The tissue/biomaterial interactions of three biomaterials of potential use in bone implants were studied in vitro. The mechanism of cell adherence to various ceramic substrata has been investigated by measurement of short term (90 minutes) cell attachment and spreading rate. We have determined the effect of two serum glycoproteins, fibronectin and vitronectin, on the adhesion of human bone derived cells (HBD-cells) cultured on three different types of ceramics {alumina (Al2O3), hydroxyapatite (HAP) and yttria-doped tetragonal zirconia polycrystal (Y-TZP)} . This investigation determined the effects of reaction temperature and bulk composition on the microstructural features of hydroxyapatites (HAp) synthesized from reaction between particulate CaHPO4·2H2O and Ca4(PO4)2O. These data were used in combination with previous work to further elucidate the mechanistic reaction path taken by these calcium phosphate cements. HAps having two different compositions (Ca/P = 1.50 and 1.67) were synthesized between 15.0 and 70.0°C. All reactions reached completion as indicated by X-ray diffraction. The surface and cross-section characteristics of the plasma-sprayed calcium phosphate coatings, along with the microstructures and elemental compositions near the titanium alloy interface, were investigated by scanning electron microscopy for two different dental implants of proprietary compositions: Integra!® (Calcitek) and BioVent ® (Dentsply). Elemental concentrations (Ca, P, Ti, AI, and V) near the interfaces were obtained by X-ray energy-dispersive spectroscopy. Coating surfaces exhibited a splat deposition topography of greatly differing microstructural scale for the two implants, along with other features characteristic of the plasma-spraying process; cross-sections of the coatings revealed minimal porosity. The preliminary results of biological and physical tests of hydroxyapatite thin films deposited on dental implants by a new technology with the KrF excimer laser ablation method were evaluated. Biological and physical properties were studied and analyzed by the lymphocyte proliferation test and scanning electron microscopy (SEM), X-ray analysis, Rutherford backscattering analysis (RBS) and particle induced X-ray emission (PIXE) methods. Ten bioceramic films from 45 samples had very good physical and biological properties. Creation of hydroxyapatite thin films with laser ablation can have a positive effect on adhesion of the film and protection for corrosion.

Rat bone marrow cells, capable of forming bonelike tissue, were cultured on three types of plasma sprayed hydroxyapatite that differed in degree of crystallinity from 15%, 43% to 69%. The interface between the mineralized extracellular matrix and the hydroxyapatite was studied with scanning and transmission electron microscopy. At the onset of bone formation, calcium and phosphorous-rich afibrillar globules, with a diameter varying from 0.2 to 0.8 μm, were produced and deposited onto the different substrata. These globules subsequently fused to form a homogeneous layer to which collagen fibres became anchored. Individual globules could be distinguished on the non-degrading 69 % …

The implantation of calcium phosphate ceramics in the bone environment elicits the differentiation of multinucleated giant cells (MNGCs). MNGC interactions with both crushed and standard hydroxyapatite (HA, Bioapatite ®) particles (cHA and sHA) were studied in rat skull defects. Light microscopy (LM) of undemineralized material after 21 and 42 days of implantation showed that the number of particles in the defect significantly decreased (about SO%) with the two HA forms. Concomitantly, average cHA particle size increased significantly due to the elimination of the smallest particles by the MNGCs. The tissue response to implanted hydroxyapatite (HAP) in rat alveolar bone was investigated using morphological and cytochemical approaches. High-resolution immunogold labeling was used to examine the distribution of two non-collagenous bone proteins, osteopontin and osteocalcin, and two plasma proteins, a2HSglycoprotein (fetuin) and albumin. Glycoconjugates were characterized by lectin-gold cytochemistry using Ricinus communis I agglutinin and Wheat germ agglutinin. Transmission electron microscopy demonstrated the presence of an electron-dense, lamina limitans-like layer at the HAP particle-bone interface and organic material within the micropores of the particles. Calcium phosphate ceramics are used in bone surgery under different forms: dense or porous ceramic s as bone substitute, thin ceramic coatings on metallic implants as an osseointegration enhancer. Their degradation depends on their physico-chemical properties and particularly on their chemical composition. Natural calcium phosphates of bone are degraded by mononuclear or multinuclear cells and the extracellular matrix induces the differentiation of the degrading-cells. An in vitro rat bone marrow cell system was used to examine the interfacial ultrastructure established between various calcium phosphates and mineralized tissue. The investigated calcium phosphates comprised hydroxyapatite (HA), fluorapatite (FA), tricalcium phosphate (TCP), tetracalcium phosphate (TECP) and magnesium whitlockite (MWL). Both scanning and transmission electron microscopy were used to examine the elaborated interface. The time in which a mineralized extracellular matrix was formed on the various materials differed from 2 weeks on HA, TCP and TECP, to 8 weeks on FA. Knowledge of the phase, composition, and crystallite size and perfection of the mineral in normal and abnormally calcified tissues provides insight into the mechanism by which this mineral was deposited. These data also can be used to develop rational therapies for pathological conditions characterized by abnormal mineral deposition. As illustrated in this review, coupling of an optical microscope with a Fourier transform infrared (FT-IR) spectrophotometer permits the mapping at 20 μm spatial resolution of changes in mineral characteristics (content, particle size, composition) in the growth plate, in bone biopsies, in mineralizing cell culture systems, and in soft tissue calcifications. Porous hydroxyapatite ceramics combined with rat marrow cells were implanted subcutaneously in the back of syngeneic Fischer rats . Fluorochrome-labeling (calcein, tetracycline) was performed post-operatively and the ceramics were harvested 4 weeks after implantation. Undecalcified thin sections of the implants were observed under light microscopy or fluoromicroscopy and the corresponding areas were also analyzed in a scanning electron microscope connected to an electron probe microanalyzer (SEM-EPMA). Many pore areas of the ceramics showed bone and osteoid formation together with active osteoblasts. Collagenase isolated rat calvaria cells forming a mineralized matrix in vitro were cultured in the presence of synthetic hydroxyapatite. Interactions between bone cells and hydroxyapatite biomaterial were followed by transmission electron microscopy. The appearance of a granular, collagen free, electron-dense layer at the periphery of the material was noted initially. Progressively, an amorphous, granular material formed and extended between the hydroxyapatite aggregates. Osteoblastic cells then synthesized an osteoid matrix which mineralized on the first formed granular collagen free layer, following a classical pattern of calcification . The biomineralization of bovine pericardium (BP) heart valve bioprostheses was investigated by simulation of the process under in vitro and in vivo conditions. The nature and composition of calcific deposits that formed in human heart valve bioprostheses were compared with the mineral formed on BP discs immersed in a calcifying medium or subcutaneously implanted into Sprague-Dawley rats. The early stage of experimental biomineralization in vitro took place on the surface only, while in vivo deposition appeared to be intrinsic, as documented by histological cross sections.
