Nanoparticles (NPs) are defined as particles less than 100 nm in at least one dimension. CuO NPs have possible applications in agriculture as micronutrient sources, pesticides, and enhancers of crop stress tolerance. Here, three aspects of CuO NP agricultural applications are studied: 1) the potential of CuO NPs to prevent wheat lodging-when crops irreversibly fall over; 2) CuO NP-induced drought tolerance in wheat seedlings; and 3) the effects of CuO NPs on outer membrane vesicle (OMV) production by Pseudomonas chlororaphis O6 (PcO6), a plant-health promoting bacterium.

FR901533 (1, also known as WS79089B), WS79089A (2), and WS79089C (3) are polycyclic aromatic natural products with promising inhibitory activity to endothelin-converting enzymes. In this work, we isolated five tridecaketide products from Streptosporangium roseum No. 79089, including 1-3, benaphthamycin (4) and a novel FR901533 analogue (5). The structure of 5 was characterized based on spectroscopic data. Compared to the major product 2, the new compound 5 has an additional hydroxyl group at C-12 and an extra methyl group at the 13-OH. Biochemical Methane Potential (BMP) tests are a crucial part of feasibility studies to estimate energy recovery opportunities from organic wastes and wastewater. Despite the large number of publications dedicated to BMP testing and numerous attempts to standardize procedures, there is no “one size fits all” mathematical model to describe biomethane formation kinetic precisely. Importantly, the kinetics models are utilized for treatability estimation and modeling processes for the purpose of scale-up. In this work we report the isolation, identification and antibacterial activity of two anthraquinones, 2-hydroxy-1-methoxyanthraquinone (1) and 2,5-dihydroxy-1-methoxy-6-methoxymethylanthraquinone (2), from the stem bark of Morinda lucida. These two natural products were selectively converted into two new glycosylated derivatives, 2-hydroxy-1-methoxyanthraquinone-4′-O-methyl-2-O-β-d-glucopyranoside (3) and 2,5-dihydroxy-1-methoxy-6-methoxymethylanthraquinone-4′-O-methyl-2-O-β-d-glucopyranoside (4) by the filamentous fungus Beauveria bassiana ATCC 7159. Structure elucidation was accomplished based on the 1D and 2D NMR, IR and mass spectra. Acute kidney injury (AKI) is a common complication associated with cardiac surgery. Those patients who develop AKI have increased hospital costs, hospital LOS and odds of death. The current diagnostic criteria rely on serum creatinine measurements and urine output data. Serum creatinine is not renal specific. It is known that there are other factors that influence serum creatine concentrations other than renal injury. Serum creatinine levels are measured as concentrations which means large changes in fluid balance may alter the serum creatinine measurement. While myalgic encephalomyelitis/chronic fatigue syndrome (ME/CFS) is relatively new and poorly understood, a recent upsurge in research has identified the disease’s core symptoms, including post-exertional malaise and orthostatic intolerance. The FDA has yet to approve any treatments for ME/CFS, partially due to a lack of validated efficacy endpoints.

