A uniform dispersion of reactants is necessary to achieve a complete reaction involving multi-components, especially for the crosslinking of rigid high-performance materials. In these reactions, miscibility is crucial for curing efficiency. This miscibility is typically enhanced by adding a third component, a plasticizer. For the reaction of the highly crystalline crosslinking agent hexamethylenetetramine (HMTA) with a strongly hydrogen-bonded phenol formaldehyde resin, furfural has been traditionally used as the plasticizer. However, the reason for its effectiveness is not clear. Polyethylene glycol (PEG) hydrogels are tunable cell culture platforms that recapitulate tissue geometry, water content, and bulk modulus. Despite these benefits, PEG hydrogels elicit an acute immune response, limiting their use in regenerative medicine, and they critically underrepresent the cell-instructive proteins found in the extracellular matrix (ECM). Here, I developed a new class of tissue-specific PEG-based materials and provided biocompatible strategies to improve the user handling and cell viability post-encapsulation when using these hydrogels. I also demonstrated that decreasing the protein fouling to PEG does not decrease the foreign body response to implanted hydrogels, a common misconception in the field. …

Obstructive sleep apnea (OSA) is characterized by recurrent episodes of airway collapse and airflow limitation during sleep. Fragmented sleep and reductions in blood oxygen saturation lead to several comorbidities, including hypertension, cardiovascular disease, and cerebrovascular disease. Longitudinal forces (tracheal traction) acting on the soft tissues surrounding the upper airway have been proposed to play a significant role in stabilizing the airway and preventing collapse. However, the relative contribution of longitudinal forces as compared to other factors that affect airway stability (airway geometry, tissue properties, muscle activity) remains unclear. The work of the Cal Poly Tissue Engineering Lab is primarily focused on the fabrication, characterization, and improvement of “Blood Vessel Mimics” (BVMs), tissue engineered constructs used to evaluate cellular response to vascular medical devices. Currently, cells are grown onto fibrous, porous tubes made using an in-house electrospinning process from PLGA, a biocompatible co-polymer. The adhesion and proliferation of cells in a BVM is reliant on the micro-scale structure of the PLGA scaffold, and as such it is of great importance for the electrospinning process to consistently produce scaffolds of similar morphologies. Likewise, moving bed bioreactors (MBBR) are gaining interest and popularity because they are relatively affordable to build. Central nervous system neural device functionality hinges on effective communication with surrounding neurons. This depends on both the permissiveness of the device material to promote neuron integration and the ability of the device to avoid a chronic inflammatory response. Here, a facile approach has been developed exploring the multiple functionalities of hydrogel particles to provide cues to impart neural integration for such materials. Ferritin is an iron-storage globular protein with an ability to uptake, mineralize and release iron ions in a controllable manner. The globular hollow shell allows storage of mineralized iron, with several channels responsible for the transit of ions into the shell and out of it. Chronic wounds present a significant burden to patients, causing pain, impairing limb function, and often resulting in the need for amputation. Treatment of chronic dermal wounds is challenging, with current therapies showing limited efficacy in clinical trials. As galectin-3 has been implicated in several wound healing processes, its efficacy as a therapeutic in skin healing was investigated in this study. An electrospun gelatin scaffold loaded with galectin-3 was developed as a delivery system. Creation of tissue-mimicking constructs is of great importance in the field of biomedical engineering. Poly (vinyl Alcohol) (PVA) is a biomaterial capable of simulating a wide range of geometries and mechanical properties of biological tissues. It is nontoxic, biocompatible, and easy to produce. PVA can be physically crosslinked by repeated cycles of freezing and thawing. The final product of this process is called PVA cryogel (PVA-C). Stem cell transplantation is under investigation to stimulate angiogenesis in patients with peripheral artery disease. To develop a cell-delivery platform that enhances cell retention and function post-transplantation, the response of human adipose-derived stem/stromal cells (ASCs) encapsulated within N-methacrylate glycol chitosan (MGC) hydrogels with or without integrin-binding RGD or IKVAV motifs was explored. ASC viability was enhanced in the MGC and MGC-RGD hydrogels relative to the MGC-IKVAV group under hypoxic (2% O2) culture conditions, with cell spreading and higher metabolic activity noted in MGC-RGD at 14 days. Common biodegradable polyesters such as poly(lactic acid) (PLA), poly(lactic-co-glycolic acid) (PLGA) and poly(ε-caprolactone) (PCL) are used as drug delivery vehicles for tissue regenerative applications. However, they are typically bioinert, with drug loading limitations. Polymerizing the active agent or precursor into its respective biodegradable polymer would control drug loading via molar ratios of drug to initiator used for synthesis. Simvastatin was chosen due to its favorable anti-inflammatory, angiogenic, and osteogenic properties. In addition, its lactone ring lends itself to ring-opening polymerization and, consequently, the synthesis of poly(simvastatin) with controlled simvastatin release.

