requires applied fields of a few Tesla at low temperatures. This is achievable through the application of superconducting magnets. Borate silica glass ceramics were produced for neutron scintillation. The Glass ceramics were doped with europium fluoride [EuF2] and cerium chloride [CeCl3]. Isotopic lithium fluoride [6LiF] and boron oxide [10B2O3] were used in most samples while non-isotopic lithium fluoride [LiF] and boron oxide [ B2O3] were used in the rest. When exposed to a neutron beam, samples doped with europium fluoride [EuF2] scintillated while samples doped with cerium chloride [CeCl3] did not. In a lab-on-a-chip (LOC) application, the measurement of small analog signals such as local temperature variation often involves detection of very low-level signals in a noisy micro-scale environment. This is true for other biomedical monitoring systems as well. These systems observe various physiological parameters or electrochemical reactions that need to be tracked electrically. Unfortunately, difficulty with stair climbing has been seen to exist, prolonging the challenges of TKR patents. X-rays create electron hole pairs within the plate which recombine upon stimulation by a laser, producing light which is read out and stored as a digital image. Processing parameters varied were primary air flow rate and collector distance. In total, twenty-one webs were produced and the physical properties of the webs were investigated including, mean fiber diameter and fiber diameter distribution, mean pore diameter and pore size distribution, web thickness, degree of crystallinity, tensile modulus and degradation rate. Crouch gait decreases walking efficiency due to the increased knee and hip flexion during the stance phase of gait. Crouch gait is generally considered to be disadvantageous for patients with cerebral palsy; however, a crouched posture may afford biomechanical advantages that lead some patients to adopt a crouch gait.

A novel far-field plasmonic resonance enhanced nanoparticle-seeded Particle Image Velocimetry (nPIV) has been demonstrated to measure the velocity in a micro channel. Chemically synthesized silver nanoparticles have been used to seed the flow. Eight naturally deposited beef cow manure patties were sampled during summer (July 19 to August 9, 2010), fall (October 26 to November 19, 2010), winter (January 14 to February 18, 2011), and spring (May 5-27, 2011) to determine whether hypothesized seasonal differences existed in the initial concentrations and decay rates of Escherichia coli (E. coli) and bovine Bacteroidetes (BoBac). Positron Emission Tomography (PET) is frequently used for medical imaging. Maturity and flexibility of PET as an imaging technique has expanded its utility beyond the medical domain. It can be used as a tool for fluid flow studies in opaque fluids and for flow within complex geometry where conventional optical flow measurement approaches fail. This study explores the capabilities of PET as flow measurement tool suited to validation of computational fluid dynamic (CFD) predictions.

Synthesis And Characterization Of Diamond-Like Carbon Thin Films For Biomedical Applications, Russell Lee Leonard

Diamond-like carbon (DLC) thin films were produced by pulsed laser deposition (PLD) on silicon, fused silica, and silicon nitride substrates. The films produced were either undoped, made using a pure graphite target, or doped, using multi-component targets made from a combination of graphite and silicon, silicon nitride, titanium dioxide, or silicon monoxide. These films were evaluated for their potential use in biomedical applications, including coatings for artificial joints, heart stents, and bronchoscopes. The films were characterized by Raman spectroscopy, atomic force microscopy, ball-on-flat tribometry, contact angle measurements, and spectrophotometry.
