Necrotic skin flaps are difficult to predict and treat due to the lack of quantitative biomarkers. Label-free multiphoton microscopy is well suited for non-invasively monitoring skin metabolism through NAD(P)H and other intrinsic fluorophores, and offers immediate future directions for assessing necrosis in the clinic. The objective of this study was to assess whether phasor FLIM could be used to evaluate skin flap status and treatment efficacy in ex vivo skin sections. Phasor maps revealed differences in growth factor treatment and region, but changes in skin flap autofluorescence at 755nm excitation and 460nm emission were not just related to NAD(P)H. A …

One of the important surgical tools in spinal surgery is the C-Arm X-ray System. The C-Arm is a large “C” shaped and manually maneuvered arm that provides surgeons and X-ray technicians the ability to take quick quality X-rays during surgery. Because of its five degrees of freedom, the C-Arm can be manually maneuvered around the patient to provide many angles and perspectives, ensuring surgical success.

Commonly used simulation models for predicting outbreaks of re-emerging infectious diseases (EIDs) take an individual-level or a population-level approach to modeling contact dynamics. These approaches are a trade-off between the ability to incorporate individual-level dynamics and computational efficiency. Agent-based network models (ABNM) use an individual-level approach by simulating the entire population and its contact structure, which increases the ability of adding detailed individual-level characteristics. However, as this method is computationally expensive, ABNMs use scaled-down versions of the full population, which are unsuitable for low prevalence diseases as the number of infected cases would become negligible during scaling-down. Our team at the Los Alamos National Laboratory has developed novel assays to detect bacterial biomarkers from patient blood at the point-of-care in order to facilitate a universal diagnostic platform. Dopamine is critical for processing of reward and etiology of drug addiction. Astrocytes throughout the brain express dopamine receptors, but consequences of astrocytic dopamine receptor signaling are not well established. This thesis illustrates effects of dopamine on cultured astrocytes and astrocytes in brain slices (ex vivo). In striatal cultures, extracellular dopamine triggered changes in astrocytic Ca2+ signaling and rapid concentration-dependent stellation of astrocytic processes that was not a result of dopamine oxidation, but instead relied on both cAMP-dependent and cAMP-independent dopamine receptor signaling. AhR was discovered as the receptor that binds with high affinity to 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) and leads to numerous of toxicological outcomes. Friedreich Ataxia (FA) is a debilitating autosomal-recessive neurodegenerative disorder which is characterized by ataxia of all four limbs, difficulty walking, areflexia, and dysarthria. Further complications of FA include diabetes, scoliosis, and hypertrophic cardiomyopathy. Approximately three-quarters of people with FA have onset before the age of 25 and in most instances, affected individuals will require the use of a wheelchair within ten years after symptoms emerge.

Laparoscopic procedures account for 15 million surgeries worldwide [1], with the initial entry into the peritoneal cavity accounting for 33-50% of all major laparoscopic complications [7]. This initial entry is the most dangerous as surgeons must enter the cavity using a sharp object with no visibility and space between the outer surface of the cavity and internal tissues. During the initial entry into the peritoneal cavity, the patients undergoing laparoscopic procedures are at a high risk for damage to internal organs and vasculature, necessitating the development of a device to protect these internal tissues and increase patient safety.

Traumatic Spinal Cord Injury (SCI) results in structural and functional neurological changes at both the brain and the level of the spinal cord. Anatomical studies indicate decreased grey matter volume in sensorimotor and non-sensorimotor regions of the cortex following SCI; whereas, neurophysiological findings mostly report altered functional activity in the sensorimotor nodes of the cortex, subcortex, and cerebellum. Therefore, it is currently unknown whether tissue atrophy observed in non-motor related areas has any concomitant functional consequences. Brain-inspired computation promises a paradigm shift in information processing, both in terms of its parallel processing architecture and the ability to learn to tackle problems deemed unsolvable by traditional algorithmic approaches. The computational capability of the human brain is believed to stem from an interconnected network of 100 billion compute nodes (neurons) that interact with each other through approximately 1015 adjustable memory junctions (synapses). The conductance of synapses is modifiable allowing the network to learn and perform various cognitive functions. Classical methods for psychometric function estimation either require excessive resources to perform, as in the method of constants, or produce only a low resolution approximation of the target psychometric function, as in adaptive staircase or up-down procedures. The purpose of this dissertation is to quantify the effects of using geared wheelchair wheels on upper extremity biomechanics and energy expenditure during functional mobility tasks in individuals with spinal cord injury (SCI). The effects of using geared wheels on hand-rim biomechanics, glenohumeral joint dynamics, and shoulder muscle activity were investigated during manual wheelchair propulsion over tiled and carpeted level-floors and up a ramp in low gear (1.5:1) and standard gear (1:1) conditions. Murine xenotropic leukemia virus (XMuLV) is one of the retroviruses, recommended by Food and Drug Administration (FDA) as a model virus for viral clearance via inactivation from therapeutics derived from Chinese hamster ovary cells (CHO). The primary goal of the research presented in this thesis was to study effects of sensory stimulus on the olfactory system of rats, focusing on the olfactory bulb (OB) and the anterior piriform cortex (aPC). Extracellular electrophysiological measurements revealed distinct frequency bands of oscillations in OB and aPC. Analysis of peripheral venous pressure (PVP) waveforms is a novel method of monitoring intravascular volume. Two cohorts were used to study the hemodynamics change of the body state and its influence on the PVP using (1) dehydration setting with infants suffering from pyloric stenosis and (2) hemorrhage setting during a craniosynostosis elective surgery. The goal of this research is to develop a minimally invasive method of analyzing the PVP waveforms and find correlations with volume loss.

Tuberculosis is the deadliest infectious disease in the world; it is especially rampant in underdeveloped countries because they do not have the infrastructure, technology, or funding to properly combat the infection. However, the development of portable point-of-care diagnosis machines can reverse this epidemic as they far surpass conventional laboratory identification. The question now is where to place these machines, which is a difficult decision with a lack of data.