Implanted biomaterials often induce inflammation that frequently leads to the foreign body response, fibrosis, and the failure of the implant. Thus, it is important to evaluate how cells interact with materials to promote a more regenerative response. It is critical to determine how to modulate the response of tissue resident innate immune cells, as they are among the first cells to interact with implanted materials. Biodegradable and biocompatible poly (L-lactic-acid) (PLLA) coating was applied on a modified zinc (Zn) substrate by dip coating, with the intent to delay the bio-corrosion and slow the degradation rate of zinc substrate. 3-(Trimethoxysilyl) propyl methacrylate (MPS) was used for modification of the zinc substrate for promoting the adhesion between the metallic substrate and the polymer coating. It is hypothesized that the delay in Zn biodegradation could be useful in the first several weeks to prevent the early loss of mechanical integrity of the endovascular stent and to improve the healing process of the diseased vascular site. Zinc has begun to be studied as a bio-degradable material in recent years due to its excellent corrosion rate and optimal biocompatibility. Unfortunately, pure Zn’s intrinsic ultimate tensile strength (UTS; below 120 MPa) is lower than the benchmark (about 300 MPa) for cardiovascular stent materials, raising concerns about sufficient strength to support the blood vessel. Thus, modifying pure Zn to improve its mechanical properties is an important research topic.

Chronic Obstructive Pulmonary Disease (COPD) is a leading cause of death worldwide. Alveolar wall destruction is a significant contributor to COPD. Inflammatory macrophages are a major source of the Extracellular Matrix (ECM) proteolysis. ECM breakdown causes air to get trapped in the alveoli, obstructing airflow. One step in curing COPD may be to convert inflammatory to pro-regenerative macrophages. Recently, decellularized ECM scaffolds have shown the ability to induce a pro-regenerative phenotype.

Selective Laser Melting (SLM) of Additive Manufacturing is an attractive fabrication method that employs CAD data to selectively melt the metal powder layer by layer via a laser beam and produce a 3D part. This method not only opens a new window in overcoming traditional NiTi fabrication problems but also for producing porous or complex shaped structures. Selectively permeable biological membranes composed of lipophilic barriers inspire the design of biomimetic carrier-mediated membranes for aqueous solute separation. This work imparts selective permeability to lipid-filled pores of silica thin film composite membranes using carrier molecules that reside in the lipophilic self-assemblies. The lipids confined inside the pores of silica are proven to be a more effective barrier than bilayers formed on the porous surface through vesicle fusion, which is critical for quantifying the function of an immobilized carrier. Electrospinning holds great promise for designing functional 3D biomimetic scaffolds for tissue engineering applications. The technique allows for the reproducible fabrication of 3D scaffolds with control over the porosity and thickness. In this work, a novel method for the synthesis of a 3D electroactive scaffold using electrospinning from polycaprolactone (PCL), Polyvinylidene Fluoride (PVDF) and Polyaniline (PANI) is reported. Additional scaffolds involving different morphologies of PCL, PCL-PVDF and PCL-PANI-PVDF were also fabricated and evaluated. Many instruments are used to find elastic properties of biological samples using methods such as tensile and bending tests, but using the atomic force microscope (AFM) is considered a non-destructive method because it can provide repeated local stiffness information without damaging the sample. It additionally allows the sample to be tested in an aqueous environment, which is optimal for soft materials such as hydrogels. The goal of this research was to develop a system of individualized medicine that could be applied to dermal wounds serving as a wound dressing and synthetic extracellular matrix while delivering stem cells to the wound bed. First, fabrication parameters for electrospinning polymer fibers were determined. This involved evaluating fiber morphology with respect to polymer selection and solution concentration. Next, construct fabrication was examined to produce an integrated void space, or cargo area, suitable to maintain stem cells.
