The use of neural prostheses to improve health of paraplegics has been a prime interest of neuroscientists over the last few decades. Scientists have performed experiments with spinal cord stimulation (SCS) to enable voluntary motor function of paralyzed patients. This dissertation introduces a novel approach at gauging patterns of informa- tion flow using brain connectivity analysis and partial directed coherence (PDC) in epilepsy. The main objective of this dissertation is to assess the key characteristics that delineate neural activities obtained from patients with epilepsy, considering both focal and generalized seizures. The use of PDC analysis is noteworthy as it es- timates the intensity and direction of propagation from neural activities generated in the cerebral cortex, and it ascertains the coefficients as weighted measures in formulating the multivariate autoregressive model (MVAR). Assessing complex brain activity as a function of the type of epilepsy and in the context of the 3D source of seizure onset remains a critical and challenging endeavor. In this dissertation, we tried to extract the attributes of the epileptic brain by looking at the modular interactions from scalp electroencephalography (EEG). A classification algorithm is proposed for the connectivity-based separation of interictal epileptic EEG from normal. Connectivity patterns of interictal epileptic discharges were investigated in different types of epilepsy, and the relation between patterns and the epileptogenic zone are also explored in focal epilepsy.

A lack of adherence to prescribed physical therapy regimens in improper healing results in poor outcomes for those affected by musculoskeletal disorders (MSDs) of the upper limb. Societal and psychological barriers to proper adherence can be addressed through the system presented in this work consisting of the following components: an ambulatory biosignal acquisition sleeve, an electromyography (EMG) based motion repetition detection algorithm, and the design of a compatible capacitive EMG acquisition module.

Studies have shown that the activity of muscarinic receptors and their affinity to agonists are sensitive to membrane potential. It was reported that in airway smooth muscle (ASM) depolarization evoked by high K+ solution increases contractility through direct effects on M3 muscarinic receptors. In this study, we assessed the physiological relevance of voltage sensitivity of muscarinic receptors on ASM contractility. Our findings reveal that depolarization by high K+ solution induces contraction in intact mouse trachea predominantly through activation of acetylcholine release from embedded nerves, and to a lesser extent by direct effects on M3 receptors. According to the World Health Organization, disabling hearing loss affects nearly 466 million people worldwide. Sensorineural hearing loss (SNHL), which is characterized as damage to the inner ear (e.g., cochlear hair cells) and/or to the neural pathways connecting the inner ear and brain, accounts for 90% of all disabling hearing loss. More concerning is that significant perceptual and physiological aspects of SNHL remain “hidden” from standard clinical diagnostics. Hidden hearing loss (HHL) manifests as the inability to understand speech in loud, noisy environments (e.g., listening in a noisy restaurant) despite a normal audiogram (i.e., normal detection of soft sounds). Pitch is an important perceptual dimension in audition, supporting auditory object segregation, melody recognition and lexical distinction. Huggins’ pitch, for example, is a phenomenon evoked by two sources of broadband noise presented binaurally with an inter-aural phase shift over a narrow frequency band. Huggins’ pitch and other dichotic pitches have been studied extensively using perceptual experiments. Several models have been proposed to explain and predict the perception of pitch; however, no studies have tried to record in vivo neuron responses to Huggins’ pitch (HP) nor have tried to explain how the HP is coded by neurons. The peripheral sensory system is critical to regulating motor plasticity and motor recovery. Peripheral electrical stimulation (ES) can generate constant and adequate sensory input to influence the excitability of the motor cortex. The aim of this proof of concept study was to assess whether ES prior to each hand function training session for eight weeks can better improve neuromuscular control and hand function in chronic stroke individuals and change electroencephalography-electromyography (EEG-EMG) coherence, as compared to the control (sham ES). The focus of this thesis was to measure the intracellular voltage of a living neural cell using a silicon transistor. The coupling of neurological tissues with silicon devices is of interest to the fields of neurology, neuroscience, electrophysiology and cellular biology. Custom-designed macro-sieve electrodes represent a novel means of facilitating chronic high specificity nerve stimulation needed to control distal nerve musculature and restore sensorimotor function. Implantation of these electrodes requires the transection of the nerve, which has shown to disrupt muscle fiber distribution. This present study assesses the feasibility of implementing these electrodes in an end-to-side nerve graft. The macro-sieve electrodes were fabricated and micro-surgically implanted into 3.2 cm nerve autografts harvested from the sciatic nerve of 12 male Lewis rats. Neuromorphic engineering involves designing artificial neural systems that mimic the way neuron circuits in the brain process information and make computations. It took the fourth most powerful computer in the world (with 705,024 processor cores and 1.4 million GB of RAM) 40 minutes to simulate just one second of human brain activity. This shows a clear difference of energy use in the human brain versus modern computers; neuromorphic engineering could be how we mimic the computing power of the brain to create energy-efficient, neuron-based computers. Electrophysiological measurements have been used in recent history to classify instantaneous physiological configurations, e.g., hand gestures. This work investigates the feasibility of working with changes in physiological configurations over time (i.e., longitudinally) using a variety of algorithms from the machine learning domain. We demonstrate a high degree of classification accuracy for a binary classification problem derived from electromyography measurements before and after a 35-day bedrest. The problem difficulty is increased with a more dynamic experiment testing for changes in astronaut sensorimotor performance by taking electromyography and force plate measurements before, during, and after a jump from a small platform. Our project took a mechanically operated 3D printed prosthetic hand and automated the process by which it makes a grasp. The purpose of this project was to provide an affordable, automatic prosthetic hand to those either capable and incapable of wrist flexion. Neurological disorders require varying types and degrees of treatments depending on the symptoms and underlying causes of the disease. Patients suffering from medication-refractory symptoms often undergo further treatment in the form of brain stimulation, e.g. electroconvulsive therapy (ECT), transcranial direct current stimulation (tDCS), deep brain stimulation (DBS), or transcranial magnetic stimulation (TMS). Nanopulse Stimulation (NPS) eliminates mouse and rat tumor types in several different animal models. NPS induces protective, vaccine-like effects after ablation of orthotopic rat N1-S1 hepatocellular carcinoma. Here we review some general concepts of NPS in the context of studies with mouse metastatic 4T1 mammary cancer showing that the postablation, vaccine-like effect is initiated by dynamic, multilayered immune mechanisms. NPS eliminates primary 4T1 tumors by inducing immunogenic, caspase-independent programmed cell death (PCD). With lower electric fields, like those peripheral to the primary treatment zone, NPS can activate dendritic cells (DCs). The activation of DCs by dead/dying cells leads to increases …

