This paper describes the design, manufacturing, and testing of a novel controllable hypoxic incubator with fully functional oxygen gas control and temperature control in a humid environment. On the current market, a majority of the few hypoxic incubators use pre-mixed gas that does not offer precise control over gas concentration. The objective for this project was to create a chamber that allows the user to set the O2 concentration to varying set points of % O2 while maintaining the chamber at a constant body temperature, CO2 level, humidity, and sterility. This document is an overview report of the Cal Poly Senior Project ASTM Test Fixture for SeaSpine orthopedic bone screws. The main application of orthopedic screws is to fasten implants directly to the bone structure, enhancing structural integrity. Testing for breaking angle, insertion torque, removal torque, axial pullout strength, and self-tapping force as specified in the ASTM F543 Annexes provides the FDA with vital screw mechanics and ensures manufacturing uniformity. In this work, a novel microfluidic cell culture platform capable of automated electrical impedance measurements and immunofluorescence and brightfield microscopy was developed for further in-vitro cellular research intended to optimize cell culture conditions. The microfluidic system design, fabrication, automation, and design verification testing are described. Electrical and optical measurements of the 16 parallel cell culture chambers were automated via a custom LabView interface. A proposed design change will enable gas diffusion, removing the need for an environmental enclosure and allow long-term cell culture experiments. The purpose of the Tierni Resistance Training System project was to design and construct functional workout apparel that has built-in resistance. For this product, the key customer requirements we set out to address were most importantly material comfort and functionality, followed by stylishness, lack of latex, safety for injured and uninjured users, and washer safety. To meet these customer requirements, we started by researching current resistance training technology, and used the findings as a springboard for our own design development. This project aims to integrate an automatic gas release system in a pre-existing scaffold fabrication process for tissue engineering applications.

To form the proper scaffold structure, the fabrication process is heavily influenced by the change in its surrounding pressure. The current production involves a pressure transducer and electric valve that is managed manually to create a suitable pressure environment for the scaffold. This method, although functional, proves to be ineffective when creating several batches; the user needs to constantly monitor the developing pressure profile and alter voltage parameters accordingly to create a linear gas release under a predetermined slope.

Infant apnea is a widespread condition in which infants fail to effectively breathe, and can lead to death. Clinical solutions exist for continuous monitoring of respirations in a hospital setting and requiring constant skin contact. This thesis investigates the construction of a proof of concept device that performs in-home monitoring without skin contact and with commonly available off-the-shelf components.

For this project, the design, implementation, characterization, calibration and possible applications of a multiple transducer stethoscope apparatus were investigated. The multi-transducer sensor array design consists of five standard stethoscope diaphragms mounted to a rigid frame for a-priori knowledge of their relative spatial locations in the x-y plane, with compliant z-direction positioning to ensure good contact and pressure against the subject’s skin for reliable acoustic coupling. This system allows these veterans to complete a slalom speed race independent of a personal guide in the water. Microfluidic devices are diagnostic tools that are small, portable, and inexpensive. This makes them ideally suited for resource-limited settings. To construct a 3D device, each layer of patterned paper is stacked using double-sided tape with holes corresponding to the pattern. It is necessary to have a paste to fill the holes in the tape and provide a medium for the fluid to wick through. This thesis successfully integrates laminate microfluidic devices with an analytic Surface Plasmon Resonance (SPR) instrument. Integration was accomplished at low-cost using materials such as polydimethylsiloxane (PDMS), Poly(methyl methacrylate) (PMMA), Tygon tubing, and a 3-way stopcock. The main components of this thesis are the design and fabrication of the low-cost, in-house fluidics that can integrate with upstream microfluidics and the validation of the in-house fluidics using the Biosensing Instruments BI-2000 SPR instrument.
