Photoacoustic (PA) tomography (PAT) is a novel imaging modality that combines the fine lateral resolution from optical imaging and the deep penetration from ultrasonic imaging, and provides rich optical-absorption–based images. PAT has been widely used in extracting structural and functional information from both ex vivo tissue samples to in vivo animals and humans with different length scales by imaging various endogenous and exogenous contrasts at the ultraviolet to infrared spectrum. Tremor, one of the most disabling symptoms of Parkinson's disease (PD), significantly affects the quality of life of the individuals who suffer from it. These people live with difficulties with fine motor tasks, such as eating and writing, and suffer from social embarrassment. Traditional medicines are often ineffective, and surgery is highly invasive and risky. The emergence of wearable technology facilitates an externally worn mechatronic tremor suppression device as a potential alternative approach for tremor management. Predictive simulation based on dynamic optimization using musculoskeletal models is a powerful approach for studying biomechanics of human gait. Predictive simulation can be used for a variety of applications from designing assistive devices to testing theories of motor controls. However, one of the challenges in formulating the predictive dynamic optimization problem is that the cost function, which represents the underlying goal of the walking task (e.g., minimal energy consumption) is generally unknown and is assumed a priori. Radiation therapy is a main component of treatment for many lung cancer patients. However, the respiratory motion can cause inaccuracies in radiation delivery that can lead to treatment complications. In addition, the radiation-induced damage to healthy tissue limits the effectiveness of radiation treatment. Motion management methods have been developed to increase the accuracy of radiation delivery, and functional avoidance treatment planning has emerged to help reduce the chances of radiation-induced toxicity. Acute Myocardial Infarction (AMI) is the leading cause of worldwide death and disability, and approximately 720,000 Americans will experience an AMI in 2018. Studies have shown that rapid hypothermia therapy (<35°C) before reperfusion in patients with AMI can reduce infarct size by 37%. Localized therapeutic hypothermia has proven the potential to cool heart tissue rapidly following AMI, 3°C in 5 minutes. Using Materialise Mimics digital imaging software and the finite volume method we analyzed temperature distributions in six patient-specific left main coronary artery (LMCA) models. A mock circulatory loop was used to determine the exiting temperatures of a standard 7 Fr catheter to feed into our model with flow rates ranging from 29.2 ml/min to 68.85 ml/min. Our work showed that therapeutic hypothermia (TH) temperatures were evident at the outlets of three out of all six heart models, which varied in each left anterior descending (LAD) and left circumflex (LCX) artery depending on flowrate. Results of this study indicate that biovariability in patient-specific vascular structures significantly impacts therapeutic hypothermia (TH) treatment methods. These results indicate that further research is needed to examine more accurate physiological effects, such as pulsatile flow and vessel wall thickness. Future models will be used to provide insight to guide more efficient TH device designs and operation parameters to optimize patient outcomes following AMI.

The objective of this research is to design and manufacture a device that exhibits some of the bio-physiological signals relevant to fetal health monitoring. Currently, limited options exist for testing the performance of monitoring devices such as the tocodynamometer (TOCO) and electrocardiograph (ECG) that measure the bio-physiological signals of a woman and her fetus. Sensor designers need ways of generating and acquiring signals that do not carry the ethical burden of human testing. Epithelial cells form multi-layered tissue scaffolding that makes up every organ in the body. Along with epithelial cells, the basement membrane (BM) and connective tissue are composed of various proteins that sculpt the organs and protect them from foreign macromolecules. Epithelial cells respond to various cues, both chemical and mechanical, from their surrounding matrices to aid in maintenance and repair of these layers through degradation and deposition of extracellular matrix (ECM) proteins. In cancer progression, epithelial cells lose their normal function of supporting tissue structure and instead adopt more aggressive behaviors through an epithelial-to-mesenchymal transition (EMT) of their cellular traits. …