The central focus of this research is to develop ME/CFS efficacy endpoints using a non-invasive, inertial measurement-based approach. Accessible endpoints will provide a way to properly evaluate potential treatments for ME/CFS. General anesthesia is well known to offer physicians access to a broad variety of invasive procedures otherwise deemed too risky. Anesthesia machines provides the means for anesthetizing patients safely in the hospital operating room. However, these devices are increasingly unable to meet the demands and needs outside of the hospital. Developing countries struggle to purchase and maintain these costly devices, leading to a 40-fold increase in anesthesia-related deaths compared to developed countries. Small-office practices in the United States experience significantly poorer anesthesia outcomes and increased legal claims versus their larger hospital counterparts, resulting in 60% more anesthesia-related deaths. High-dose propofol is being investigated for its potential antidepressant effect. Propofol is titrated to induce burst suppression, a specific EEG pattern. However, propofol is difficult to dose due to uncertainty in each patient’s pharmacokinetics (PK) and pharmacodynamics (PD), and the lack of a commercially available monitor of propofol concentration. Clinicians currently infer the proper drug dose after observing the EEG response to the given dose. In this report we share our development of an automated controller to optimally administer propofol-induced burst suppression. The technology is superior to current video laryngoscopes in that it utilizes machine learning techniques to guide the healthcare provider in real-time, providing augmented reality cues to anatomical features, feedback to prevent critical levels of deoxygenation of the patient, and an automated system to assess the difficulty of airway and call on the assistance of other physicians if first-pass intubation is not successful. Central Valley Water Reclamation Facility (CVWRF) in Salt Lake City is the largest municipal wastewater treatment plant in Utah and must meet new and rigorous nutrient effluent standards–over 95% reduction in phosphorus output by 2025. Filtrate from CVWRF anaerobic digesters contains high levels of nitrogen, phosphorus, and magnesium. Supersaturation of these constituents leads to nuisance struvite precipitation that clogs belts, pumps, and pipes downstream of anaerobic digesters. Struvite is a mineral precipitate composed of equimolar magnesium, ammonium, and phosphate. Controlled precipitation of struvite helps prevent clogging and scaling, removes phosphate from wastewater, and generates a marketable fertilizer product.

The purpose of this project was to characterize the potential of various strains of purple non-sulfur bacteria for the production of fixed nitrogen fertilizers for the manned Mars missions. Six strains of Rhodopseudomonas palustris (CGA009, CGA010, TIE-1, NifA*, and PB23) were all investigated. Through initial growth trial experiments, R. palustris NifA* and PB23 were selected for their engineered nitrogen fixation and rapid growth respectively. Growth curves and ammonium concentrations were collected over time in pilot scale batch photobioreactors (200 mL). Biomass production was then scaled up to benchtop photobioreactors (1.5 L) and then 14 L total volume for both NifA* …

Choroidal neovascularization (CNV) is a major cause of blindness in patients with age-related macular degeneration. CNV is characterized by new blood vessel growth and subretinal fluid accumulation, which results in mechanical pressure on retinal pigment epithelial (RPE) cells. The overexpression of RPE-derived angiogenic factors plays an important role in inducing CNV. In this work, we investigated the effect of mechanical stress on the expression of angiogenic factors in porcine RPE cells and determined the impact of conditioned medium on in-vitro angiogenesis.

Nanofertilizers are increasingly explored for sustainable micronutrient delivery in agriculture. Pre-treating seeds with nanofertilizers prior to planting (i.e., seed priming) reduces concerns about nanoparticle (NP) fertilizer non-target dispersion; however, priming formulations and concentrations must be carefully selected to avoid germination inhibition and toxicity. Here we investigate changes in corn seed germination and seedling development after seed priming with ZnO NPs, ZnO bulk and ZnCl2. Biomass feedstocks contain inorganic compounds generally classified as ash. The ash consists of compounds of potassium, calcium, magnesium, silicon, phosphorus. and other elements. These elements have been reported to influence both the pyrolysis reactions as well as the destabilization of the pyrolysis oils during storage. The inorganic elements have also been reported to deposit on catalyst surfaces during in situ catalytic pyrolysis leading to the eventual deactivation of acidic catalysts such as zeolites. A major challenge for astronauts in long-duration space travel is combatting the hazardous spaceflight environment caused by microgravity and increased levels of ionizing radiation. Microgravity damages cellular DNA by increasing the production of harmful reactive oxygen species, while ionizing radiation damages DNA by creating double-stranded DNA (dsDNA) breaks. Diesel exhaust particles (DEPs) are major air pollutants that lead to numerous human disorders, especially pulmonary diseases, partly through the induction of oxidative stress. Resveratrol is a polyphenol that ameliorates the production of reactive oxygen species (ROS) and delays aging-related processes. These properties make spider silk a desirable material for many commercial products, ranging from textiles to biomedical materials. This study investigated the effect of milk oligopeptides and (α)-tocopherol on inhibition of linoleic acid oxidation using Fe2+-vitamin C induced linoleic acid oxidation model through analysis of malondialdehyde, peroxide value, and conjugated diene and triene in the model. The alteration of milk oligopeptides maximal absorption wavelength, fluorescent feature, and secondary structure were further investigated to elucidate the interactions between milk oligopeptide and (α)-tocopherol that altered the inhibitory effect of linoleic acid oxidation. Results showed that Pro-Tyr-Tyr-Ala-Lys (PYYAK) and Ile-Pro-Ile-Gln-Tyr (IPIQY) with (α)-tocopherol significantly inhibited the oxidation of linoleic acid and reduced the formation of malondialdehyde by 38% and …

