Nanoparticles have been of interest to the pharmaceutical industry since the 1980s. The first FDA approved nanoparticle-based therapies included liposomal anesthesia agents. Since then, the amount of FDA-approved nanoparticle therapies remains low. This is because nanoparticle-patient interactions can be very complex and are not well understood. Complicating factors also include increasing obesity rates among the patient population and many small animal pre-clinical trials are completed with healthy, lean animals. The biochemical differences between lean and obese patients prevents early studies from accurately predicting nanoparticle clinical behaviors. Many nanoparticles fail in trails. Phosphorylated myo-inositol compounds including inositol phosphates (InsPs) as well as the phosphatidylinositol polyphosphate lipids (PIPns) are critical biomolecules that regulate many of the most important biological processes and pathways. They are aberrant in many disease states due to their regulatory function. The same is true of the phospholipid phosphatidylserine (PS) which can serve as a marker to begin apoptosis. Calcific Aortic Valve Disease (CAVD) is a major disorder in the developed countries among elderly. It is characterized by calcific deposition and stiffening of the aortic valve cusps. CAVD is a highly cell-mediated condition where valvular interstitial cells (VICs) become activated and differentiate into osteoblast-like cells. This is associated with upregulation of calcific markers like Alkaline phosphatase (ALP) and Runx2. ECM remodeling in another characteristic of stenotic aortic valves due to VIC activation. This dissertation combines self-assembly phenomena of amphiphilic molecules with soft materials to create and characterize mechanoelectrical transducers and sensors whose sensing elements are thin-film bioinspired membranes comprised of phospholipids or amphiphilic polymers. We show that the structures of these amphiphilic molecules tune the mechanical and electrical properties of these membranes. We show that these properties affect the mechanoelectrical sensing characteristic and range of operation of these membrane transducers. In the experiments, we construct and characterize a membrane-based hair cell embodiment that enables the membrane to be responsive to mechanical perturbations of the hair. Bacterial cellulose (BC) is a natural hydrogel made of nanofibers. This material has been used in commercial products, including wound dressings. BC can be modified and optimized for improved performance in multiple applications. This work will focus on producing and characterizing resorbable cellulose, a composite for bone applications, and a composite for a synthetic venous valve leaflet.

BC can be produced and modified to perform as a degradable tissue scaffold. This is achieved by an oxidation procedure after the initial production and purification of native BC. A material characterization of oxidized BC was performed to identify the changes in properties …