Cytosolic DNA sensors are a subgroup of pattern recognition receptors (PRRs) and are activated by the abnormal presence of the DNA in the cytosol. Their activation leads to the upregulation of pro-inflammatory cytokines and chemokines and can also induce cell death. The presence of cytosolic DNA sensors and inflammatory cytokines in TS/A murine mammary adenocarcinoma and WEHI 164 fibrosarcoma cells was demonstrated using real time reverse transcription polymerase chain reaction (RT-PCR), western blotting and enzyme-linked immunosorbent assay (ELISA). Nano-pulse stimulation (NPS), previously called nsPEFs, induced a vaccine-like effect after ablation of orthotopic N1-S1 hepatocellular carcinoma (HCC), protecting rats from subsequent challenges with N1-S1 cells. To determine immunity, immune cell phenotypes were analyzed in naïve, treated and protected rats. NPS provides a positive, post-ablation immuno-therapeutic outcome by alleviating immunosuppressive T regulatory cells (Treg) in the tumor microenvironment (TME), allowing dendritic cell influx and inducing dynamic changes in natural killer cells (NKs), NKT-cells and T-lymphocytes in blood, spleen and liver. Several studies have shown that different control plasmids may cause antitumor action in different murine tumor models after gene electrotransfer (GET). Due to the differences in GET protocols, plasmid vectors, and experimental models, the observed antitumor effects were incomparable. Therefore, the current study was conducted comparing antitumor effectiveness of three different control plasmids using the same GET parameters. The Anterior Cruciate Ligament (ACL) is the primary source of rotational stability in the knee by preventing the tibia from sliding in front of the femur. When the ACL is torn, it typically must be repaired through reconstructive surgery which results in proprioceptive deficiencies in the knee. Proprioception plays an important role in understanding where one’s knee is in space, sensing movement and reacting accordingly. Melanins are organic polymers made up of 5,6-dihydroxyindole carboxylic acid (DHICA) and 5,6-dihydroxyindole (DHI). Biochemical systems synthesize melanins through a complex reaction pathway. Previous research exhibited that naturally-occurring melanins (NatMel) and their synthetic analogues (SynMel) can potentially serve as electrode materials for aqueous charge storage devices. Small surface area creates technical challenges for these melanins as charge storage materials. Biosensors are called upon to provide valuable benefits for human society in vital fields such as disease diagnosis, food inspection, environment monitoring, etc. Among the various biosensor architectures, the field effect transistor (FET) biosensors are promising as the next generation nanoelectronic biosensors, particularly attractive for point-of-care biomedical applications. The FET biosensors typically operate by measuring the conductance change of the semiconducting channel induced by the adsorption of the target biomolecules on it.