Organosilanes (e.g., R’-SiOR3) provide hydrophobic functionality in thin-film coatings, porous gels, and particles. Compared with tetraalkoxysilanes (SiOR4), organosilanes exhibit distinct reaction kinetics and assembly mechanisms arising from steric and electronic properties of the R’ group on the silicon atom. Here, the hydrolysis and condensation pathways of n-propyltrimethoxy silane (nPM) and a tri-fluorinated analog of nPM, 3,3,3-trifluoropropyl trimethoxy silane (3F), were investigated under aqueous conditions at pH 1.7, 2.0, 3.0, and 4.0. Indigoidine is a natural blue dye with antioxidant and antimicrobial activities. It has also been used as an indicator for gene expression based on its distinctive blue color. Similar to the industry blue dye indigo, indigoidine has a promising potential to be applied in industry as a blue dye. However, the indigoidine production level in the original microorganisms was very low. Heterologous expression of the responsible synthetase gene in Escherichia coli can facilitate the fast and large-scale production of indigoidine. Background

Endophytic microorganisms are a rich source of bioactive natural products. They are considered as promising biofertilizers and biocontrol agents due to their growth-promoting interactions with the host plants and their bioactive secondary metabolites that can help manage plant pathogens. Identification of new endophytes may lead to the discovery of novel molecules or provide new strains for production of valuable compounds.

Results

In this study, we isolated an endophytic bacterium from the leaves of Taxus chinensis, which was identified as Pseudomonas sp. 102515 based on the 16S rRNA gene sequence and physiological characteristics. Analysis of its secondary metabolites revealed …

Spider silk has received significant attention due to its fascinating mechanical properties. Given the solitary and cannibalistic behavior of spiders, spider silk farming is impractical. Unlike spiders, silkworms are capable of producing large quantities of a fibrous product in a manner mimetic to spiders, and there already exists an industry to process cocoons into threads and textiles for many applications. The combination of silk farming (sericulture), a millennia old practice, and modern advancements in genetic engineering has given rise to an innovative biomaterial inspired by nature; transgenic silkworm silk.

A renewable energy source, biogas, comprises of methane (80%) and carbon dioxide (15%), and is a great alternative to the conventional fossil-based fuels, such as coal, gas and oil. Biogas is created during anaerobic biological digestion of waste materials, such as landfill material, animal manure, wastewater, algal biomass, industrial organic waste etc. A biogas potential from organic waste in the United States is estimated at about 9 million tons per year and technology allows capture of greenhouse gases, such as methane and carbon dioxide, into a form of a fuel. The area around a plant’s roots hosts a complex and diverse microbial community. This environment can include a large number of bacteria that live on the surface of the root and benefit from the nutrients that the roots exude into the soil. These microbes can in turn be beneficial to the plant by protecting the roots from harmful fungi or stressful environmental conditions such as drought. This dissertation is focused on exploring the potential of bacteria for the biosynthesis of natural products with the purposes of generating novel natural product derivatives and of improving the titer of pharmaceutically important natural products.