My research focuses on electrokinetic transport. Particularly, in this dissertation, we focus on fabrication and testing of micro electrodes with nanostructured surfaces to minimize the electrode polarization (EP) effects for biosensor applications. In the first study, electrochemical deposition of gold nanoparticles on to planar gold electrodes was used to generate rough surfaces. Dendritic nanostructures that reduced EP up to two orders of magnitude was obtained by optimizing the deposition conditions. These structures also enhanced dielectrophoresis (DEP) response of our bio-chips, making them usable in physiological buffers. Manipulating a single biomolecule requires sensitive approaches to avoid damage to the molecule. One of the important surgical tools in spinal surgery is the C-Arm X-ray System. The C-Arm is a large “C” shaped and manually maneuvered arm that provides surgeons and X-ray technicians the ability to take quick quality X-rays during surgery. Because of its five degrees of freedom, the C-Arm can be manually maneuvered around the patient to provide many angles and perspectives, ensuring surgical success.

In E. coli, fatty acid synthesis is catalyzed by the enzyme acetyl-CoA carboxylase (ACC), which converts acetyl-CoA into malonyl-CoA. Malonyl-CoA is a major building block for numerous of bioproducts. Multiple parameters regulate the homeostatic cellular concentration of malonyl-CoA, keeping it at a very low level. Understanding how these parameters affect the bacterial production of malonyl-CoA is fundamental to maximizing it and its bioproducts. This critical design report describes the product development of a prosthesis for use on sand. Quality of Life Plus (QL+), a national non-profit organization aimed to develop prostheses for veterans and people with disabilities, introduced this project and its accompanying challenger, Sgt. Brady, to Cal Poly’s Interdisciplinary Senior Project class in September 2018. After consulting with Sgt. Brady and QL+ and performing extensive research, the Sand Foot team defined customer requirements and engineering specifications to meet these requirements. The purpose of this project was to devise a solution for Jorge Segura, a US Marine who was wounded in Afghanistan and had to have his non-dominant arm amputated above the elbow. His current prosthetic attachments are not suited to cooking tasks, so he needed a way to cook more effectively with either prosthetic attachments or accessibility devices. Methods for prolonged compassionate care for persons with Profound Intellectual and Multiple Disabilities (PIMD) require a rotating cast of import people in the subjects life in order to facilitate interaction with the external environment. As subjects continue to age, dependency on these people increases with complexity of communications while the quality of communication decreases. It is theorized that a machine learning (ML) system could replicate the attuning process and replace these people to promote independence. Introduction: The mechanical characteristics of the plantar tissues during walking is not well understood as most of the current research focuses on testing specific plantar regions in cadavers or while the feet of the participants are raised. In this work, it is hypothesized that a viscoelastic geometric ellipsoid model used to assess multiple structures of the foot would be accurate and robust. Through a partnership with Indian non-profit Bhagwan Mahaveer Viklang Sahayata Samiti, we designed a functional, robust, and and low cost electrically powered prosthetic hand that communicates with unilateral, transradial, urban Indian amputees through a biointerface. The device uses compliant tendon actuation, a small linear servo, and a wearable garment outfitted with flex sensors to produce a device that, once placed inside a prosthetic glove, is anthropomorphic in both look and feel. The prosthesis was developed such that future groups can design for manufacturing and distribution in India.

This dissertation presents a model-based method for estimating arterial wall parameters from noninvasively measured arterial pulse signals via a microfluidic-based tactile sensor. The sensor entails a polydimethylsiloxane (PDMS) microstructure embedded with 5×1 transducer array built on Pyrex/Polyethylene terephthalate (PET) substrate. The arterial pulse causes a time-varying deflection on the top of the PDMS microstructure, which registers as a resistance change by the transducer at the site of the artery.

This work presents a novel counter-flow design for thermal stabilization of microfluidic thermal reactors. In these reactors, precise control of temperature of the liquid sample is achieved by moving the liquid sample through the thermal zones established ideally through the conduction in the solid material of the device. The goal here is to establish a linear thermal distribution when there is no flow and to minimize the temperature change at flow condition. The Surf Leg Prosthetic project team in conjunction with QL+ and Operation Surf designed a transtibial prosthetic to be used for surfing. The senior project team focused on the missions of QL+ and Operation Surf as they went through the design process during the 2018-2019 academic year. The problem solved was that a standard prosthetic for a transtibial amputee does not provide the flexibility in enough degrees of freedom for a user to squat and balance on a surfboard.
