Biofuels from Waste to Road (WASTE2ROAD) is an EU funded project under the Grant Agreement No. 818120 within the LC-SC3-RES-21-2018 call, “Development of next generation biofuels and alternative renewable fuel technologies for road transport”, as a Research and Innovation Action of the European Union’s Horizon 2020 Programme. Developing A Control System To Better Understand The Effects Of Pyruvate Decarboxylase Activity On Clostridium Thermocellum Metabolism, Nicholas Cervenka

In order for cellulosic biofuels from Clostridium thermocellum to be commercially viable, the ethanol yield and titer of the microbe must be increased. To accomplish this, it has been suggested to introduce the Pyruvate Decarboxylase (PDC) enzyme into C. thermocellum. In order to demonstrate effects on ethanol production by PDC prior to genetic modification, a cell free system (CFS) has been developed. A purified enzyme system was developed with the CFS to function as a control. Using the purified enzyme system, PDC from Saccharomyces cerevisiae was demonstrated to be a good candidate for further testing in the CFS.

"Biofuel is a promising clean alternative to fossil fuels. Currently, first generation biofuels are commercially produced by using corn grain as biomass feedstock. However, the use of edible matter of crops, may lead to a competition between food and fuel. Therefore, there is a significant push in both industry and academia to commercialize second generation biofuel manufacturing technology, which uses non-edible matter from crops. Most research focuses on individual manufacturing processes for producing second generation biofuel, but the economic and environmental impacts of a large-scale adoption of second generation biofuel manufacturing have been less widely reported.

Low-value biomass materials such as processing co-products and agricultural wastes possess abundant potential in value-added products production. The effort for utilization of these materials, especially as the substrate for fermentation process was limited by the low availability of fermentable sugar. This is due to the presence of lignin and hemicellulose in biomass structure, which entails appropriate pretreatment procedure to be conducted to expose the cellulose for the enzymatic reaction.
